    Abstract: The Hemiptera (aphids, cicadas, and true bugs) are a key insect order, with high diversity for feeding ecology and excellent experimental tractability for molecular genetics. Building upon recent sequencing of hemipteran pests such as phloem-feeding aphids and blood-feeding bed bugs, we present the genome sequence and comparative analyses centered on the milkweed bug Oncopeltus fasciatus , a seed feeder of the family Lygaeidae. Results The 926-Mb Oncopeltus genome is well represented by the current assembly and official gene set. We use our genomic and RNA-seq data not only to characterize the protein-coding gene repertoire and perform isoform-specific RNAi, but also to elucidate patterns of molecular evolution and physiology. We find ongoing, lineage-specific expansion and diversification of repressive C2H2 zinc finger proteins. The discovery of intron gain and turnover specific to the Hemiptera also prompted the evaluation of lineage and genome size as predictors of gene structure evolution. Furthermore, we identify enzymatic gains and losses that correlate with feeding biology, particularly for reductions associated with derived, fluid nutrition feeding. Conclusions With the milkweed bug, we now have a critical mass of sequenced species for a hemimetabolous insect order and close outgroup to the Holometabola, substantially improving the diversity of insect genomics. We thereby define commonalities among the Hemiptera and delve into how hemipteran genomes reflect distinct feeding ecologies. Given Oncopeltus ’s strength as an experimental model, these new sequence resources bolster the foundation for molecular research and highlight technical considerations for the analysis of medium-sized invertebrate genomes.

    Abstract: Dengue fever is a public health issue of global concern. Taiwan is located in the subtropical region featuring humid and warm weather, which is conducive to the breeding of mosquitoes and flies. Together with global warming and the increasing frequency of international exchanges, in addition to the outbreak of pandemics and dengue fever, the number of people infected has increased rapidly. This study is dedicated to the development of a new mosquito-trapping system. Research has shown that specific wavelengths, colors, and temperatures are highly attractive to both Aedes aegypti and Aedes albopictus. In this study, we create equipment which effectively improves the trapping capabilities of mosquitoes in a wider field. The design of the special Secondary Freeform Lens Device (SFLD) is used to expand the range for trapping mosquitoes and create illumination uniformity; it also directs light downward for the protection of users’ eyes. In addition, we use the correct amount of stearic acid as a mosquito attractant to allow a better control effect against mosquitoes during the day. In summary, when the UV LED mosquito trapping system is combined with a quadratic free-form lens, the experimental results show that the system can extend the capture range to 100 π m2 in which the number of captured mosquitoes is increased by about 350%.

    Abstract: Dengue fever is the most serious vector-borne disease in Asia. There are still no dengue vaccines or therapeutic drugs, and vector-mosquito control is the main prevention and treatment method. The prevention and control of dengue-fever vector mosquitoes in Taiwan is still dominated by larval control. However, the removal of the source of mosquito-borne diseases has not been fully implemented, and the removal process of vector-borne mosquitoes cannot keep up with their breeding rate. In addition, chemical agents used in the elimination of pathogenic mosquitoes may cause mosquito resistance and environmental pollution. Therefore, it is important to develop new prevention and control technologies. This study is dedicated to the development of a mosquito trapping optical system with high efficiency and low glare that is safe for humans. The system is mainly equipped with ultraviolet light-emitting diodes (UV-LED), a freeform-surfaced optical reflector, and a photocatalyst. The reflector can lead light downward for the protection of user eyes, and expand the range of trapping mosquitoes to 225 π m2. Based on practical experiments, captured mosquitoes increased by about 450% through the proposed system compared to conventional traps using UV LED. In addition, the proposed system is shown to be 45% more enhanced in trapping capability after a photocatalyst (titanium dioxide) coating is applied to its reflector.

    Abstract: In this study, our proposed ultraviolet light-emitting diode (UV LED) mosquito-trapping lamp is designed to control diseases brought by insects such as mosquitoes. In order to enable the device to efficiently catch mosquitoes in a wider area, a secondary freeform lens (SFL) is designed for UV LED. The lens is mounted on a 3 W UV LED light bar as a mosquito-trapping lamp of the new UV LED light bar module to achieve axially symmetric light intensity distribution. The special SFL is used to enhance the trapping capabilities of the mosquito-trapping lamp. The results show that when the secondary freeform surface lens is applied to the experimental outdoor UV LED mosquito-trapping lamp, the trapping range can be expanded to 100π·m2 and the captured mosquitoes increased by about 300%.

    Abstract: The current state of the COVID-19 pandemic is a global health crisis. To fight the novel coronavirus, one of the best-known ways is to block enzymes essential for virus replication. Currently, we know that the SARS-CoV-2 virus encodes about 29 proteins such as spike protein, 3C-like protease (3CLpro), RNA-dependent RNA polymerase (RdRp), Papain-like protease (PLpro), and nucleocapsid (N) protein. SARS-CoV-2 uses human angiotensin-converting enzyme 2 (ACE2) for viral entry and transmembrane serine protease family member II (TMPRSS2) for spike protein priming. Thus in order to speed up the discovery of potential drugs, we develop DockCoV2, a drug database for SARS-CoV-2. DockCoV2 focuses on predicting the binding affinity of FDA-approved and Taiwan National Health Insurance (NHI) drugs with the seven proteins mentioned above. This database contains a total of 3,109 drugs. DockCoV2 is easy to use and search against, is well cross-linked to external databases, and provides the state-of-the-art prediction results in one site. Users can download their drug-protein docking data of interest and examine additional drug-related information on DockCoV2. Furthermore, DockCoV2 provides experimental information to help users understand which drugs have already been reported to be effective against MERS or SARS-CoV.     Abstract: To enhance driving safety at night, a new freeform-surface street light luminaire was proposed and evaluated in this study that meets the requirements of the International Commission on Illumination (CIE) M3 class standard for road lighting. The luminaire was designed using simulations to optimize the location of the bulb according to the requirements of the standard. The light source IES file was experimentally obtained for the optimized luminaire prototype with a 150 W ceramic metal halide lamp using an imaging goniophotometer. The trial road lighting simulation results computed by the lighting software DIALux indicated that the proposed luminaire provided an average road surface brightness of 1.1 cd/m 2 (compared to a minimum requirement of 1.0 cd/m 2 ), a brightness uniformity of 0.41 (compared to a minimum requirement of 0.4), a longitudinal brightness uniformity of 0.64 (compared to a minimum requirement of 0.6), and a glare factor of 7.6% (compared to a maximum limit of 15%). The findings of the image goniophotometer tests were then confirmed by the results of a certified mirror goniophotometer test conducted by the Taiwan Accreditation Foundation (TAF). The results of this study can be used to provide improved street lighting designs to meet enhanced international standards.

    Abstract: Uterine mesenchymal tumors are genetically heterogenous; those with uniform cytomorphology, best exemplified by endometrial stromal tumors, often contain various fusion genes. Novel fusions involving ESR1 and GREB1 , key factors in sex hormone pathways, have been implicated in rare uterine mesenchymal tumors. Particularly, the fusions between 5′- ESR1/GREB1 and 3′- NCOA2/NCOA3 were recently identified in 4 uterine tumors resembling ovarian sex-cord tumor (UTROSCT). By RNA sequencing, pathology review, and FISH screening, we identified 4 uterine sarcomas harboring rearranged GREB1 , including GREB1-NCOA2 and the novel GREB1-NR4A3 , GREB1-SS18 , and GREB1-NCOA1 , validated by RT-PCR and/or FISH. They occurred in the myometrium of postmenopausal women and were pathologically similar despite minor differences. Tumor cells were generally uniform and epithelioid, with vesicular nuclei and distinct to prominent nucleoli. Growth patterns included solid sheets, trabeculae/cords, nests, and fascicles. Only 1 tumor showed small foci of definitive sex-cord components featuring well-formed tubules, retiform structures, Leydig-like cells, and lipid-laden cells and exhibiting convincing immunoreactivity to sex-cord markers (calretinin, α-inhibin, and Melan-A). In contrast, all the 4 classic UTROSCT we collected occurred in premenopausal patients, consisted predominantly of unequivocal sex-cord elements, prominently expressed multiple sex-cord markers, and harbored ESR1-NCOA3 fusion. Combined with previously reported cases, GREB1 -rearranged tumors involved significantly older women ( P =0.001), tended to be larger and more mitotically active, showed more variable and often inconspicuous sex-cord differentiation, and appeared to behave more aggressively than ESR1 -rearranged UTROSCT. Therefore, these 2 groups of tumors might deserve separate consideration, despite some overlapping features and the possibility of belonging to the same disease spectrum.

    Abstract: To investigate the risk and genetic association of oxcarbazepine-induced cutaneous adverse reactions (OXC-cADRs), including Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN), in Asian populations (Chinese and Thai). Methods: We prospectively enrolled patients with OXC-cADRs in Taiwan and Thailand from 2006 to 2014, and analyzed the clinical course, latent period, drug dosage, organ involvement, complications, and mortality. We also investigated the carrier rate of HLA-B*15:02 and HLA-A*31:01 of patients with OXC-cADRs and compared to OXC-tolerant controls. The incidence of OXC-SJS/TEN was compared with carbamazepine (CBZ)–induced SJS/TEN according to the nationwide population dataset from the Taiwan National Health Insurance Research Database. Results: We enrolled 50 patients with OXC-cADRs, including 20 OXC-SJS/TEN and 6  drug reaction with eosinophilia and systemic symptoms, of Chinese patients from Taiwan and Thai patients from Thailand. OXC-cADRs presented with less clinical severity including limited skin detachment (all ≦5%) and no mortality. There was a significant association between HLA-B*15:02 and OXC-SJS ( p = 1.87 × 10 −10 ; odds ratio 27.90; 95% confidence interval [CI] 7.84–99.23) in Chinese and this significant association was also observed in Thai patients. The positive and negative predictive values of HLA-B*15:02 for OXC-SJS/TEN were 0.73% and 99.97%, respectively. HLA-A*31:01 was not associated with OXC-cADRs. The incidence and mortality of OXC-SJS/TEN was lower than CBZ-STS/TEN in new users ( p = 0.003; relative risk 0.212; 95% CI 0.077–0.584). Conclusions: Our findings suggest that HLA-B*15:02 is significantly associated with OXC-SJS in Asian populations (Chinese and Thai). However, the severity and incidence of OXC-SJS/TEN are less than that of CBZ-SJS/TEN. The need for preemptive HLA-B*15:02 screening should be evaluated further.
