    Kurzfassung: Cancer‐associated fibroblasts (CAFs) reconstitute cancer stemness. This study aims to investigate whether the loss of CAF‐derived exosomal miR‐34c‐5p contributes to the maintenance of stem‐like properties of laryngeal squamous cell carcinoma (LSCC). Methods Exosomes from primarily cultured CAFs and paired normal fibroblasts (NFs) were collected and identified. The differential expression of exosomal miR‐34c‐5p between CAFs and NFs was detected by next‐generation sequencing. In vitro and in vivo assays were performed to examine the effects of miR‐34c‐5p on the maintenance of stem‐like properties. Results MiR‐34c‐5p expression is significantly reduced in CAF‐derived exosomes. In vitro and in vivo assays revealed that exosomal miR‐34c‐5p can regulate the stem‐like properties of LSCC cells, such as proliferation, invasion, sphere and plate colony formation, chemoresistance, tumorigenicity in nude mice, as well as the expression of cancer stem cell genes. Conclusions Loss of miR‐34c‐5p in CAF‐derived exosomes contributes to the maintenance of stem‐like phenotypes of LSCC.

    Kurzfassung: Reliable quantitative information regarding sediment sources is essential for target mitigation, particularly in settings with a large number of loose provenances caused by earth disasters. The lakes in the Jiuzhaigou World Natural Heritage Site (WNHS) are facing serious environmental problems of silting and swamping, which threaten the sustainability of the area, especially after the earthquake on 8 August 2017 (the “8.8 earthquake”). Therefore, a field investigation was conducted after the “8.8 earthquake” (June 2020), and the Arrow Bamboo and Rhino Lakes, which were affected by the earthquakes to different degrees, were selected as the research objects. Based on the data of 27 environmental indicators from 31 surface sediment and soil samples in and around the lakes, the spatial distribution characteristics of the lake sediment sources were quantified using composite fingerprint recognition technology. Furthermore, a high protection standard of a WHNS and a process treatment scheme for reducing the siltation of the Jiuzhaigou lakes were proposed. The results showed that the contribution ratio of loose matter sources entering the lake on the road-side of the Arrow Bamboo and Rhino Lakes (16.5% and 21.8%, respectively) was lower than that on the forest-side (83.5% and 78.2%, respectively), indicating that physical barriers such as roads can effectively reduce the sediment input, while the lake forest side contributes a large number of loose matter sources, which has not attracted attention in the past and requires protection. High protection standards for the Jiuzhaigou WHNS are suggested. Accordingly, the entire control scheme of Jiuzhaigou lake sediment reduction including “monitoring–control–interception–buffer–cleaning” is provided. Source erosion monitoring is the first step in blocking the sediment source. Vegetation restoration and surface coverage should be conducted in areas where water and soil losses have occurred. Necessary engineering measures should be implemented to intercept loose material sources at points where geological disasters occur frequently. A buffer zone should be established between the lake and the mountain to intercept the sediment. Sediment caused by geological disasters with low interference must also be cleaned from the lake. The level of nutrients in the lake must be controlled by the regular cleaning of plant debris from the lake and lakeside.

    Kurzfassung: Due to the unique geographical characteristics of cold alpine and high-altitude regions, glaciers, permafrost, ground ice, rock glaciers, and other periglacial geomorphology have developed with fragile habitats, and these areas are often the birthplaces of many river basins and natural hazards. With global warming and the extensive cryogenesis and physical weathering, the thermal state of permafrost and the mass balance of glaciers have been changed, and thus it can be deduced that the surface deformation is of great concern. To obtain ground subsidence or uplift over a large area to understand local surface changes, the small baseline subset interferometric synthetic aperture radar (SBAS-InSAR) technique was applied to process 89-scene of Sentinel-1A images ranging from December 25, 2017 to January 2, 2021 to obtain surface deformation for these 3 years for the eastern Tianshan Mountains, China. The surface deformation characteristics of the area were analyzed to provide a basic dataset for environmental protection policies and mitigation or reduction of natural hazards in this region, and to verify the applicability of SBAS-InSAR technology in alpine and high-altitude areas. The results show that the SBAS-InSAR technique processing with sentinel-1A dataset cannot be effectively used to acquire ground deformation in areas covered by trees, scrub/shrub, glaciers, snow, and ground ice, where the decohered phenomenon is serious. In other regions, SBAS-InSAR can effectively measure surface subsidence or uplift. Surface deformation is significant throughout the study area, with rates ranging from −70.7 to 50.8 mm/a and with an average rate of 1.1 mm/a. There are obvious regions of uplift in the northwest, northeast, and central sections of the study area, with uplift greater than 155.73 mm in 3 years, and three obvious regions of subsidence in the northeast and west sections of the study area, with subsidence of at least −125.20 mm in 3 years. The remaining areas of deformation are scattered, with smaller amounts of settlement and uplift and with an isolated and sporadic distribution. Areas with elevations of 3,150 to 4,275 m.a.s.l., slopes of 15°–50°, and southwest, west, and northwest aspects are geologic disaster-prone regions and should receive more attention and more field monitoring. The results of this study have important implications for local environmental protection and hazard prevention.

    Kurzfassung: Background: Peste des petits ruminants virus (PPRV) is the causative agent of PPR, which can cause an acute, highly contagious and fatal disease of sheep and goats, resulting in significant economic losses for commercial animal husbandry due to its high mortality and morbidity. As professional antigen-presenting cells, dendritic cells (DCs) play a unique role in innate immunity. This study aimed to gain a deeper understanding of the transcriptional response of bone marrow-derived dendritic cells (BMDCs) stimulated with PPRV. Results: Transcriptional profiling was performed using RNA sequencing. Herein, we reported that compared to untreatedBMDCs, 4492 differentially expressed genes (DEGs) were identified following PPRV stimulation, out of these DEGs 2311 were upregulated and 2181 were downregulated, respectively. A total of three gene ontology (GO) term clusters of biological process, cell component and molecular function were significantly enriched in 963 GO terms in the PPRV-stimulated BMDCs. These GO clusters were related to inflammatory response, cell division and vacuole, anchoring junction, positive regulation of cellular component and nucleoside binding. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways of DEGs were enriched in a chemokine signaling pathway, protein processing in endoplasmic reticulum, cell cycle and mTOR signaling pathway. Additionally, identified DEGs of BMDCs were further validated by qRT-PCR and the results were in accordance with the change of the genes. This study suggested the effects of PPRV stimulation on the maturation and function of BMDCs. Conclusion: We found that the dramatic BMDCs transcriptome changes triggered were predominantly related to an inflammatory response and chemokine signaling pathway.

    Kurzfassung: Renal hypoxia and its associated oxidative stress is a common pathway for the development of kidney diseases, and using dietary antioxidants such as flavan-3-ols to prevent kidney failure has received much attention. This study investigates the molecular mechanism by which flavan-3-ols prevent hypoxia-induced cell death in renal tubular epithelial cells. Human kidney proximal tubular cells (HKC-8) were exposed to hypoxia (1% O2) in the presence of flavan-3-ols (catechin, epicatechin, procyanidin B1, and procyanidin B2). Cell death was examined using flow cytometric analysis. Gene expression was determined using a PCR array and Western blotting, and its network and functions were investigated using STRING databases. Here, we show that the cytoprotective activity of catechin was the highest among these flavan-3-ols against hypoxia-induced cell death in cultured HKC-8 cells. Exposure of HKC-8 cells to hypoxia induced oxidative stress leading to up-regulation of DUOX2, NOX4, CYBB and PTGS2 and down-regulation of TXNRD1 and HSP90AA1. Treatment with catechin or other flavan-3-ols prevented the down-regulation of TXNRD1 expression in hypoxic HKC-8 cells. Overexpression of TXNRD1 prevented hypoxia-induced cell death, and inactivation of TXNRD1 with TRi-1, a specific TXNRD1 inhibitor, reduced the catechin cytoprotection against hypoxia-induced HKC-8 cell death. In conclusion, flavan-3-ols prevent hypoxia-induced cell death in human proximal tubular epithelial cells, which might be mediated by their maintenance of TXNRD1 expression, suggesting that enhancing TXNRD1 expression or activity may become a novel therapeutic strategy to prevent hypoxia-induced kidney damage.

    Kurzfassung: The fragile habitat of alpine mining areas can be greatly affected by surface disturbances caused by mining activities, particularly open-pit mining activities, which greatly affect the periglacial environment. SBAS-InSAR technology enables the processing of SAR images to obtain highly accurate surface deformation information. This paper applied SBAS-InSAR technology to obtain three years of surface subsidence information based on the 89-scene Sentinel-1A SLC products, covering a mining area (tailings and active areas) in the Tianshan Mountains and its surroundings from 25th December 2017 to 2nd January 2021. The data were adopted to analyze the characteristics of deformation in the study region and the mining areas, and the subsidence accumulation was compared with field GNSS observation results to verify its accuracy. The results showed that the study area settled significantly, with a maximum settlement rate of −44.80 mm/a and a maximum uplift rate of 28.04 mm/a. The maximum settlement and accumulation of the whole study area over the three-year period were −129.39 mm and 60.49 mm, respectively. The mining area had a settlement value of over 80 mm over the three years. Significantly, the settlement rates of the tailings and active areas were −35 mm/a and −40 mm/a, respectively. Debris accumulation in the eastern portion of the tailings and active areas near the mountain was serious, with accumulation rates of 25 mm/a and 20 mm/a, respectively, and both had accumulation amounts of around 70 mm. For mine tailing pile areas with river flows, the pile locations and environmental restoration should be appropriately adjusted at a later stage. For gravel pile areas, regular cleaning should be carried out, especially around the mining site and at the tunnel entrances and exits, and long-term deformation monitoring of these areas should be carried out to ensure safe operation of the mining site. The SBAS-InSAR measurements were able to yield deformations with high accuracies over a wide area and cost less human and financial resources than the GNSS measurement method. Furthermore, the measurement results were more macroscopic, with great application value for surface subsidence monitoring in alpine areas.

    Kurzfassung: Retinoid X receptor alpha (RXRA) is a well-characterized factor that regulates lipid metabolism; however, the regulatory mechanism in muscle cells of poultry is still unknown. The overexpression and the knockdown of RXRA in myoblasts (CS2 cells), RT-PCR, and western blotting were used to detect the expression levels of genes and proteins related to PPAR-signaling pathways. Intracellular triglycerides (TGs), cholesterol (CHOL), and nonesterified free fatty acids (NEFAs) were detected by the Elisa kit. Fat droplets were stained with Oil Red O. The double-fluorescein reporter gene and chromatin immunoprecipitation (CHIP) were used to verify the relationship between RXRA and candidate target genes. The RXRA gene was highly expressed in duck breast muscle, and its mRNA and its protein were reduced during the differentiation of CS2 cells. The CS2 cells, with the overexpression of RXRA, showed reduced content in TGs, CHOL, NEFAs, and lipid droplets and upregulated the mRNA expression of CD36, ACSL1, and PPARG genes and the protein expression of CD36 and PPARG. The knockdown of RXRA expression in CS2 cells enhanced the content of TGs, CHOL, NEFAs, and lipid droplets and downregulated the mRNA and protein expression of CD36, ACLS1, ELOVL6, and PPARG. The overexpression of the RXRA gene, the activity of the double-luciferase reporter gene of the wild-type CD36 promoter was higher than that of the mutant type. RXRA bound to −860/−852 nt, −688/−680 nt, and −165/−157 nt at the promoter region of CD36. Moreover, the overexpression of CD36 in CS2 cells could suppress the content of TGs, CHOL, NEFAs, and lipid droplets, while the knockdown expression of CD36 increased the content of TGs, CHOL, NEFAs, and lipid droplets. In this study, the transcription factor, RXRA, inhibited the accumulation of TGs, CHOL, NEFAs, and fat droplets in CS2 cells by promoting CD36 expression.
