    Abstract: Proteasome inhibitors have been suggested as potential anticancer agents in many clinical trials. Recent evidence indicates that proteasomal deubiquitinase (DUB) inhibitors, bearing a different mechanism from that of traditional proteasome inhibitors, would be appropriate candidates for new anticancer drug development. In the present study, we describe the deubiquitinase inhibition of 19S regulatory particles (19S RP) by AC17, a 4-arylidene curcumin analog synthesized in our laboratory. Although 4-arylidene curcumin analogs were reported to act as inhibitory κB (IκB) kinase (IKK) inhibitors, AC17 instead induced a rapid and marked accumulation of ubiquitinated proteins without inhibiting proteasome proteolytic activities. In contrast to its parent compound, curcumin, which is a proteasome proteolytic inhibitor, AC17 serves as an irreversible deubiquitinase inhibitor of 19S RP, resulting in inhibition of NF-κB pathway and reactivation of proapoptotic protein p53. In addition, in a murine xenograft model of human lung cancer A549, treatment with AC17 suppresses tumor growth in a manner associated with proteasome inhibition, NF-κB blockage, and p53 reactivation. These results suggest that 4-arylidene curcumin analogs are novel 19S deubiquitinase inhibitors with great potential for anticancer drug development. Mol Cancer Ther; 12(8); 1381–92. ©2013 AACR.

    Abstract: In recent years, oil and gas exploration of the Baikouquan Formation in the northwestern margin of the Junggar Basin has seen major breakthroughs have been made in oil and gas exploration. Sandstone and conglomerate reservoirs are developed in the Baikouquan Formation in this area, but the factors affecting the differences in petro physical properties of these two types of reservoirs are still unclear. In this paper, taking the Baikouquan Formation in the Hongcheguai area on the northwestern margin of the Junggar Basin as an example, a large number of core observations, thin sections, scanning electron microscopy, and petro physical property test data were used to evaluate the petro logical characteristics, reservoir space types, digenesis and pore evolution of the sandstone and conglomerate reservoirs. The results show that the sandstone and conglomerate rocks of the Baikouquan Formation in the study area have a large content of tuffaceous components. The sandstone reservoirs are dominated by primary intergranular pores, and the conglomerate reservoirs are dominated by secondary dissolved pores. In addition, the porosity is negatively correlated with rock permeability for the conglomerate rocks of the Baikouquan Formation. However, the porosity is positively correlated with rock permeability of the sandstone reservoirs. According to the analysis of the digenetic process, the compaction and cementation can lead to a significant decrease in the petro physical properties of the reservoir. While dissolution can improve the petro physical properties of the sandstone reservoirs to a small extent, and can improve that of the conglomerate reservoirs to a greater extent. Moreover, local fractures are important factors affecting the petro physical properties of the conglomerate reservoirs. The existence of abnormal high pressure has a certain effect on the preservation of primary pores. On the whole, the controlling factors of the petro physical properties of the conglomerate reservoirs in the Baikouquan Formation include sedimentation, diagenesis, and tectonic actives, while that of the sandstone reservoirs mainly include sedimentation and diagenesis.

    Abstract: Stroke is a leading cause of death worldwide, but it is unclear whether circulating lipids and lipid-lowering drugs are causally associated with stroke and its subtypes. Methods We used two-sample Mendelian randomization (MR) to examine the effects of blood lipids and lipid-lowering drugs on stroke and its subtypes. Results The inverse variance weighted Mendelian randomization (IVW-MR) revealed the low-density lipoprotein cholesterol (LDL-C) (OR, 1.46; 95% CI, 1.17–1.83; p  = 0.0008) and apolipoprotein B (apoB) (OR, 1.46; 95% CI, 1.21–1.77; p  = 0.0001) was positively correlated with large artery stroke (LAS). However, no causal effect was found in LDL-C and apoB on LAS risk when we conducted mvMR. The IVW-MR also found a suggestive evidence that decreased LDL-C levels mediated by the PCSK9 (proprotein convertase subtilisin-kexin type 9) gene were associated with a reduced risk of any stroke (AS) (OR, 1.31; 95% CI, 1.13–1.52; p  = 0.0003), any ischemic stroke (AIS) (OR, 1.29; 95% CI, 1.10–1.51; p  = 0.001), and LAS (OR, 1.73; 95% CI, 1.15–2.59; p  = 0.008), while NPC1L1 (Niemann-Pick C1-like protein)-mediated LDL-C levels were associated with a higher risk of small vessel stroke (SVS) (OR, 6.10; 95% CI, 2.13–17.43; p  = 0.0008). The SMR revealed that expression of PCSK9 was associated with risk of AS (OR, 1.15; 95% CI, 1.03–1.28; p  = 0.01), AIS (OR, 1.02; 95% CI, 1.14–1.29; p  = 0.03), cardioembolic stroke (CES) (OR, 1.28; 95% CI, 1.01–1.61; p  = 0.04). And, a significant association was found between the expression of NPC1L1 and the risk of SVS (OR, 1.15; 95% CI, 1.00–1.32; p  = 0.04). Conclusion We cautiously find that LDL-C and apoB was positively correlated with LAS. These findings suggest that the reducing LDL-C levels could be an effective prevention strategy for reducing the risk of stroke.

    Abstract: Cerebrovascular disease, among the most prevalent neurological disorders, poses a substantial threat to human health with its elevated mortality and disability rates, placing considerable strain on healthcare systems. Although several studies in recent years have suggested a potential association between digestive system diseases and cerebrovascular diseases, the findings remain inconsistent. Methods Genome-wide association study (GWAS) summary data for 12 digestive diseases and cerebrovascular diseases were used to conduct Mendelian randomization (MR) analysis. In this investigation, we endeavored to elucidate the causal relationship between digestive system diseases and cerebrovascular diseases. Employing a comprehensive approach, including two-sample MR (TSMR), multivariate MR (MVMR), and two-step MR analysis, we leveraged summary statistics data obtained from published GWAS. The primary analysis method employed was inverse variance weighted (IVW), with MR-Egger and weighted median (WM) as secondary methods. Sensitivity analysis included heterogeneity testing, horizontal multivariate testing, MR-PRESSO, and a “leave-one-out” method. Additionally, the F -statistic was utilized to assess the strength of instrumental variables, ensuring robust results. Results In the TSMR analysis, this study found a significant causal relationship between genetically predicted gastroesophageal reflux disease (GERD) and any stroke (AS), any ischemic stroke (AIS), large-artery atherosclerotic stroke (LAS), intracranial aneurysm (IA), and subarachnoid hemorrhage (SAH). In MVMR analysis, this study found that even after adjusting for systolic blood pressure (SBP), body mass index (BMI) and type 2 diabetes (T2D), the causal relationship remains exist. In the two-step MR mediation analysis, it was found that BMI, SBP and T2D play mediating role in the causal relationship between GERD and cerebrovascular diseases. Conclusion This study indicates a clear positive causal relationship between GERD and cerebrovascular diseases, and this causal association remains significant even after adjusting for BMI, SBP and T2D. The mediation MR analysis suggests that BMI, SBP and T2D may mediate the causal relationship between GERD and the risk of cerebrovascular diseases.

    Abstract: Methylation of the adenine base at the nitrogen 6 position (m6A) is the most common post-transcriptional epigenetic modification of RNA, and it plays a very important role in regulating gene expression. To investigate the role of m6A methylation in the expression of non-coding RNA and miRNA, we used a system of adenine base editors (ABEs). Here, we mutated regions up- and downstream of miRNA 675 m6A modification sites in the H19 locus using HEK293T, L02, MHCC97L, MHCC97H, A549, and SGC-7901 cells. Our results showed that a T–A base transversion had occurred in all cell lines. Moreover, mutation of the regions upstream of the miRNA 675 m6A modification site led to reduced expression of H19 and the induction of cell apoptosis in HEK293T cells. To further confirm our results, L02 and MHCC97L cells were detected using ABEs system. The results indicated increased cell apoptosis and reduced expression of miR675 as well as H19. To confirm the relationship between H19 and miR675 expression, overexpression and knockdown studies were performed. The results showed that reduced HI9 expression induced cell apoptosis through miR675. Taken together, these results indicate that m6A modification can regulate the expression of H19 and miR675 which induce cell apoptosis.

    Abstract: Metal‐N‐C is a type of attractive electrocatalyst for efficient CO 2 reduction to CO. Because of the ambiguity in their atomic structures, the active sites and catalytic mechanisms of the catalysts have remained under debate. Here, the effects of N and C hybrid coordination on the activity of Ni‐N‐C catalysts were investigated, combining theoretical and experimental methods. The theoretical calculations revealed that N and C hybrid coordination greatly enhanced the capability of single‐atom Ni active sites to provide electrons to reactant molecules and strengthens the bonding of Ni to N and C in the Ni‐N‐C complexes. During the reaction process, the C and N coordination synergistically optimized the reaction energies in the conversion of CO 2 to CO. A good agreement between theoretical calculations and electrochemical experiments was achieved based on the newly developed Ni‐N‐C electrocatalysts. The activity of hybrid‐coordination NiN 2 C 2 was more than double that of single‐coordination NiN 4 .
