Abstract:
Zusammenfassung: Ketoconazol hemmt die mitogeninduzierbare DNS‐Synthese, Immunglobulinsynthese und Lymphokinproduktion wie auch die antikörpervermittelte zelluläre Zytotoxizität (ADCC) in Kulturen menschlicher mononukleärer Zellen (MNC). Nur die Aktivität natürlicher Killerzellen (NK‐Zellen) – gemessen an Monozyten‐verarmten MNC – blieb unbeeinflußt Ketoconazol selbst ist nicht mitogen, induziert aber innerhalb von 5 Stunden eine Lymphokinproduktion in T‐Zellen. Die optimalen Konzentrationsbereiche lagen je nach Testsystem zwischen 10 −5 bis 10 −11 M. Die Untersuchungen warden an Zellen gesunder Probanden durchgeführt. Nicht alle Probandenzellen erwiesen sich als Ketoconazolsensitiv. Summary: Ketoconazole in vitro inhibits the mitogen‐induced DNA synthesis, immunoglobulin synthesis and lymphokine production of human mononuclear cells (MNC). In contrast to natural killer cell (NK) cytotoxicity, measured in monocyte‐depleted cultures, the antibody‐dependent cellular cytotoxicity (ADCC) of MNC was inhibited as well. Ketoconazole is not mitogenic by itself but is capable to induce T cell lymphokine production within 5 h. Depending on the test system optimal ketoconazole concentrations ranged from 10 −5 bis 10 −11 M. We investigated cells from healthy blood donors. The ketoconazole sensitivity could not be detected in all blood donors.
