    Abstract: An actinomycete, strain D1 T , was isolated from a freshwater sediment sample collected from the San Pablo river in the La Risueña community, Santiago de Cuba province, Cuba. The strain was identified as a member of the genus Nocardiopsis by means of a polyphasic taxonomic study. It produced a light yellow non-fragmented substrate mycelium, a white well-developed aerial mycelium and straight to flexuous hyphae. No specific spore chains were observed. Strain D1 T contained meso- diaminopimelic acid, no diagnostic sugars, and MK-10(H 2 ), MK-10(H 4 ), MK-10 and MK-10(H 6 ) as predominant menaquinones, but not phosphatidylcholine as diagnostic polar lipid of the genus Nocardiopsis . The predominant fatty acids were iso-C 16 : 0 , 10-methyl-C 18 : 0 and anteiso-C 17 : 0 . Strain D1 T showed the highest degree of 16S rRNA gene sequence similarity to Nocardiopsis synnematoformans DSM 44143 T (99.8 %), Nocardiopsis dassonvillei subsp. albirubida NBRC 13392 T (99.8 %) and Nocardiopsis dassonvillei subsp. dassonvillei DSM 43111 T (99.6 %). A genomic OrthoANIu value between D1 T and N. dassonvillei subsp. dassonvillei DSM 43111 T of 97.63 % and a dDDH value of 78.9 % indicated that strain D1 T should be classified in N. dassonvillei . However, phenotypic characteristics distinguished strain D1 T from its nearest neighbour taxon. On basis of these results we propose to classify strain D1 T (=LMG 30468 T =CECT 30033 T ) as a representative of a novel subspecies of the genus Nocardiopsis , for which the name Nocardiopsis dassonvillei subsp. crassaminis subsp. nov. is proposed. In addition, the genomic distance between N. dassonvillei subsp. albirubida NBRC 13392 T and N. dassonvillei subsp. dassonvillei DSM 43111 T as determined through OrthoANIu (93.64 %) and dDDH (53.40 %), along with considerable phenotypic and chemotaxonomic differences reported in earlier studies, indicated that the classification of this taxon as Nocardiopsis alborubida Grund and Kroppenstedt 1990 is to be preferred over its classification as N. dassonvillei subsp. albirubida Evtushenko et al. 2000.

    Abstract: During a bioprospection of bacteria with antimicrobial activity, the actinomycete strain A38 T was isolated from a sediment sample of the Carpintero river located in the Gran Piedra Mountains, Santiago de Cuba province (Cuba). This strain was identified as a member of the genus Micromonospora by means of a polyphasic taxonomy study. Strain A38 T was an aerobic Gram-positive filamentous bacterium that produced single spores in a well-developed vegetative mycelium. An aerial mycelium was absent. The cell wall contained meso -diaminopimelic acid and the whole-cell sugars were glucose, mannose, ribose and xylose. The major cellular fatty acids were isoC 15:0 , 10 methyl C 17:0 , anteiso-C 17:0 and iso-C 17:0 . The predominant menaquinones were MK-10(H 4 ) and MK-10(H6). Phylogenetic analysis of 16S rRNA gene sequences revealed that this strain was closely related to Micromonospora tulbaghiae DSM 45142 T (99.5 %), Micromonospora citrea DSM 43903 T (99.4 %), Micromonospora marina DSM 45555 T (99.4 %), Micromonospora maritima DSM 45782 T (99.3 %), Micromonospora sediminicola DSM 45794 T (99.3 %), Micromonospora aurantiaca DSM 43813 T (99.2 %) and Micromonospora chaiyaphumensis DSM 45246 T (99.2 %). The results of OrthoANIu analysis showed the highest similarity to Micromonospora chalcea DSM 43026 T (96.4 %). However, the 16S rRNA and gyrB gene sequence-based phylogeny and phenotypic characteristics provided support to distinguish strain A38 T as a novel species. On the basis of the results presented here, we propose to classify strain A38 T (=LMG 30467 T =CECT 30034 T ) as the type strain of the novel species Micromonospora fluminis sp. nov.

    Abstract: The increase in antibiotic resistance demands innovative strategies to combat microorganisms. The current study evaluated the antibacterial and antivirulence effects of ethanol extracts from Persea americana seeds obtained by the Soxhlet (SE) and maceration (MaE) methods. The UHPLC-DAD-QTOF analysis showed mainly the presence of polyphenols and neolignan. Ethanol extracts were not cytotoxic to mammalian cells (CC50 〉 500 µg/mL) and displayed a moderate antibacterial activity against Pseudomonas aeruginosa (IC50 = 87 and 187 µg/mL) and Staphylococcus aureus (IC50 = 144 and 159 µg/mL). Interestingly, no antibacterial activity was found against Escherichia coli. SE and MaE extracts were also able to significantly reduce the bacterial adhesion to A549 lung epithelial cells. Additionally, both extracts inhibited the biofilm growth at 24 h and facilitated the release of internal cell components in P. aeruginosa, which might be associated with cell membrane destabilization. Real-time PCR and agarose electrophoresis gel analysis indicated that avocado seed ethanol extracts (64 µg/mL) downregulated virulence-related factors such as mexT and lasA genes. Our results support the potential of bioproducts from P. americana seeds as anti-adhesive and anti-biofilm agents.

    Abstract: Plants are phytochemical hubs containing antioxidants, essential for normal plant functioning and adaptation to environmental cues and delivering beneficial properties for human health. Therefore, knowledge on the antioxidant potential of different plant species and their nutraceutical and pharmaceutical properties is of utmost importance. Exploring this scientific research field provides fundamental clues on (1) plant stress responses and their adaptive evolution to harsh environmental conditions and (2) (new) natural antioxidants with a functional versatility to prevent and treat human pathologies. These natural antioxidants can be valorized via plant-derived foods and products. Cuba contains an enormously rich plant biodiversity harboring a great antioxidant potential. Besides opening new avenues for the implementation of sustainable agroecological practices in crop production, it will also contribute to new strategies to preserve plant biodiversity and simultaneously improve nature management policies in Cuba. This review provides an overview on the beneficial properties of antioxidants for plant protection and human health and is directed to the valorization of these plant antioxidants, emphasizing the need for biodiversity conservation.

    Abstract: To investigate the main chemical components and the anti-inflammatory activity of extracts of Adelia ricinella L. aerial parts. Methods Three extracts obtained by soxhlet extraction and ethanol/water mixtures were evaluated in their chemical composition by UPLC-DAD-MS/MS. The in vitro anti-inflammatory activity of the prepared extracts was assessed through three different assays: COX-1 and COX-2 enzymatic inhibition, cell-based COX assays on RAW264.7 macrophages (ATCC) measuring the COX-2 protein expression by Western blot and the measurement of the PGE2 concentration in the supernatants of the culture medium. Also was determinate the effect of the three extracts on the RAW 264.7 cell viability. Key findings Few differences in the phytochemical profile were found between the three prepared extracts, identifying a blend of thirteen flavonoids derived from luteolin and apigenin, with orientin as main constituent. Plant extracts (alcoholic and aqueous) did not affect the macrophage cell viability (IC50 & gt; 256 μg/ml) and significantly reduced COX-1 and COX-2 enzyme activities. Additionally, COX-2 expression and PGE2 release were suppressed after 24 h of LPS stimulation and treatment with plant extracts (8–64 µg/ml). Conclusions A. ricinella extracts showed the ability to reduce the inflammatory effect exerted by LPS in murine macrophages. However, further studies should confirm their anti-inflammatory activity.

    Abstract: Mosquitoes represent the most important agent disseminating infectious diseases like yellow fever, dengue, chikungunya, and malaria, among others. An essential strategy for its control is killing them at immature stages using industrial insecticides. However, those chemicals often generate resistance and affect the environment and human health. Agricultural and plant by-products constitute a new sustainable option to obtain harmless and eco-friendly bioinsecticides to prevent mosquitoes from spreading. This study aimed to investigate the phytochemical profile of Persea americana Mill (Lauraceae) seed extracts and their insecticide activity against Aedes aegypti at larval and pupal stages. The ethanol extracts from avocado seeds were obtained by Maceration/stirring (MaE) and Soxhlet extraction (SE) methods. The main chemical profile was determined by quantitative and UPLC assays. Insecticide activity was assessed by the exposition of mosquitoes at larval and pupal stages to seed extracts. Human cell lines were used to evaluate the cytotoxicity. Soxhlet methodology was more efficient in the extraction of P. americana seeds metabolites (42.13±1.76 mg/mL) compared with MaE (20.46±1.66 mg/mL) (p 〈 0.05). Additionally, SE showed a higher amount of polyphenols (5.12±0.18 mg/mL). The UPLC spectra analysis revealed the presence of polyphenols, mainly catechin, and neolignan constituents. Both extracts showed larvicidal and pupicidal effects, but SE was more active at lower concentrations. Moreover, no significant toxic effects on human monocytes and fibroblast cell lines were found after treatment. In sum, avocado seed by-products can be considered an eco-friendly insecticide and its use may help to substantially decrease the vector-transmitted diseases in developing countries.

    Abstract: La Bioinformática es una ciencia ha tenido un desarrollo acelerado debido a la gran disponibilidad de información biológica. Sus aplicaciones en la Biotecnología, Farmacología, Biología y Medicina hacen de esta, un área atractiva de investigación. Sin embargo, la carencia de personal calificado y la limitada incorporación de este contenido son evidentes. Se expone experiencia en la carrera Licenciatura en Biología en la asignatura optativa Introducción a la Bioinformática en el cuarto año. El tema escogido para las clases prácticas fue el análisis de la Hemoglobina. Los resultados muestran mejora en la calidad del proceso de enseñanza aprendizaje de esta asignatura, a partir de la integración de saberes de diferentes disciplinas promueve un aprendizaje autónomo y la calidad del estudio de los estudiantes.

    Abstract: Context: Several factors can affect the erythrocyte cell membrane integrity targeting important hematological alterations that can be avoided by the cytoprotective effect offered by some plant extracts. Aims: To evaluate the cytoprotective activity of Adelia ricinella L. extracts on red blood cells (RBCs) membrane after hypotonic and oxidative treatments. Methods: Total phenols and flavonoid contents were spectrophotometrically determined in three extracts: AR1 (ethanol 95%), AR2 (ethanol 50%), and AR3 (aqueous extract). Luteolin and apigenin were quantified using HPLC-DAD techniques. Hypotonic erythrocyte membrane stabilizing activity of plant extracts, their antioxidant protective effect on H2O2-induced cell damage, and cytotoxicity on Vero cells were explored. Control cells were treated with sodium diclofenac or ascorbic acid. Results: AR2 extract showed the highest values of total phenols/flavonoids, as well as, for luteolin and apigenin with 207.5 and 1.86 µg/mL respectively. The extracts did not exert spontaneous hemolysis following the INVITOX protocol, presumably by the protective effect of high flavonoid content. A concentration-dependent pattern was observed on the hypotonic erythrocyte membrane stabilizing assay, in which both ethanol extracts but mainly AR1 (IC50 = 16.46 µg/mL) showed a significant activity with lower IC50 values than diclofenac-control group. On the other hand, AR2 (IC50 = 17.49 µg/mL) displayed the most potent cytoprotective effect on RBCs after H2O2-induced cell damage. Adelia ricinella extracts were not cytotoxic to mammalian Vero cells (IC50 〉 256 μg/mL). Conclusions: The study suggests that Adelia ricinella extracts can promote erythrocyte cytoprotection by protecting both membrane layers, thus preventing potential hematological alterations induced by oxidizing damage and probably, in inflammation-related diseases.

    Abstract: Pleurotus ostreatus mushroom preparations have been investigated because of their ability to modulate the immune function. However, there is still no consensus regarding the activation and polarizing effect on macrophages by Pleurotus-derived bioproducts. This study examined the immune-activating effect of a mycelium-derived P. ostreatus aqueous extract (HW-Pm) on macrophage functions, by means of the determination of nitric oxide (NO) production, the mRNA expression of inducible nitric oxide synthase (iNOS), Arginase-1 and FIZZ and the cytokine levels. The phagocytic activity and the activation of NF-κB in U937 reporter cells were also investigated. No cytotoxicity was observed in macrophages treated with HW-Pm (IC50 〉 1024 μg/mL) by the resazurin test. HW-Pm induced high levels of NO production and iNOS expression in macrophages. In contrast, HW-Pm did not induce Arginase-1 and FIZZ mRNA expressions. The mushroom extract increased TNF-α and IL-6 production and the phagocytic function in murine macrophages. It also stimulated the activation of the NF-κB promoter. The P. ostreatus mycelium extract has a potential application as a natural immune-enhancing agent, by targeting macrophage activation towards the classically activated subset and stimulating macrophage-mediated innate immune responses.
