    Abstract: Larotrectinib (Lar) is a highly selective and potent small‐molecule inhibitor used in patients with tropomyosin receptor kinase (TRK) fusion‐positive cancers, including colon cancer. However, the underlying molecular mechanisms specifically in patients with colon cancer have not yet been explored. Our data showed that Lar significantly suppressed proliferation and migration of colon cancer cells. In addition, Lar suppressed the epithelial–mesenchymal transition (EMT) process, as evidenced by elevation in E‐cadherin (E‐cad), and downregulation of vimentin and matrix metalloproteinase (MMP) 2/9 expression. Furthermore, Lar was found to activate autophagic flux, in which Lar increased the ratio between LC3II/LC3I and decreased the expression of p62 in colon cancer cells. More importantly, Lar also increased AMPK phosphorylation and suppressed mTOR phosphorylation in colon cancer cells. However, when we silenced AMPK in colon cancer cells, Lar‐induced accumulation of autolysomes as well as Lar‐induced suppression of the EMT process were significantly diminished. An in vivo assay also confirmed that tumour volume and weight decreased in Lar‐treated mice than in control mice. Taken together, this study suggests that Lar significantly suppresses colon cancer proliferation and migration by activating AMPK/mTOR‐mediated autophagic cell death.

    Abstract: Laser sealing for vacuum plate glass is a key step in developing the cost-effective smart vacuum-glass window for the drive towards net-zero energy buildings. In this paper, the pores, cracks, and interface with laser welding are analyzed in depth using PbO-TiO2-SiO2-RxOy system sealing solder to prepare vacuum flat glass. The microstructure of the sealing layer was analyzed by a BX41M-LED metallographic microscope, and the interfacial bonding characteristics were observed by thermal field emission scanning electron microscopy (SEM). The solder was analyzed by an energy spectrometer, and the influence of laser power, sealing temperature, and sealing speed on the gas holes and the crack sand interface separation of the sealing layer are reported. The results show that when the laser power reached 80 W at the welding speed of 2 mm/s, the bulk solder disappeared to most of the quantity and the sealing surface density was higher, due to which negligible pores and micro cracks were found. Thus, the sealing quality of the sealing layer is considered to be suitable when the temperature of 470 °C was maintained and the solder has 68.93% of Pb and 3.04% Si in the atom fraction to achieve the wet the glass substrate surface whilst improving the bonding quality.

    Abstract: A novel dual-wideband double-layer magnetoelectric dipole unidirectional antenna with a modified horned reflector for 2G/3G/LTE applications is proposed. Firstly, a double-layer electric dipole structure is presented to provide a dualwideband, whose folded lower layer mainly serves the lower frequency band while the inclined upper layer works for the upper frequency band. In addition, to reduce the size of the antenna and improve impedance matching, a new feeding structure designed with inverted U-shaped and tapered line is introduced. Finally, a modified horn-shaped reflector, instead of a ground plane, is employed to achieve stable and high gains. The antenna prototype can achieve a bandwidth of 24.4% (790 MHz–1010 MHz) with a stable gain of 7.2 ± 0.6 dBi for the lower band, and a bandwidth of 67.3% (1.38 GHz–2.78 GHz) with a gain of 7.5 ± 0.8 dBi for the upper band covering all the frequency bands for 2G/3G/LTE systems. To the best of our knowledge, it is the first double-layer magnetoelectric dipole antenna proposed. Compared with the existing ME dipole antennas, the proposed antenna, which is completely made of copper, can be easily fabricated at low cost and thus is practicable for 2G/3G/LTE applications.

    Abstract: The neutron shielding glass is widely used in nuclear/fusion plants. To improve its temperature resistance and heat insulation, a Gadolinium (Gd)-containing laminate vacuum multiple glass is proposed by using the vacuum insulation method. A 3D finite element model validated by theoretical calculation was developed to analyse the heat transfer path and numerical simulation of the multiple glass was carried out to obtain the temperature distribution and the maximum temperatures of the organic glass in relation to dynamic working temperatures, the sealing agent width, view size, and vacuum thermal conductivity. The results show that the vacuum layer between common glasses can make the work temperature of neutron shielding glass increase. The multiple glass has good heat-shielding performance and it is expected to work in a high-temperature environment. In addition, the vacuum layer between the common glasses and the sealing agent width decay with respect to the view size and vacuum thermal conductivity show an increase in the working temperature of the neutron shielding glass. It was concluded that the order of affecting the temperatures of the organic glass follows the pattern of: view size 〉 vacuum thermal conductivity 〉 sealing agent width.

    Abstract: β barrel outer membrane proteins (β-OMPs) play vital roles in mitochondria, chloroplasts, and Gram-negative bacteria. Evolutionarily conserved complexes such as the mitochondrial sorting and assembly machinery (SAM) mediate the assembly of β-OMPs. We investigated the SAM-mediated assembly of the translocase of the outer membrane (TOM) core complex. Cryo–electron microscopy structures of SAM–fully folded Tom40 and the SAM-Tom40/Tom5/Tom6 complexes at ~3-angstrom resolution reveal that Sam37 stabilizes the mature Tom40 mainly through electrostatic interactions, thus facilitating subsequent TOM assembly. These results support the β barrel switching model and provide structural insights into the assembly and release of β barrel complexes.

    Abstract: Highlights An improved YOLOv5s deep learning model was used to identify peppers in complex background. The deep-level features on 3D (O-XYZ) coordinate of peppers were extracted using RealSense depth camera. An image database set of pepper in different scenes was established. A pepper recognition and location system were constructed based on improved YOLOv5s network. The proposed method achieved a mean average precision of 95.6% and minimum depth error of 0.001 m. Abstract. In order to investigate the impact of different scenes on the recognition performance and obtain the location information of picking targets, the recognition and location system based on improved YOLOv5s network and RealSense depth camera was constructed in this study. An image database in different scenes was established including light intensity, occlusion and overlap degree of pepper. An improved YOLOv5s deep learning model with bidirectional feature pyramid network (BiFPN) was used for the deep feature extraction and high-precision detection of pepper, and the effects of different scenes on recognition accuracy of the model were studied. The results showed that mean average precision (mAP) of YOLOv5s model reached 0.956, which was respectively 6.1%, 9.3%, 44.4%, and 8.2% higher than that of YOLOv4, YOLOv3, YOLOv2, and Faster R-CNN model. The model had good robustness under daytime and evening scenes with the mAP value higher than 0.9. The detection accuracy of the model in the leaf occlusion scenes was better than that of fruit overlap. The detection error was 0.001m which could not affect the picking positioning precision when the Z value of three-dimensional coordinates (O-XYZ) of pepper was 0.2 m. The improved algorithm can accurately recognize and extract three-dimensional coordinates of pepper, which reduces the calculations by eliminating lots of duplicate and redundant prediction boxes and provides a reference for trajectory planning of pepper picking operation.
