    Abstract: Nos últimos dez anos, tem-se verificado o aumento do interesse pela terapia homeopática devido a sua inclusão no SUS, oficializada na "Política Nacional de Práticas Integrativas e Complementares” (PNPIC). A Universidade Federal do Rio de Janeiro (UFRJ) possui dois hospitais universitários de grande importância no Sistema Único de Saúde (SUS): o Hospital Universitário Clementino Fraga Filho (HUCFF) e o Instituto de Puericultura e Pediatria Martagão Gesteira (IPPMG). O presente estudo avaliou, ao todo, a opinião de 142 médicos, sendo 110 do HUCFF e 32 do IPPMG, sobre a homeopatia, através de um questionário específico. Após coleta e tratamento estatístico dos dados, os resultados indicaram que apenas 14 médicos já tinham indicado e/ou utilizado a terapia homeopática anteriormente, apesar da inexistência de ambulatório de homeopatia em ambos os hospitais avaliados. A análise dos questionários evidenciou, também, que a maioria dos médicos entrevistados, além de desconhecerem a terapia homeopática, acham irrelevante que o serviço de homeopatia seja oferecido nesses hospitais. Dessa forma, verificamos a necessidade de inclusão da homeopatia na graduação de medicina da UFRJ com a ampliação do currículo dos médicos da UFRJ, como já é feito na UNIRIO e na UFF, e de mapear a percepção dos profissionais da saúde acerca da homeopatia, fornecendo dados que favoreceram a implantação da homeopatia na Atenção Básica à Saúde, como previsto na PNPIC.

    Abstract: Lettuce seeds bioassays have been used in many different tests such as: alellopathyc models; developing of new drugs; ecotoxicity tests. In most cases, lettuce (Lactuca sativa L., Asteraceae) has been used because of its sensitivity, simultaneous and rapid germination, reliability of germination percentage and homogeneity of seeds. The main goal was to evaluate the effects of ultra-high diluted gibberellic acid (GA3) on lettuce seeds germination and seedling growth. Experiment was performed using Petri dishes containing one disk of Whatman nº01 paper watered with 1ml of water. In each Petri dish 10 lettuce seeds(Lactuca sativa L.) cv Regina 500 were placed and 2ml of the different treatment solutions were add: GA33µmol, GA3 3CH (10-6), GA3 12CH (10-24), water 12CH and water (no dilution and succussion). One milliliter solutions were added every 2 days of experiment. The experiment was repeated twice and each one consisted in 5 Petri dishes per treatment (n=100). All seeds were maintained in germination incubator under controlled temperature (25°C) and photoperiod (16L/8D). The tested substances were prepared according to Brazilian Homeopathic Pharmacopoeia (Brazil, 2011). The experiment was blinded all the time. All seeds germinated at same time (2 days) and after 7 days the germination rate was the same in all treatments. Root was affected just by Water 12 CH, in which shown the longest length (4.59 cm) when compared with others treatments. Shoot length was higher where gibberellin was added in concentration upper then Avogrado’s number.

    Abstract: Background: the number of cases of Covid-19 reached more than 11 million people in Brazil, but mostly in Caxias city (Rio de Janeiro) high death rates are very worrysome. In this scenario, homeopathy therapy, along with sanitary and therapeutic measures, should be recommended by health authorities. Aims: to evaluate the use of homeopathic complex (Bryonia alba, China officinalis and Metallum album 30cH), concerning the severity of the illness, during the coronavirus pandemic. Methodology: the complex was administrated at multiple families from Caxias, with (FW) and without (FWO) symptoms similar to COVID-19’s, therefore qualifying the project as a quasi-experimental longitudinal observational cohort study (applied in 51 families or 128 people). With this, the influence of homeopathic intervention in sick (FW) and health (FWO) groups was estimated. All volunteers used three drops of the medicine, once a day, per 60 days. The medicine was prepared accordingly to the Brazilian Homeopathic Pharmacopoeia1. The follow-up was carried out within seven days, which resulted in a total of 8 weeks, since the first interview. The data was organized and statistically analyzed with a 5% significance level, for categorical variables, by Fisher Exact test, in Excel sheet and SPSS 17.0 platform. The ethical committee approved this project under the code: CAAE 31601020.3.0000.5243. Results and Discussion: the preliminary results showed the absence of deaths in both groups (FW and FWO), without any statistically significant difference related to the severity of illness. Most of the families (98.03%) were thrilled to contribute in this particular homeopathic research and reported to have had general health symptoms’ improvements (84.31%). Despite the previous comorbidities presented in 52.9% of the families and considering that Covid-19 infected 0.020% of them, only 0.098% looked for medical assistance and none needed hospitalization. Conclusion: no differences between previous sick and health groups were detected after homeopathy intervention.

    Abstract: Background: The use of sensitive techniques can be useful to increase the understanding of homeopathic systems behavior. Aim: To analyze physicochemical properties of Zincum metallicum and lactose mixtures prepared according to Brazilian Homeopathic Pharmacopeia. Methodology: Zincum metallicum was prepared using lactose as vehicle, from 1 to 3 cH, and from 1 to 6dH. All samples including respective controls (dynamized lactose) were analyzed by Atomic Absorption Spectroscopy (AAS), X-ray Diffraction (XRD), Transmission Electron Microscopy (TEM) with X-Ray Energy Dispersive Spectroscopy (EDX), Scanning Electron Microscopy (SEM), and Differential Scanning Calorimetry (DSC). Results: AAS analysis showed the dynamization process decreases zinc concentration in a proportional manner to the increase of homeopathic potencies. Quantities of 4ppm and 132 ppm of zinc were detected in 6dH and 3cH, respectively. XRD analysis showed crystalline form of lactose was not modified by dynamization. Amorphous lactose spheres were observed by TEM in all samples, with mean size ranging from 800 to 200 nm. EDX obtained in the TEM confirmed zinc presence throughout the amorphous matter and the absence of individualized zinc crystalline particles. These results are in accordance with XRD which showed absence of zinc diffraction peaks, in all lactose samples containing zinc suggesting this metal could be modified from crystalline to amorphous state, and consequently adsorbed into lactose molecules.     Abstract: Background: Leishmaniasis is a serious public health problem especially in developing countries [1]. The therapeutic potential of biotherapics against several microorganism has been described in vitro [2,3] and in vivo studies [4,5,6,7,8,9]. Considering the resistance of leishmaniasis to conventional treatment as well as previous studies with biotherapic, we evaluated the effects of Leishmania infantum 30x (BioLi30x) biotherapy. Aim: evaluate the antileishmanial effects of BioLi30x in in vivo and in vitro models. Methodology: The in vivo experiments were performed using BALB/c mice (n=138), divided into 8 groups: G1-healthy, G2-infected with L. infantum, G3-BioLi30x pre-treated, G4-BioLi30x pre/post-treated, G5-BioLi30x post-treated, G6-H2O30x post-treated, G7-Antimonium crudum 30x post-treated and G8-Glucantime® post-treated. After 49 days of treatment, the animals were submitted to euthanasia (ethical approval ECUA/UFRJ/066/14). Liver and spleen histological changes were evaluated, and serum samples were aliquoted and storage at -20°C for cytokine assays. The in vitro assays were performed using RAW 264.7 macrophages treated with BioLi30x and infected with L. infantum. The morphological aspects were evaluated by scanning electron microscopy (SEM), and the nitric oxide (NO) release was quantified in the supernatant of infected macrophages. Results: The histological analysis from 4 independent experiments showed livers with normal appearance (G1); periportal chronic hepatitis (G2,G4,G5,G8); discreet (G3,G7), moderate (G4,G5,G6), and severe (G2,G8) vacuolar hydropic degeneration; congestion and neutrophilic inflammation (G2,G4,G5,G6,G8), and possible amastigotes within macrophages (G2-G8). Spleens presented healthy appearance only in G1. All treated animals presented histological alterations, with different lesions severity, which involved spleen pulp hyperplasia with moderate disruption (G2,G8), as well as megakaryocytes and macrophages proliferation (G2- G8). SEM analyses showed BioLi30x treatments induced significant protozoan morphology alterations when compared to H2O30x. Besides, a 19% increase in the NO release was detected in RAW supernatants, when compared to H2O30x. Conclusions: BioLi30x and Antimonium crudum 30x modified the infection animal process, involving several cellular mechanisms as well as different histological damage. The in vitro experiments will be repeated in order to confirm these preliminary results.

    Abstract: Introduction: Candidiasis is an opportunist infection, caused by yeast of the genus Candida, which emerges as one of the main causes of systemic infections in hospitalized patients. Candida albicans is the most common causing agent of these infections. According to the Brazilian Homeopathic Pharmacopeia[1] , nosodes are medicines compounded from chemically undefined biological products. Living nosodes are prepared using the etiologic agent of an illness in its infective form, were first developed by Brazilian physician Roberto Costa (RC). Roberto Costa’s research indicated that living nosodes present a higher capability to stimulate the host’s immunological system [2]. Aim: This study aims to evaluate cellular alterations induced in C. albicans yeasts and RAW 264-7 macrophages by Candida albicans RC. Methodology: To prepare Candida albicans RC, one part of C. albicans infective yeast suspension (108 cell/ml) was diluted in 9 parts of sterile distilled water and submitted to 100 mechanical succussions. This process was successively repeated to the potencies of 12x and 30x1. Water 30x was prepared by the same technique, as control. The cell viability of C. albicans previously treated with nosodes in both potencies and respective controls was evaluated using the samples at the concentration of 10% (V/V), in a volume of 1ml, distributed in 1-3 days. The viability of the yeast cells was analyzed by MTT (3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolic) (5mg/ml) assay [3] and by Propidium Iodide (PI) incorporation methods. Additionally, using macrophages RAW 264-7 as a cell model, Nitric Oxide (NO) production and cell viability were also evaluated. For this, the following protocol of cell treatment was employed: on each experimental day, RAW 264-7 cells were treated 4 times (4 stimuli) with RC nosode 30x at the concentration of 10% (V/V). Results: The nosodes (12x and 30x) did not present cytotoxic effects on macrophage cells (n=1), or on C. albicans yeasts (n=2), as detected by MTT and PI methods. Moreover, no statistically significant differences on NO production were detected among the experimental groups (n=6). Conclusion: Preliminary results of in vitro assays indicate that nosodes (12x and 30x) do not alter mitochondrial activity or cell viability of C. albicans. Similarly, treatment by RC nosodes does not seem to alter NO release and mitochondrial activity of RAW macrophages. New experiments are being performed to confirm these preliminary data.

    Abstract: Several in vitro and in vivo studies investigated the biological properties of mistletoe preparations [1-4]. In Europe, where phytochemical constituents of aqueous Viscum album extracts were previously described, the most widespread and common form of mistletoe is the white-berried species (Viscum album) [5] . The aim of the present study was to analyze the biological activity and phytochemical features of different Viscum album ssp mother tinctures collected in 2 different harvesting periods. Leaves, stems and berries of Viscum album samples were subjected to ethanolic extraction (45% v/v) following the homeopathic pharmacopeia [6,7]. The mother tinctures (MT) prepared were: V. album (from 3 host trees Malus domestica, Quercus sp, Ulmus sp), V. album ssp. austriacum (host tree Pinus sylvestris) and V. album ssp. abietis (host tree Abies pectinata). All samples were collected at Höfli farm (Switzerland) in July and August and subjected to chemical analysis by means of high-performance liquid chromatography-mass spectrometry (HPLC-MS/MS) and Thin-Layer Chromatography (TLC). Tumor (Yoshida and Molt4) and non-tumor cells (Ma104) were incubated with MT (1; 0.1; 0.01%) for 4, 24 and 48 hours at 37ºC. Proliferation was indirectly measured using WST-1 (tumor cells) and MTT (non-tumor cells). All cells (1.5 x 104/100 μL) were plated in triplicate in 96-well plates. The cell proliferation inhibition rate was calculated as percent inhibition relative to control cells treated with the MT solvent (45% ethanol v/v). Apoptosis/necrosis was measured using 2 x 105 cells incubated with Annexin V-FITC and 7-AAD at room temperature in the dark, and analyzed in flow cytometer [2] . Antimicrobial activity was assessed for Candida albicans, Cryptococcus neoformans, Escherichia coli and Staphylococcus aureus following the Clinical Laboratory Standards Institute manuals [8-9] and compared with 45% ethanol v/v (MT solvent). The chromatographic assays of MT samples showed phenolic acids and lignans as main chemical classes identified. WST-1 and MTT experiments revealed different antitumor potential of MT. In general, Yoshida cells were more sensitive than Molt4. The maximal cytotoxic effect with 100% of mortality was attained by incubation of Yoshida and Molt-4 cells with 1% MT of V. album ssp. abietis (harvested in July) for 4 hours. Preliminary results with non-tumor cells indicate lower cytotoxicity compared to tumor cells. Annexin V/7ADD showed predominance of late apoptotic/necrotic events after 4 and 24 hours of incubation with all MT. The antimicrobial assay showed that MT of Viscum album ssp. austriacum (host tree Pinus sylvestris) exhibited minimum inhibitory concentration 7.25 mg/mL for the 2 tested bacterial strains. The results of the present study suggest that Viscum album samples prepared according to the homeopathic pharmacopeia exhibit promising biological activities in a dose-dependent manner. The effects of potentiation will be assessed and compared to non-potentiated samples in future experiments. Therefore, the present study contributes with data on the main chemical components in homeopathic preparations of Viscum album, drawing attention to the relevance of extract solvent, seasonal aspects, host tree and harvested parts of plant harvested to the quality of thee mother tinctures.

    Abstract: Plant tissue culture techniques have been used to evaluate the effects of many different substances and/ or conditions in plant growth and development. It provides information of great value about problems related to basic and applied aspects of plant as well as contributed to understanding of factors responsible for growth, metabolism, synthesis of secondary compounds, stress response. Considering all this wide range of applications and as all plant tissue culture techniques are undergone under axenic and controlled conditions (culture medium composition, light and temperature, for instance), it seems to be a value model for Ultra High Diluted (UHD) studies. Lippia alba is a Brazilian plant that tissue cultures protocols and in vitro essential oil production have already been described in scientific literature. None of all scientific papers evaluated the effects of UHD substances on in vitro development or secondary metabolic production. The main goal was to evaluate the use of plant tissue culture to investigate the effects of UHD benzilaminopurine (BA) on Lippia alba shoot culture. Nodal segments obtained from plants growth in vitro was subcultured to Murashigue & Skoog semi-solid medium added with 2ml of these different solutions: BA 3µmol, BA 12CH (10-24), water 12CH and water (no dilution and succussion). Weekly 1 ml of solutions were added to cultures. The experiment was repeated twice and each one consisted in 3 culture vessel with 5 nodal segments per treatment (n=30). All plants were maintained in growth room under controlled temperature (25°C), light and photoperiod (16L/8D). The tested substances were prepared according to the method of stepwise dilution and succussion as describe in Brazilian Homeopathic Pharmacopoeia. The experiment was blinded all the time. After 60d, plantlets were evaluated for number of shoots, shoot length, rooted plants (%), callus development (%) and fresh biomass. Data were submitted to ANOVA following by Duncan’s and t-test. Plants from water 12CH and BA 12CH increased the number of new shoots and promoted the highest shoot length. By adding BA 3µmol the organogenetic response was inhibited since neither shoot nor root were developed. However, it was observed a significant basal callus development. Plant tissue culture could be adapted for UHD studies. More studies are being conducted in way to analyze other experimental conditions and biochemical/phytochemical parameters.
