    Abstract: Objective: To establish a stable myocardial ischemia animal model, and to find the characteristics of its myocardium proteome and also to seek the biomarkers for myocardial ischemia. Methods: Establishment and assessment of a stable model with myocardial ischemia Chinese experimental mini pigs (25±4 kg) were instrumented with a size-matched Ameroid constrictor on the anterior descending branch under general anesthesia in sterile conditions. The sham group didn’t place the Ameroid constrictor, the other were the same to the model group. After the operations, we dynamically observed the pathophysiology indicators from the two group animals for 12 weeks. Myocardium samples were obtained from the animals with myocardial ischemia and the sham operated animals after operation 12 weeks. Myocardium samples were separated by two-dimensional electrophoresis (2-DE) to identify differentially expressed proteins. A cluster of protein spots differentially expressed were selected and identified with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF/TOF-MS). Results: Compared with the sham group, the differential expressed proteins in the model group 5 up-regulated protein spots and 3 down-regulated protein spots were detected in the 2-DE separation of pooled samples. Albumin, aldose reductase, troponin T, isocitrate dehydrogenase subunit alpha, myosin light chain 3 were significantly highly expressed in the myocardium samples, while NADH-ubiquinone oxidoreductase, ropomyosin alpha-1 chain, Hsp27 were decreased in the myocardium proteome. Conclusion: The detected differential expressed myocardium proteins of animals with myocardial ischemia were associated with the related proteins of energy metabolism obstruction, cytoskeleton proteins, cell damaged and apoptosis - related proteins. Thus, we supposed that energy metabolism obstruction may be the most important changes of physiological and biochemical process in myocardial ischemia, accompanied by cytoskeletal damage, apoptosis and a series of molecular level changes.

    Abstract: Response gene to complement 32 (RGC-32) was considered as an apoptosis-promoting factor. To explore the novel functions of RGC-32, a DNA array analysis was performed in Human Microvascular Endothelial Cells (HMEC) treated with SiRNA, which showed that glucose metabolism was one of most dramatically changed biological process. qPCR analysis also proved that many glucose metabolism related genes were changed dramatically in RGC32 o/e HMEC, one of which was Insulin-like growth factor binding protein 4(IGFBP4). We propose that RGC-32 may be a potential factor involved in the cycle of endothelial glucose metabolism besides glucose induced apoptosis. Then we found that expression of RGC-32 was dramatically increased in mice hearts and adipose tissue after high fat diet treatment, as well as in HMEC after high glucose (30mM) treatment. In high fat diet treated endothelial specific RGC-32 transgenic mice (VE-Cad/RGC-32), decreases in area under the curve (AUC) of glucose tolerance test were observed when compared to their wild type littermates. Upregulated P53 and downregulated IGFBP4 mRNA expression level were also observed in the heart of VE-Cad/RGC-32 mice.Furthermore, we found that P53 was upregulated in RGC-32 o/e HMEC and blocking of p53 was able to decrease the RGC-32-related induction of apoptotic cells. We also observed that RGC-32 can not only decrease IGFBP4 but also reverse the decrease of IGFBP4 in high glucose or insulin condition. Interestingly, after treated the cells with pifithrin-a, a specific P53 inhibitor, decreased IGFBP4 by RGC-32 was rescued. These results demonstrate that RGC-32 may have some kind of connections with glucose metabolism besides its well-known apoptosis-promoting characters and P53 may be a mediator of RGC-32 related glucose metabolism disorder as well as apoptosis.These findings will provide some clues for novel insights into the functions of RGC-32.

    Abstract: Objective . To explore new diagnostic patterns for syndromes to overcome the insufficiency of obtainable macrocharacteristics and specific biomarkers. Methods . Chinese miniswines were subjected to Ameroid constrictor, placed around the proximal left anterior descending branch. On the 4th week, macrocharacteristics, coronary angiography, echocardiography, and hemorheology indices were detected for diagnosis. IL-1, IL-6, IL-8, IL-10, TNF- α , and hsCRP in serum were detected, and Decision Tree was built. Results . According to current official-issued standard, model animals matched the diagnosis of blood stasis syndrome with myocardial ischemia based on findings, including 〉 90% occlusion, attenuated left ventricular segmental motion, dark red or purple tongues, and higher blood viscosity. Significant decrease of IL-10 and increase of TNF- α were found in model animals. However, in the Decision Tree, besides IL-10 and TNF- α , IL-8 helped to increase the accuracy of classification to 86%. Conclusions . The Decision Tree building with TNF- α , IL-10, and IL-8 is helpful for the diagnosis of blood stasis syndrome in myocardial ischemia animals. What is more is that our data set up a new path to the differentiation of syndrome by feature patterns consisting of multiple biomarkers not only for animals but also for patients. We believe that it will contribute to the standardization and international application of syndromes.

    Abstract: Nowadays, it is quite challenging to clarify Chinese medicine's results and its internal biological foundation in the traditional Chinese medicine (TCM) field. The key to break through this problem is to make the right methodological choices. From our study, a pattern consisting of many kinds of characterizations has been found, and every characterization corresponds with its internal biological indicators. If the relationship of every characterization and its biological indicators can be structured, and of each pattern and its biological indicators, it will help us to understand TCM better. Therefore, it is a better method to construct and analyze the "pattern-characterization-biological indicator" network. The aim of this paper is to distinguish two common Chinese medicine patterns, qi deficiency pattern and qi stagnation pattern, and their characterizations in terms of internal biology by using literature-mining methods. Furthermore, the results will be validated by clinical data to examine the methodological reliability. A neuro-endocrine-immune (NEI)-related gene data dictionary and a human phenotype ontology (HPO) characterizations terminology database have been established for these two patterns. Relevant literatures about the characterizations of these two patterns can be found on PubMed. Two different literature-mining software PubMiner and GenCliP, were used on the principle of "pattern-characterization-biological indicator" co-occurrence to find the characteristic NEI gene and the chemical messenger (CM) of Qi deficiency and qi stagnation patterns and to explore the difference in the bioactive substances between the two patterns, such as Hormones, Receptors, Cytokine, Neurotransmitters, etc. Biological networks of the two patterns and their various characterizations were separately constructed by using two literature-mining methods. After integrating and analyzing all kinds of networks, we found that qi deficiency pattern genes based on the NEI network include CD4, CHAT, EPO, GCG, INS, PTH, PRL, REN, SHBG and MAOA; and the key chemical transmitters include IgA, IgM, IgG, IL6, INFα, C3, C4, IL2, T, TSH, T3, T4, NO, EPO, E2, Serotonin, Histamine, ACTH, Hydrocortisone, Insulin, Cytokine, Calcitriol, Aldosterone, Adenosine, Somatostatin, Progesterone Acetylcholine, NE and Dopamine. Qi stagnation pattern genes based on the NEI network include EGF, EGFR, INS, PRL, SHBG, SNAP25, BDNF, COMT, DRD4, CD4 and IL6; and the key chemical transmitters include T3, E2, Prolactin, Serotonin, Steroids, T, ACTH, TSH, NE, ACTH and IL6. By comparing the literature data with clinical data, we found that abnormalities of the endocrine system, especially the thyroid, adrenal gland and gonadal gland, are closely related to the occurrence of coronary heart disease (CHD). The abnormalities in the endocrine system affect the immune system and nervous system, which eventually leads to CHD. The CHD of qi deficiency pattern emphasizes the imbalance of the immune system, while that of qi stagnation patients focuses on the imbalance of the nervous system. To some extent, it is feasible to use literature-mining methods to construct and analyze the "pattern-characterization-biological indicator" network as a new method of finding syndromes for the biological indicators. It provides an advanced, effective and concise method for the objectivity and internalization of traditional medicine. However, the universality and reliability of this method will need to be further validated by other syndrome studies.

    Abstract: Background and Objective: Inflammation was one of the most important pathophysiological processes during myocardial ischemia. However, it is still poorly understood about the possibilities of these pro- or anti-inflammation factors be used as prognostic parameters for subsequent attenuation of heart functions. Methods and Results: Chinese experimental mini swines (25±4 kg) were instrumented with a size-matched Ameroid constrictor on the left anterior descending branch (LAD) under general anesthesia in sterile conditions (N=15). And a sham group (N=6) was included. 4 weeks after thoracotomy, more than 90% occlusion of the LAD was proved by coronary angiography. Early stage of ischemia was found by echocardiography which showed focal ischemia in the anterior wall of left ventricular (from apex to papillary muscle level) and unchanged ejection fraction. Meanwhile, Enzyme-linked immunosorbent assay of serum showed that IL-8 (86.83 ±10.96 vs. 130.3 ±10.94-pg/ml, P =0.0322) and IL-10 (0.9619 ±0.1421 vs. 1.660 ±0.1607-pg/ml, P =0.0111) were dramatically decreased compared to sham group, while no significant changes were found in IL-1 (0.3075 ±0.03558 vs. 0.3045 ±0.057371 -ng/ml, P = 0.9643), IL-6 (307.7 ±27.81 vs. 315.8 ±23.07-pg/ml, P =0.8637), high sensitivity C reactive protein (hsCRP) (12.94 ±1.324 vs. 9.943 ±1.442-ng/ml, P =0.2087) and TNF-a(1.769 ±0.1467 vs. 1.260 ±0.3209-ng/ml, P =0.1121). Furthermore, 8 weeks after thoracotomy, ejection fraction(EF) of myocardial ischemia animals were dramatically attenuated (44.69 ±4.494 vs. 60.60 ±3.420-%, P =0.0428). Subsequent EF (8 weeks postoperation) showed a good linear regression with the IL-10 and hsCRP concentration in the early stage of myocardial ischemia (4 weeks postoperation) (EF= 0.525+ 0.132* IL-10- 0.015* hsCRP, P =0.001) when stepwise regression was performed with all inflammation related factors above as dependent variables. Conclusion: These results demonstrate that lower IL-10 and higher hsCRP in serum may be predictors for the attenuation of heart function caused by myocardial ischemia. Further Clinical trials may help to prove the efficiency of these novel insights of prognosis for ischemia heart diseases.
