    Abstract: The emergence and spread of carbapenem-resistant Klebsiella pneumoniae (CRKP) is a serious medical problem worldwide. Acquired OXA-48-like carbapenemases encoded by plasmids are important causes of carbapenem resistance in K. pneumoniae . To explore the links between plasmids and bla OXA-48 -like genes in K. pneumoniae , we systematically analyzed the variants of bla OXA-48 -like plasmid replicon types, phylogenetic patterns, geographic distribution, conjugative transfer regions, and the genetic environments surrounding bla OXA-48 -like of 191 bla OXA-48 -like-harboring plasmids, which were identified from 4451 plasmids of K. pneumoniae downloaded from GenBank. Our results showed that seven different variants of bla OXA-48 -like genes were identified from the 191 bla OXA-48 -like-harboring plasmids in K. pneumoniae , with bla OXA-48 , bla OXA-232 , and bla OXA-181 being highly prevalent. In K. pneumoniae , bla OXA-48 was mainly carried by the composite transposon Tn 1999.2 located on IncL/M-type conjugative plasmids, which were mainly geographically distributed in Switzerland, Germany, and China. In K. pneumoniae , the bl a OXA-232 gene was mainly carried by 6.1-kb ColKP3-type mobilizable plasmids, which were mainly isolated in India. In K. pneumoniae , bla OXA-181 was mainly carried by a group of 50-kb ColKP3-IncX3 hybrid conjugative plasmids and a group of small ColKP3-type mobilizable plasmids with lengths of 5.9–9.3 kb, the former was sporadically discovered in China, South Korea, India, and Czech Republic, while the latter was almost all isolated in India. In addition, five bla OXA-245 -harboring 65.9-kb IncL plasmids of K. pneumoniae isolated in Spain were found to have the genetic context of bla OXA-245 more complicated than that of bla OXA-48 -harboring IncL/M-type plasmids, with two copies of IS 1R inserted both upstream and downstream of bla OXA-245 - lysR . These findings enhance our understanding of the genetic diversity of bla OXA-48 -like-harboring plasmids in K. pneumoniae .

    Abstract: Deuterium incorporation can effectively stabilize the chiral centers of drug and agrochemical candidates that hampered by rapid in vivo racemization. In this work, the synthetically challenging chiral-center deuteration of alcohols has been achieved via a single-electron umpolung reductive-deuteration protocol using benign D2O as deuterium source and mild SmI2 as electron donor. The broad scope and excellent functional group tolerance of this method has been showcased by the synthesis of 43 respective α-deuterioalcohols in high yields and ≥98% deuterium incorporations. The potential application of this versatile method has been exemplified in the synthesis of 6 deuterated drug derivatives, 1 deuterated human hormone, and 3 deuterated natural products. This method using D2O is greener and more efficient compared to traditional pyrophoric-metal-deuteride-mediated reductive deuterations.

    Abstract: New Delhi metallo-β-lactamases (NDMs), including at least 28 variants, are a rapidly emerging family of β-lactamases worldwide, with a variety of infections caused by NDM-positive strains usually associated with very poor prognosis and high mortality. NDMs are the most prevalent carbapenemases in Escherichia coli ( E. coli ) worldwide, especially in China. The vast majority of bla NDM cases occur on plasmids, which play a vital role in the dissemination of bla NDM . To systematically explore the relationships between plasmids and bla NDM genes in E. coli and obtain an overall picture of the conjugative and mobilizable bla NDM -positive plasmids, we analyzed the variants of bla NDM , replicon types, phylogenetic patterns, conjugative transfer modules, host STs, and geographical distributions of 114 bla NDM -positive plasmids, which were selected from 3786 plasmids from 1346 complete whole genomes of E. coli from the GenBank database. We also established links among the characteristics of bla NDM -positive plasmids in E. coli . Eight variants of bla NDM were found among the 114 bla NDM -positive plasmids, with bla NDM – 5 (74 bla NDM – 5 genes in 73 plasmids), and bla NDM – 1 (31 bla NDM – 1 genes in 28 plasmids) being the most dominant. The variant bla NDM – 5 was mainly carried by the IncX3 plasmids and IncF plasmids in E. coli , the former were mainly geographically distributed in East Asia (especially in China) and the United States, and the latter were widely distributed worldwide. IncC plasmids were observed to be the predominant carriers of bla NDM – 1 genes in E. coli , which were mainly geographically distributed in the United States and China. Other bla NDM – 1 -carrying plasmids also included IncM2, IncN2, and IncHI1. Moreover, the overall picture of the conjugative and mobilizable bla NDM -positive plasmids in E. coli was described in our study. Our findings enhance our understanding of the genetic diversity and characteristics of bla NDM -positive plasmids in in E. coli .

    Abstract: We discovered that highly regioselective H/D exchange can be achieved on the α ‐position of the pentafluorophenyl (Pfp) ester using Et 3 N as the catalyst and D 2 O as the deuterium source. Our computational and experimental results showed that the Pfp group can significantly increase the acidity of the α ‐hydrogen of the ester. This mild reaction condition tolerated a variety of functional groups. 90%∼98% yields and 91%∼98% deuterium incorporations were obtained with all the tested Pfp esters. Taking advantage of the high reactivity of Pfp esters, a series of valuable α ‐deuterio carboxylic acid derivatives, including herbicide, drugs and drug intermediates, have been synthesized using α ‐deuterio Pfp esters as precursors. magnified image

    Abstract: Functionalization of amide bond via the cleavage of a non-carbonyl, C-N σ bond remains under-investigated. In this work, a transition-metal-free single-electron transfer reaction has been developed for the C-N σ bond cleavage of N-acylazetidines using the electride derived from sodium dispersions and 15-crown-5. Of note, less strained cyclic amides and acyclic amides are stable under the reaction conditions, which features the excellent chemoselectivity of the reaction. This method is amenable to a range of unhindered and sterically encumbered azetidinyl amides.

    Abstract: Subgenome expression dominance plays a crucial role in the environmental adaptation of polyploids. However, the epigenetic molecular mechanism underlying this process has not been thoroughly investigated, particularly in perennial woody plants. Persian walnut (Juglans regia) and its wild relative, Manchurian walnut (Juglans mandshurica), are woody plants of great economic importance and are both paleopolyploids that have undergone whole-genome duplication events. In this study, we explored the characteristics of subgenome expression dominance in these 2 Juglans species and examined its epigenetic basis. We divided their genomes into dominant subgenome (DS) and submissive subgenome (SS) and found that the DS-specific genes might play critical roles in biotic stress response or pathogen defense. We comprehensively elucidated the characteristics of biased gene expression, asymmetric DNA methylation, transposable elements (TEs), and alternative splicing (AS) events of homoeologous gene pairs between subgenomes. The results showed that biased expression genes (BEGs) in 2 Juglans species were mainly related to external stimuli response, while non-BEGs were related to complexes that might be involved in signal transduction. DS genes had higher expression and more AS events while having less DNA methylation and TEs than homoeologous genes from the SS in the 2 Juglans species. Further studies showed that DNA methylation might contribute to the biased expression of gene pairs by modifying LTR/TIR/nonTIR TEs and improving the AS efficiency of corresponding precursor mRNAs in a particular context. Our study contributes to understanding the epigenetic basis of subgenome expression dominance and the environmental adaptation of perennial woody plants.

    Abstract: New Delhi metallo-β-lactamase (NDM)-producing clinical strains in Acinetobacter spp. have been recently reported in many countries and have received considerable attention. The vast majority of bla NDM cases occur on conjugative plasmids, which play a vital role in disseminating bla NDM . To characterize the conjugative plasmids bearing bla NDM genes in Acinetobacter spp., we analyzed the variants of bla NDM , conjugative transfer regions, genetic contexts of bla NDM , and the phylogenetic pattern of the 62 predicted bla NDM -positive plasmids, which were selected from 1,191 plasmids of Acinetobacter species from GenBank. We identified 30 conjugative plasmids from the 62 bla NDM -harboring plasmids in Acinetobacter species, with the oriT sites similar to plasmid pNDM-YR7 in our study, genes coding for relaxases of the MOB Q family, genes encoding type IV coupling proteins (T4CPs) of the TrwB/TraD subfamily, and VirB-like type IV secretion system (T4SS) gene clusters. The genome sizes of all 30 pNDM-YR7-like plasmids ranged from 39.36 kb to 49.65 kb, with a median size of 44.56 kb. The most common species of Acinetobacter containing the bla NDM -positive conjugative plasmids was A. baumannii , followed by Acinetobacter lwoffii and Acinetobacter indicus . Notably, pNDM-YR7 is the first report on a bla NDM -positive conjugative plasmid in Acinetobacter junii . Moreover, all 30 bla NDM -positive conjugative plasmids in Acinetobacter species were found to contain genetic contexts with the structure IS Aba14 - aph(3′)-VI -IS Aba125 - bla NDM - ble . Our findings provide important insights into the phylogeny and evolution of bla NDM -positive plasmids of Acinetobacter species and further address their role in acquiring and spreading bla NDM genes in Acinetobacter species. IMPORTANCE Conjugative plasmids harboring the bla NDM gene play a vital role in disseminating carbapenem resistance. In this study, we first report a conjugative plasmid, pNDM-YR7, in Acinetobacter junii . Based on the genomic characteristics of the bla NDM -positive pNDM-YR7, we performed in silico typing and comparative analysis of bla NDM -positive plasmids using the 1,191 plasmids of Acinetobacter species available in the NCBI RefSeq database. We analyzed the characteristics of bla NDM -positive plasmids, including the variants of bla NDM , genetic features associated with bla NDM , conjugative transfer regions, and the phylogenetic pattern of the bla NDM -positive plasmids. All 30 bla NDM -positive conjugative plasmids were found to contain an IS Aba14 - aph(3′)-VI -IS Aba125 - bla NDM - ble region. This study provides novel insights into the phylogeny and evolution of bla NDM -harboring conjugative plasmids and contributes to the repertoire of knowledge surrounding bla NDM -positive plasmids in the genus Acinetobacter .
