    Abstract: As an essential infrastructure, scientific data repositories play an important role in promoting the practice of open research data. As a bridge between polices and researchers, it makes the data sharing possible. However, it is a challenge to build a repository to be trustworthy and in compliant with FAIR principles. More and more guidelines on how to select a repository and what a trustworthy repository should be like have been brought up. The research elaborates several popular principles and extracts their common requirements. Upon those, the paper analyzes the repositories registered on the website of re3data and summarizes their current development. Moreover, the research focuses on the featured practices in some typical repositories, including domain-specific and generalist data repositories. At last, the research analyzes the development trend of international scientific data repositories in terms of trustworthiness, openness, and ecologization, which can be used as an instructive reference for the construction and development of similar platforms in China.

    Abstract: Uncovering genetic variation through resequencing is limited by the fact that only sequences with similarity to the reference genome are examined. Reference genomes are often incomplete and cannot represent the full range of genetic diversity as a result of geographical divergence and independent demographic events. To more comprehensively characterize genetic variation of pigs ( Sus scrofa ), we generated de novo assemblies of nine geographically and phenotypically representative pigs from Eurasia. By comparing them to the reference pig assembly, we uncovered a substantial number of novel SNPs and structural variants, as well as 137.02-Mb sequences harboring 1737 protein-coding genes that were absent in the reference assembly, revealing variants left by selection. Our results illustrate the power of whole-genome de novo sequencing relative to resequencing and provide valuable genetic resources that enable effective use of pigs in both agricultural production and biomedical research.

    Abstract: Digital health applications have been shown to be effective in the management of chronic diseases with simple treatment targets. The potential clinical value of digital health applications in rheumatoid arthritis (RA) has not been well studied. OBJECTIVE To investigate whether assessing patient-reported outcomes using digital health applications could result in disease control for patients with RA. DESIGN, SETTING, AND PARTICIPANTS This is a multicenter, open-label randomized clinical trial in 22 tertiary hospitals across China. Eligible participants were adult patients with RA. Participants were enrolled from November 1, 2018, to May 28, 2019, with a 12-month follow-up. The statisticians and rheumatologists who assessed disease activity were blinded. Investigators and participants were not blind to group assignment. Analysis was conducted from October 2020 to May 2022. INTERVENTIONS Participants were randomly assigned at a 1:1 ratio (block size of 4) to a smart system of disease management group (SSDM) or a conventional care control group. Upon the completion of the 6-month parallel comparison, patients in the conventional care control group were instructed to use the SSDM application for an extension of 6 months. MAIN OUTCOMES AND MEASURES The primary outcome was the rate of patients with disease activity score in 28 joints using the C-reactive protein (DAS28-CRP) of 3.2 or less at month 6. RESULTS Of 3374 participants screened, 2204 were randomized, and 2197 patients with RA (mean [SD] age, 50.5 [12.4] years; 1812 [82.5%] female) were enrolled. The study included 1099 participants in the SSDM group and 1098 participants in the control group. At month 6, the rate of patients with DAS28-CRP of 3.2 or less was 71.0% (780 of 1099 patients) in the SSDM group vs 64.5% (708 of 1098 patients) in the control group (difference between groups, 6.6%; 95% CI, 2.7% to 10.4%; P  = .001). At month 12, the rate of patients with DAS28-CRP of 3.2 or less in the control group increased to a level (77.7%) that was comparable with that (78,2%) in the SSDM group (difference between groups, −0.2%; 95% CI, −3.9% to 3.4%; P  = .90). CONCLUSIONS AND RELEVANCE In this randomized clinical trial of RA, the use of a digital health application with patient-reported outcomes was associated with an increase in disease control rate.     Abstract: To evaluate the efficacy and safety of HLX01, a rituximab biosimilar, as combination therapy with methotrexate in Chinese patients with active rheumatoid arthritis who had inadequate responses to methotrexate. Methods In this double-blind, placebo-controlled phase 3 trial, biologic-naïve patients with moderate-to-severe active rheumatoid arthritis and inadequate responses to methotrexate were randomized 2:1 to receive 1000 mg HLX01 or placebo intravenously on days 1 and 15. On the first day of weeks 24 and 26, patients in both groups received 1000 mg HLX01 via intravenous infusion. The primary endpoint was the American College of Rheumatology (ACR) 20 response rate at week 24. Secondary endpoints including efficacy, safety, immunogenicity, pharmacokinetics and pharmacodynamics were assessed up to week 48. Results Between 28 May 2018 and 11 September 2020, 275 patients were randomized to the HLX01 group ( n = 183) or the placebo group ( n = 92). At week 24, the proportion of patients achieving ACR20 response was significantly greater in the HLX01 group compared with the placebo group in the intention-to-treat population (60.7% vs 35.9%; P 〈 0.001) and per-protocol set (60.3% vs 37.1%; P 〈 0.001). Most secondary efficacy endpoints favoured HLX01 when assessed at weeks 12, 24, 36 and 48. Incidences of treatment-emergent adverse events were similar between groups. Infusion-related reactions occurred more frequently following the initial two doses of HLX01 than the subsequent doses. Conclusions HLX01 plus methotrexate improved clinical outcomes compared with placebo in Chinese patients with rheumatoid arthritis who had inadequate responses to methotrexate. This treatment regimen was well tolerated, showing comparable safety profiles to placebo. Trial registration ClinicalTrials.gov , NCT03522415 . Registered on 11 May 2018.

    Abstract: There is great mental stress due to the coronavirus disease 2019 (COVID-19) pandemic. However, there are no detailed psychological studies of the children with chronic kidney disease (CKD) and their guardians during the COVID-19 pandemic. Objective This study explores the psychological pressure on children with CKD and their guardians. Methods An online survey was conducted at 20 of the largest pediatric nephropathy departments in China, including the Rutter Parent Questionnaire, Self-rating Anxiety Scale (SAS), and Self-rating Depression Scale (SDS). Overall, 885 children (589 children with CKD associated with 296 children of the control group) completed the survey together with their guardians. Results There was no statistical difference between CKD children and control children regarding their Rutter behavior scores and abnormal behaviors. Nevertheless, the abnormal behavior of children might aggravate the anxiety and depression of guardians in both CKD and control groups ( p & lt; 0.05). We confirmed that the anxiety and depression of guardians in the CKD group were both significantly higher than those in the control group ( p & lt; 0.05). The guardians in the CKD group with lower annual income were more likely to experience anxiety ( p & lt; 0.05). Furthermore, the guardians whose children were older than 11 years old might be more anxious than those who were 6–11 years old. Besides, the guardians in the CKD group who watched the news for 30–60 min daily were less likely to have depression than those who watched & lt; 10 min ( p & lt; 0.05). The subgroup results showed that the gender, the time of watching the news, the annual income of guardians, and children's age might be the most critical factors influencing guardians' psychological burden. Conclusion The guardians in the CKD group have more severe anxiety and depression during the pandemic. The children's abnormal behavior, adolescents' pressure, low household income, and the panic about the pandemic may be the main reasons for the anxiety and depression of guardians.

    Abstract: Afforestation is one of the most important forestry practices, but its impact on soil microbial communities remains poorly understood. In this study, we sampled the soil from 0–15 cm and 15–30 cm soil depths of 7-, 13-, 24-, 33-, and 53-year-old Chinese cedar (Cryptomeria japonica var. sinensis) plantations. To investigate the effect of stand age on soil microbial communities and their potential drivers, we measured phospholipid fatty acids (PLFAs) and soil physicochemical properties. At the 0–15 cm soil depth, the biomass of total PLFAs and functional microbial groups such as bacteria (B), fungi (F), Gram-negative bacteria (GN), Gram-positive bacteria (GP), actinomycetes (ACT), and arbuscular mycorrhizal fungi (AMF) increased sharply in 7- to 13-year-old stands, but then gradually leveled off in older stands. On the other hand, the biomass of total PLFAs and functional microbial groups at the 15–30 cm soil depth peaked in the 33-year-old stand. The biomass of total PLFAs and functional microbial groups was strongly influenced by stand age and soil depth, and was significantly lower at the 15–30 cm soil depth than at the 0–15 cm soil depth except for the 7-year-old stand. The F/B and fungi/total PLFAs ratios of both soil depths were markedly lower in the 13-year-old stand than in the remaining four stand ages, while the proportions of the bacterial group (GP and GN) showed contrasting trends. The biomass of all functional microbial groups and the GP/GN ratio were mainly mediated by soil organic carbon (SOC) concentration and the soil organic carbon to total phosphorus (C/P) ratio at the 0–15 cm soil depth, but primarily affected by ammonium nitrogen (NH4+-N) concentration at the 15–30 cm soil depth. The F/B ratio of the two soil depths was prominently affected by nitrate nitrogen (NO3−-N) concentration. Our results highlighted that SOC concentration and mineral N (i.e., NH4+-N and NO3−-N) concentration mainly drove changes in the soil microbial biomass and community composition with stand age in Chinese cedar plantations, and that the 13-year-old stand may be the key period for management.

    Abstract: To investigate the potential value of a contrast enhanced computed tomography (CECT)-based radiological-radiomics nomogram combining a lymph node (LN) radiomics signature and LNs’ radiological features for preoperative detection of LN metastasis in patients with pancreatic ductal adenocarcinoma (PDAC). Materials and methods In this retrospective study, 196 LNs in 61 PDAC patients were enrolled and divided into the training (137 LNs) and validation (59 LNs) cohorts. Radiomic features were extracted from portal venous phase images of LNs. The least absolute shrinkage and selection operator (LASSO) regression algorithm with 10-fold cross-validation was used to select optimal features to determine the radiomics score (Rad-score). The radiological-radiomics nomogram was developed by using significant predictors of LN metastasis by multivariate logistic regression (LR) analysis in the training cohort and validated in the validation cohort independently. Its diagnostic performance was assessed by receiver operating characteristic curve (ROC), decision curve (DCA) and calibration curve analyses. Results The radiological model, including LN size, and margin and enhancement pattern (three significant predictors), exhibited areas under the curves (AUCs) of 0.831 and 0.756 in the training and validation cohorts, respectively. Nine radiomic features were used to construct a radiomics model, which showed AUCs of 0.879 and 0.804 in the training and validation cohorts, respectively. The radiological-radiomics nomogram, which incorporated the LN Rad-score and the three LNs’ radiological features, performed better than the Rad-score and radiological models individually, with AUCs of 0.937 and 0.851 in the training and validation cohorts, respectively. Calibration curve analysis and DCA revealed that the radiological-radiomics nomogram showed satisfactory consistency and the highest net benefit for preoperative diagnosis of LN metastasis. Conclusions The CT-based LN radiological-radiomics nomogram may serve as a valid and convenient computer-aided tool for personalized risk assessment of LN metastasis and help clinicians make appropriate clinical decisions for PADC patients.

    Abstract: While the scientific research value, economic value and social value of research data have become increasingly apparent, the significance of open research data has reached a consensus. This article gives an introduction to open research data policies and measures in China, and reports on the status of constructing necessary infrastructure, specifically open data repositories. We compare open data repositories in China and Western countries in terms of scale, subject distribution, data policies, service and content operations. In addition, this article summarizes methods and motivations for data sharing among researchers in China. Finally, the paper discusses the characteristics, potential problems and challenges of China's open research data practices. We conclude with some suggestions for the future development of open research data in China from data policy, infrastructure construction, compliance with international standards and norms, credibility and influence improvement, incentives for data sharing and encouraging data sharing research practices.
