    Kurzfassung: Red blood cell fragments in blood smears from dogs are described morphologically and pathogenetically. Categories include microangiopathic fragmentation, spherocytic fragmentation, Heinz body fragmentation, metabolic fragmentation associated with systemic disease, and artifactual fragmentation. Microangiopathic fragmentation is associated with direct physical damage to normal circulating red blood cells as they pass through abnormal capillary beds. Spherocytic fragmentation is a common feature of immune-mediated hemolytic anemia and results from the removal of portions of antibody-coated erythrocyte plasma membranes by phagocytes of the reticuloendothelial system. Heinz body fragmentation occurs when rigid particles of oxidized hemoglobin are torn from affected red cells as they circulate through the spleen. Metabolic fragmentation is an ill-defined syndrome most commonly associated with cholesterol loading of red cell membranes caused by lipid metabolism abnormalities. Resulting spiculated red cells are more susceptible to traumatic disruption. All the types of red cell fragmentation described in dogs have been observed and documented in man.

    Kurzfassung: Fiberoptic bronchoscopy was used to obtain cytologic specimens from all lung lobes of 9 normal Beagle dogs. Three specimen collection techniques (bronchial lavage, bronchial brushing and bronchial pinch biopsy imprints) and two staining procedures (Wright-Giemsa and Papanicolaou) were used and evaluated. Bronchial lavage was the most satisfactory technique for collection of samples from the deep lung and bronchial brushings were preferred for potential bronchial tree mural lesions. Wright-Giemsa was the stain of choice because mast cells could not be identified and eosinophilic leukocytes could be identified only with difficulty in Papanicolaou stained specimens. Total and differential cell counts were determined on all bronchial lavages from all lung lobes in order to establish baseline reference values. Total nucleated cell counts ranged from 260-1200/μl. There were no significant differences among mean total nucleated cell counts for the different lung lobes. Mean total nucleated cell counts were between 420 and 630 cellsμl. Approximately 95% of all nucleated cells in normal lavages were undifferentiated alveolar macrophages. Most of the other cells seen were neutrophils, eosinophils, possible globule leukocytes and mast cells. Ciliated and nonciliated epithelial cells comprised less than 1% of the total nucleated cell population.

    Kurzfassung: Monoclonal antibody (MAb) B72.3, which binds to a human tumor-associated glycoprotein termed TAG-72, was applied to a wide range of epithelial and nonepithelial neoplasms from dogs. Immunoreactivity was detected by the use of an avidin-biotin complex (ABC) immunoperoxidase method. A variety of epithelial neoplasms, but none of the nonepithelial neoplasms, were positive (≥ 5% staining) for MAb B72.3. MAb B72.3 stained 100% (4/4) of gastric, 100% (6/6) of intestinal, 50% (2/4) of pancreatic, and 80% (4/5) of rectal adenocarcinomas but only 20% (1/5) of squamous cell carcinomas and 20% (1/5) of complex tubular mammary gland adenocarcinomas. None of the hepatocellular carcinomas and perianal and sebaceous gland adenocarcinomas stained. Most types of benign epithelial neoplasms did not stain, except for 75% (6/8) of rectal adenomas and 25% (1/4) of squamous cell papillomas. Normal gastrointestinal mucosa adjacent to and entrapped in neoplasms did stain with MAb B72.3. None of the benign and malignant nonepithelial neoplasms of mesenchymal, neuroendocrine, or lymphohematopoietic tissue origin stained. The results of this study demonstrate that MAb B72.3 has selective immunoreactivity for adenocarcinomas, especially those arising from the digestive tract; however, limited immunoreactivity was observed for other types of carcinomas and benign epithelial neoplasms and for normal gastrointestinal mucosa in the dog.

    Kurzfassung: Flow cytometric analysis of DNA ploidy was performed on 51 formalin-fixed, paraffin-embedded samples of canine transitional cell carcinoma of the urinary bladder. The DNA ploidy data obtained were compared to several clinicopathologic features. In addition, the DNA ploidy of 15 hyperplastic/inflamed and 8 normal canine urinary bladders was measured. Forty-three of the 51 neoplastic samples had interpretable DNA histograms. DNA aneuploidy was found in 34/43 (79%) of the transitional cell carcinoma samples. Of the 34 aneuploid neoplasms, 16 (47%) were hyperdiploid, 17 (50%) were tetraploid, and 1 (3%) was hypertetra-ploid. No significant correlation was found between the presence of DNA aneuploidy and the growth pattern. histologic grade, clinical stage, or individual morphologic features of this neoplasm. Additionally, the DNA ploidy was not related to the sex, age, or survival time of dogs with transitional cell carcinoma. All of the normal and all but one of the hyperplastic/inflamed urinary bladders were diploid. The results from this study demonstrated that DNA ploidy can be measured from paraffin-embedded canine samples by flow cytometry, a majority of the canine transitional cell carcinomas were aneuploid, and a significant correlation did not exist between the DNA ploidy and specific clinicopathologic features of this neoplasm.

    Kurzfassung: Tumor-associated glycoprotein 72 (TAG-72) is a large, high molecular weight, mucinlike antigen that is expressed in a wide variety of human carcinomas. Three different TAG-72 monoclonal antibodies (MAbs), designated B72.3, CC49, and CC83, were applied to the following archived samples from the dog: 1) 51 transitional cell carcinomas of the urinary bladder, 2) 15 hyperplastic/inflamed urinary bladders, and 3) eight normal urinary bladders. Immunoreactivity was detected with an avidin-biotin complex immunoperoxidase method. Fifty-three percent (27/51) of transitional cell carcinomas were positive (≥ 5% staining) for MAb B72.3. MAb B72.3 staining of these transitional cell carcinomas did not statistically correlate with any of the examined features of malignancy, including histologic grade, clinical stage, DNA ploidy, or presence of vascular/lymphatic invasion. In regard to the staining of transitional cell carcinoma by the other two TAG-72 antibodies, 53% (27/ 51) of the samples were positive for MAb CC83 and 63% (32/51) were positive for MAb CC49. The finding that similar populations of neoplastic urothelial cells in serial sections from the same neoplasm stained with all three TAG-72 antibodies supports the hypothesis that an antigen similar to TAG-72 was expressed in canine transitional cell carcinoma. None of the normal urinary bladders nor any of the hyperplastic/inflamed urinary bladders stained with any of the three TAG-72 antibodies tested. The results of these studies demonstrated that the staining of canine transitional cell carcinoma with all three TAG-72 antibodies was specific for neoplastic urothelial cells and that an antigen similar to TAG-72 was expressed.

    Kurzfassung: Sighthounds, including deerhounds, have unique physiological traits that result in laboratory test results that may lie outside reference intervals for the general dog population. Although reference intervals for most analytes are thought to be similar among sighthounds, breed‐specific reference intervals are available mainly for greyhounds. The aim of this study was to establish reference intervals for haematology and serum biochemical profiles in deerhounds. Methods Venous blood samples were collected from healthy deerhounds. Haematological and biochemical analytes were examined and reference intervals were established using the 5th and 95th percentiles. Results The reference intervals obtained from 96 dogs for platelets, reticulocytes, total thyroxine, chloride, gamma glutamyl transferase, bilirubin and glucose were lower than the general dog population. Reference intervals for mean cell volume, potassium, urea, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase and cholesterol were higher than the general dog population. Reference intervals for eosinophils and globulin were wider than that of the general population. Clinical Significance These results confirm that differences in haematological and biochemical values exist in the deerhound. Some appear to be shared by all sighthounds but others may be unique to this breed.

    Kurzfassung: A retrospective study of red blood cell parameters in 53 dogs with experimental radiation-induced hemangiosarcoma showed 24 had anemia. Morphologic alterations in red blood cells in peripheral blood films from anemic dogs included signs of regeneration (anisocytosis and polychromasia), hypochromasia, red cell fragmentation and acanthocytosis. The degree and type of red cell changes varied from dog to dog and generally correlated with the principal site of tumor involvement. Blood from dogs with tumors principally involving liver had red cell regeneration, fragmentation and acanthocytosis. Blood from dogs with tumors primarily involving the heart had only red cell fragmentation. Blood films from dogs with skeletal and pulmonary hemangiosarcomas were similar to blood films from dogs with hepatic hemangiosarcoma except that red cell alterations generally were less severe. Scanning and transmission electron micrographic evaluation of neoplastic tissue showed large amounts of fibrin within neoplastic vascular sinuses and disruption and distortion of red blood cells traversing these abnormal vascular beds. The red blood cell fragmentation syndrome associated with radiation-induced hemangiosarcomas therefore was considered to be a microangiopathic hemolytic anemia of localized origin.
