    Abstract: Nanozyme is a collection of nanomaterials with enzyme-like activity but higher environmental tolerance and long-term stability than their natural counterparts. Improving the catalytic activity and expanding the category of nanozymes are prerequisites to complement or even supersede enzymes. However, the development of hydrolytic nanozymes is still challenged by diverse hydrolytic substrates and following complicated mechanisms. Here, two strategies are informed by data to screen and predict catalytic active sites of MOF (metal–organic framework) based hydrolytic nanozymes: (1) to increase the intrinsic activity by finely tuned Lewis acidity of the metal clusters; (2) to improve the density of active sites by shortening the length of ligands. Finally, as-obtained Ce-FMA-MOF-based hydrolytic nanozyme is capable of cleaving phosphate bonds, amide bonds, glycosidic bonds, and even their mixture, biofilms. This work provides a rational methodology to design hydrolytic nanozyme, enriches the diversity of nanozymes, and potentially sheds light on future evolution of enzyme engineering.

    Abstract: Light exposure can profoundly affect neurological functions and behaviors. Here, we show that short-term exposure to moderate (400 lux) white light during Y-maze test promoted spatial memory retrieval and induced only mild anxiety in mice. This beneficial effect involves the activation of a circuit including neurons in the central amygdala (CeA), locus coeruleus (LC), and dentate gyrus (DG). Specifically, moderate light activated corticotropin-releasing hormone (CRH) positive (+) CeA neurons and induced the release of corticotropin-releasing factor (CRF) from their axon terminals ending in the LC. CRF then activated tyrosine hydroxylase-expressing LC neurons, which send projections to DG and release norepinephrine (NE). NE activated β-adrenergic receptors on CaMKIIα-expressing DG neurons, ultimately promoting spatial memory retrieval. Our study thus demonstrated a specific light scheme that can promote spatial memory without excessive stress, and unraveled the underlying CeA-LC-DG circuit and associated neurochemical mechanisms.

    Abstract: The Neo-REGATTA study evaluated the effectiveness and safety of Docetaxel, oxaliplatin, and S-1 (DOS regimen) followed by radical resection vs. chemotherapy in advanced gastric adenocarcinoma patients with single non-curable factor. Methods This cohort study prospectively enrolled advanced gastric adenocarcinoma patients with single non-curable factor between November 2017 and June 2021. Patients without progression after four cycles of DOS were divided into resection group and chemotherapy group. The outcomes included overall survival (OS), progression-free survival (PFS) and safety. Effectiveness analysis was also performed by propensity score matching (PSM). Results A total of 73 patients were enrolled and 13 patients were withdrawn due to disease progression after 4 cycles of DOS. Afterwards, 35 and 25 participants were in the resection and chemotherapy groups, respectively. After a median follow-up time of 30.0 months, the median PFS and OS were 9.0 months, and 18.0 months for the chemotherapy group, but not reached in the resection group. After PSM, 19 matched participants were in each group, and the median PFS and OS were longer in resection group than that in chemotherapy group. The most common grade 3 or 4 adverse events both in the resection group and chemotherapy groups were neutropenia (5.7%, 8.0%) and leukopenia (5.7%, 8.0%). Conclusions Radical resection might provide survival benefit compared with continuous chemotherapy alone in advanced gastric adenocarcinoma patients who had a disease control after DOS, with a good safety profile. Trial registration The study protocol was registered on ClinicalTrial.gov (NCT03001726, 23/12/2016).

    Abstract: We establish some framework so that the generalized Conley index can be easily used to study the multiple solution problem of semilinear elliptic boundary value problems. Both the parabolic flow and the gradient flow are used. Some examples are given to compare our approach here with other well-known methods. Our abstract results with parabolic flows may have applications to parabolic problems as well.

    Abstract: The Beijing 2008 Olympics Research and Development Project (B08RDP), initiated in 2004 under the World Meteorological Organization (WMO) World Weather Research Programme (WWRP), undertook the research and development of mesoscale ensemble prediction systems (MEPSs) and their application to weather forecast support during the Beijing Olympic Games. Six MEPSs from six countries, representing the state-of-the-art regional EPSs with near-real-time capabilities and emphasizing on the 6–36-h forecast lead times, participated in the project. The background, objectives, and implementation of B08RDP, as well as the six MEPSs, are reviewed. The accomplishments are summarized, which include 1) providing value-added service to the Olympic Games, 2) advancing MEPS-related research, 3) accelerating the transition from research to operations, and 4) training forecasters in utilizing forecast uncertainty products. The B08RDP has fulfilled its research (MEPS development) and demonstration (value-added service) purposes. The research conducted covers the areas of verification, examining the value of MEPS relative to other numerical weather prediction (NWP) systems, combining multimodel or multicenter ensembles, bias correction, ensemble perturbations [initial condition (IC), lateral boundary condition (LBC), land surface IC, and model physics], downscaling, forecast applications, data assimilation, and storm-scale ensemble modeling. Seven scientific issues important to MEPS have been identified. It is recognized that the daily use of forecast uncertainty information by forecasters remains a challenge. Development of forecaster-friendly products and training activities should be a long-term effort and needs to be continuously enhanced. The B08RDP dataset is also a valuable asset to the research community. The experience gained in international collaboration, organization, and implementation of a multination regional EPS for a common goal and to address common scientific issues can be shared by the ongoing projects The Observing System Research and Predictability Experiment (THORPEX) Interactive Grand Global Ensemble—Limited Area Models (TIGGE-LAM) and North American Ensemble Forecast System—Limited Area Models (NAEFS-LAM).

    Abstract: Sea ice is crucial in many natural processes and human activities. Understanding the dynamical couplings between the inception, growth and equilibrium of sea ice and the rich fluid mechanical processes occurring at its interface and interior is of relevance in many domains, ranging from geophysics to marine engineering. Here we investigate experimentally the complete freezing process of water with dissolved salt in a standard natural convection system, i.e. the prototypical Rayleigh–Bénard cell. Due to the presence of a mushy phase, the studied system is considerably more complex than the freezing of fresh water in the same conditions (Wang et al. , Proc. Natl Acad. Sci. USA , vol. 118, issue 10, 2021, e2012870118). We measure the ice thickness and porosity at the dynamical equilibrium state for different initial salinities of the solution and temperature gaps across the cell. These observables are non-trivially related to the controlling parameters of the system as they depend on the heat transport mode across the cell. We identify in the experiments five out of the six possible modes of heat transport. We highlight the occurrence of brine convection through the mushy ice and of penetrative convection in stably stratified liquid underlying the ice. A one-dimensional multi-layer heat flux model built on the known scaling relations of global heat transport in natural convection systems in liquids and porous media is proposed. Given the measured porosity of the ice, it allows us to predict the corresponding ice thickness, in a unified framework.

    Abstract: The lipophilic polycyclic aromatic hydrocarbon (PAH) phenanthrene is relatively abundant in polluted air and water and can access and accumulate in human tissue. Phenanthrene has been reported to interact with cardiac ion channels in several fish species. This study was undertaken to investigate the ability of phenanthrene to interact with hERG ( human Ether-à-go-go-Related Gene ) encoded Kv11.1 K + channels, which play a central role in human ventricular repolarization. Pharmacological inhibition of hERG can be proarrhythmic. Whole-cell patch clamp recordings of hERG current (I hERG ) were made from HEK293 cells expressing wild-type (WT) and mutant hERG channels. WT I hERG1a was inhibited by phenanthrene with an IC 50 of 17.6 ± 1.7 µM, whilst I hERG1a/1b exhibited an IC 50 of 1.8 ± 0.3 µM. WT I hERG block showed marked voltage and time dependence, indicative of dependence of inhibition on channel gating. The inhibitory effect of phenanthrene was markedly impaired by the attenuated inactivation N588K mutation. Remarkably, mutations of S6 domain aromatic amino acids (Y652, F656) in the canonical drug binding site did not impair the inhibitory action of phenanthrene; the Y652A mutation augmented I hERG block. In contrast, the F557L (S5) and M651A (S6) mutations impaired the ability of phenanthrene to inhibit I hERG , as did the S624A mutation below the selectivity filter region. Computational docking using a cryo-EM derived hERG structure supported the mutagenesis data. Thus, phenanthrene acts as an inhibitor of the hERG K + channel by directly interacting with the channel, binding to a distinct site in the channel pore domain.
