Abstract: Molds can carry out amylase production, but the production of the enzyme is affected by a carbon catabolite repression system, thus reducing amylase production. This study aimed to increase amylase production in Aspergillus tamarii FNCC 6151 with irradiation by gamma rays. Spore suspensions were irradiated at 100, 200, 300, 400 and 500 Gy and the survival of the spores was determined. The spore suspension irradiated at 400 Gy was diluted and grown on medium plates containing starch as a carbon source and 2% glucose as a repressor. After staining with iodine, colonies showing clear zones around them were purified and grown in liquid media for four days and their alpha-amylase and glucoamylase activities were analyzed. The mutant Mut1 showed a clear zone. This mutant produced 1.127 U/mL of alpha-amylase and 0.011 U/mL of glucoamylase. Meanwhile, the parental strain produced 0.68 U/mL of alpha-amylase and 0.079 U/mL of glucoamylase. The random mutagenesis using gamma-ray into A. tamarii FNCC 6151 increased alpha-amylase and glucoamylase production by 1.8- and 1.4-fold respectively.

Abstract: Skin damage among healthcare workers has been reported by many centers around the world. Occupational hand dermatitis is one of the most commonly known occupational skin diseases and a socially significant health issue. The use of gloves is one of the risk factors for the occurrence and/or aggravation of hand dermatitis. This cross-sectional study involved healthcare workers in 14 referral hospitals for COVID-19 throughout Indonesia. Questionnaires were distributed to the participants, which consisted of the subject’s characteristics, glove-related skin problems, history of glove use, and clinical history. This study involved a total of 845 healthcare workers. Approximately 156 healthcare workers (18.46%) had glove-induced hand dermatitis during the pandemic. Itchy skin was the most common symptom (44.23%), and the palm was the most frequently complained area (48.72%). There was a significant association between glove use and glove-induced hand dermatitis among healthcare workers. In particular, equal to or more than 2 hours per day of glove use was significantly associated with hand dermatitis. Glove-induced hand dermatitis also had a significant association with the subject’s history of atopic dermatitis and previous history of hand dermatitis. The use of gloves by healthcare workers should be considered carefully, especially in individuals at increased risk, including those who use gloves for 2 hours or more per day and those who have a history of atopic or hand dermatitis, in order to prevent the incidence of glove-induced hand dermatitis among healthcare workers, as well as to provide a safe working environment.

Abstract: Storing and preserving onion using pesticides is common in Brebes District. The study determined the use of ozone to reduce microorganisms and to maintain the onion quality during months of post-harvesting. There were three chambers with 2.5 m 3 in size, and with a capacity of 20 kg onion used for this study. Each chamber received different treatment for 2 months; chamber I: no treatment; chamber II: pesticide; chamber III: ozone. The ozone treatment maintained better temperature and humidity for preservation. Ozone treatment was also observed to maintain low level of mass damage (1.8%) as compared to control and pesticide (6.5% and 2.4%). The protein, ash, fat and vit B1 of onions were remained high in ozone-treated chamber. Total Plate Count (CFU/mL) and the Mold Yeast Count (colonies/mL) decreased on day 30 and 60 after pesticides and ozone treatment. This study showed the importance of ozone treatment to maintain better quality of onion after months of storage as compared to pesticide treatment. Therefore, ozone can be a potential use to replace the pesticide for preservation at post-harvesting.

Abstract: Background: Tuberculosis (TB) remains one of the top ten causes of mortality worldwide. It is caused by a single infectious agent, Mycobacterium tuberculosis. Indonesia has the second highest cases globally and has a high burden of rifampicin-resistant (RR-TB). Xpert MTB/RIF has been used as a frontline test to detect RR-TB to improve treatment efficacy. Objective: This study aims to summarise the bacteriologic conversion rate as the patient’s cured hallmark and related factors among adults confirmed to have pulmonary tuberculosis in a referral hospital in Aceh Province. Method: The authors assessed 1,244 bacteriologic test results of sputum samples from TB presumptive patients from January 1st to December 31st 2018, in the Clinical Microbiology Laboratory and evaluated bacteriologic conversion. Result: There were 361 bacteriologic-confirmed TB diagnoses, where 43 (11.9%) were RR-TB. Most TB patients (26.3%) were in close contact with active TB patients. Conclusion: The study showed TB patients' low follow-up rate and bacteriologic conversion. The high prevalence of TB among close contacts suggests a need for constant surveillance of this high-risk group to end the TB epidemic. Male, young adult, and Aceh Besar residents have observed factors in sputum conversion results.

Abstract: Regional Original Income is revenue that is obtained by the regional government for the implementation of government and services to the community. One source of local revenue is the restaurant tax. However, often the tax is not optimal because it is considered to result in high economic costs and less support for business or investment including restaurant tax. The purpose of this study was to determine the provisions of administrative sanctions against restaurant tax offenders. This research uses normative juridical research with data in the form of books, journals, research results, and related laws and regulations. The results showed that restaurant tax as a source of local revenue was regulated by Regional Regulation. Provisions on administrative sanctions are regulated in regional regulations by adjusting the conditions in each area. Local regulations on restaurant taxes should contain clear and strict sanctions so they can be effective. Keywords: administrative sanctions, restaurant tax, local own income
