    Abstract: The Hippo signaling pathway is associated with cell proliferation and organ size, and its transcriptional coactivator Yes‐associated protein (YAP), emerges as a crucial oncoprotein in multiple cancers. It was increasingly recognized that nonreceptor tyrosine phosphatase 14 (PTPN14) was relevant to the cell membrane and cytoskeleton, and had a critical effect on cell adhesion, growth, and actin cytoskeleton organization. Furthermore, PTPN14 was also certified to operate the translocation and phosphorylation of YAP. The present experiment was aimed to explore the impact of PTPN14 on gastric cancer (GC) cell proliferation and migration through regulating the phosphorylation of YAP. Methods The pEGFP‐N1‐PTPN14 recombinant plasmid was stably transfected into three differentiation degrees GC cell lines, including MKN‐28, SGC‐7901, and BGC‐823. Quantitative reverse transcription‐polymerase chain reaction and Western blot assay were performed to analyze the messenger RNA (mRNA) and protein levels. The proliferative and migratory capacity of cells was appraised by Cell Counting Kit‐8 assay and transwell chamber. Results Compared with the normal control and vector transfection group, the capacity of these three cell lines, which transfected with the pEGFP‐N1‐PTPN14 to proliferate and migrate in vitro was increased obviously ( P   〈  .05). There was no YAP mRNA detected in MKN‐28 cell line. Meanwhile, after transfecting the pEGFP‐N1‐PTPN14 plasmid, the mRNA level of YAP in SGC‐7901 was reduced ( P   〈  .05), and it was increased in BGC‐823 ( P   〈  .05). The YAP protein level in SGC‐7901 and BGC‐823 has no apparent transformation by transfecting, but the protein level of phospho‐Ser127 YAP and phospho‐Ser397 YAP is upregulated ( P   〈  .05). Conclusion PTPN14 could enhance the proliferative and migratory ability of GC cells by promoting the YAP phosphorylation in the Hippo signaling pathway. Taken together, PTPN14 might be involved in the occurrence and development of GC and become a molecular regulator to treat GC.

    Abstract: Helicobacter pylori (H. pylori) infection is a major risk factor for gastric cancer ( GC ); however, only a minority of infected individuals develops GC . We aim to assess the association between serostatus of antibody against H. pylori flagellin A (FlaA) and risk of GC and to evaluate the value of serum FlaA antibody as a novel screening biomarker for GC risk. Methods A hospital‐based case–control study including 232 cases and 264 controls was conducted. Logistic regression was adopted to analyze the association between the serostatus of FlaA antibody and risk of GC . Serum FlaA antibody was measured by an enzyme‐linked immunosorbent assay ( ELISA ). Receiver operating characteristic ( ROC ) curve was used to evaluate the screening efficacy and to identify a cutoff point of serum FlaA antibody level. Results Helicobacter pylori infection was associated with an increased risk of GC ( p  =   .007). A positive association between serum FlaA antibody and GC risk was observed in overall subjects and H. pylori‐ positive subjects ( OR [95% CI ]: 6.8 [4.3–10.7] and 6.9 [3.6–13.4], respectively; p  〈   .001). The seropositivity of FlaA antibody was strongly related to GC risk in a dose‐dependent manner ( p for trend  〈  .001). The optimal cutoff value ( OD ) was 0.1403, providing a sensitivity of 74.1% and a specificity of 64.4%. The area under the ROC curve ( AUC ) was 0.74 in overall subjects and 0.73 in H. pylori ‐positive subjects, respectively. Conclusions FlaA was an independent risk factor for H. pylori ‐related GC . Serum FlaA antibody may serve as a novel noninvasive biomarker for early detection of GC .

    Abstract: Transmembrane and intracellular signal transduction of G protein is closely related to the pathophysiology of Alzheimer's disease (AD). Objective To explore the effects of Sanjiao acupuncture on G protein signal transduction pathways in the pathogenesis of AD. Methods 36 senescence-accelerated (SAM) prone 8 mice were divided into three groups that remained untreated (SAMP8, n=12) or received Sanjiao acupuncture (SAMP8+SA, n=12) or control acupuncture (SAMP8+CA, n=12). An additional control group of SAM resistant 1 mice was included (SAMR1 group, n=12). Morris water maze tests were used to investigate learning and memory abilities. Immunoprecipitation and Western blotting were used to study expression of G protein subunits and their activities in the cortex/hippocampus. Results Behavioural analysis showed that acupuncture attenuated the severe cognitive deficits observed in untreated/CA-treated SAMP8 mice. The findings of the G protein activation assays via immunoprecipitation and Western blots were that the physiologically coupled activation rate (PCAR) and maximal coupled activation rate (MCAR) of Gα s and Gα i were decreased in the cortex of SAMP8 vs SAMR1 mice. Sanjiao acupuncture induced an upregulation in the PCAR of Gα s and Gα i . In the hippocampus of untreated SAMP8 mice, the PCAR of Gα s and MCAR of both Gα s and Gα i declined, and Sanjiao acupuncture was associated with an upregulation in the MCAR of Gα s and Gα i . There were no significant differences in Gα s and Gα i expression between the groups. Conclusions Sanjiao acupuncture attenuates cognitive deficits in a mouse model of AD via upregulation of G protein activity and stabilisation of the cellular signal.

    Abstract: In order to analyze the influence of meso-structure and meso-parameters on deep granite characteristics, a construction method of variable radius proportional clump model was proposed with particle flow method. The clump particle flow structure was constructed which suited the mechanical characteristics of granite. The deep cavern numerical calculation model of gradual particle density was constructed using the variable radius proportional clump model construction method, and the macroscopic fracture law of deep cavern surrounding rock was analyzed. The results show that meso parameters have lower effects on the compressive and tensile ratios of particle structures in the ball and clump models. It is also found that clump structure is greatly influenced by particle proportion and size while ball model is only slightly influenced by particle size. Furthermore, the compressive and tensile strength curves and fracture modes of numerical simulations and laboratory tests are in good agreement. In addition, the calculated results of the tunnel after simulated excavation are very close to the engineering practice. There are obvious shear failure areas on the right vault and left wall of the tunnel, and the shape and depth of the fracture area are basically the same.

    Abstract: As we know, grid quality can produce big effect on the numerical precision of 3D forward modeling. How to generate high-quality grids becomes the key for high-accuracy EM responses. So we develop a weighted adaptive finite-element method based on hybrid posterior error estimation in combination with unstructured vector finite-element method and Backward Euler scheme. We introduce a weighting factor to adjust the relative weight of posterior errors defined by the continuities of normal current density and tangential magnetic field to create an effective mesh. The numerical experiments on a conductive cube model show that our algorithm performs better than the traditional adaptive method.

    Abstract: Semantic segmentation is a key technology for remote sensing image analysis widely used in land cover classification, natural disaster monitoring, and other fields. Unlike traditional image segmentation, there are various targets in remote sensing images, with a large feature difference between the targets. As a result, segmentation is more difficult, and the existing models retain low accuracy and inaccurate edge segmentation when used in remote sensing images. This paper proposes a multi-attention-based semantic segmentation network for remote sensing images in order to address these problems. Specifically, we choose UNet as the baseline model, using a coordinate attention-based residual network in the encoder to improve the extraction capability of the backbone network for fine-grained features. We use a content-aware reorganization module in the decoder to replace the traditional upsampling operator to improve the network information extraction capability, and, in addition, we propose a fused attention module for feature map fusion after upsampling, aiming to solve the multi-scale problem. We evaluate our proposed model on the WHDLD dataset and our self-labeled Lu County dataset. The model achieved an mIOU of 63.27% and 72.83%, and an mPA of 74.86% and 84.72%, respectively. Through comparison and confusion matrix analysis, our model outperformed commonly used benchmark models on both datasets.
