    Abstract: Hydrogen sulfide (H 2 S) has antihypertension and anti-inflammatory effects, and its endogenous-generation key enzyme cystathionine γ lyase (CSE) is expressed in CD4 + T cells. However, the role of CD4 + T-cell endogenous CSE/H 2 S in the development of hypertension is unclear. Methods: Peripheral blood lymphocytes were isolated from hypertensive patients or spontaneously hypertensive rats, then H 2 S production and expression of its generation enzymes, cystathionine β synthase and CSE, were measured to determine the major H 2 S generation system changes in hypertension. Mice with CSE-specific knockout in T cells (conditional knockout, by CD4 cre mice hybridization) and CD4 null mice were generated for investigating the pathophysiological relevance of the CSE/H 2 S system. Results: In lymphocytes, H 2 S from CSE, but not cystathionine β synthase, responded to blood pressure changes, supported by lymphocyte CSE protein changes and a negative correlation between H 2 S production with systolic blood pressure and diastolic blood pressure, but positive correlation with the serum level of interleukin 10 (an anti-inflammatory cytokine). Deletion of CSE in T cells elevated BP (5–8 mm Hg) under the physiological condition and exacerbated angiotensin II–induced hypertension. In keeping with hypertension, mesenteric artery dilation impaired association with arterial inflammation, an effect attributed to reduced immunoinhibitory T regulatory cell (Treg) numbers in the blood and kidney, thus causing excess CD4 + and CD8 + T cell infiltration in perivascular adipose tissues and kidney. CSE knockout CD4 + T cell transfer into CD4 null mice, also showed the similar phenotypes’ confirming the role of endogenous CSE/H 2 S action. Adoptive transfer of Tregs (to conditional knockout mice) reversed hypertension, vascular relaxation impairment, and immunocyte infiltration, which confirmed that conditional knockout–induced hypertension was attributable, in part, to the reduced Treg numbers. Mechanistically, endogenous CSE/H 2 S promoted Treg differentiation and proliferation by activating AMP-activated protein kinase. In part, it depended on activation of its upstream kinase, liver kinase B1, by sulfhydration to facilitate its substrate binding and phosphorylation. Conclusion: The constitutive sulfhydration of liver kinase B1 by CSE-derived H 2 S activates its target kinase, AMP-activated protein kinase, and promotes Treg differentiation and proliferation, which attenuates the vascular and renal immune-inflammation, thereby preventing hypertension.

    Abstract: We aimed to determine the risk conferred by metabolic syndrome ( METS ) and diabetes mellitus ( DM ) to recurrent stroke in patients with minor ischemic stroke or transient ischemic attack from the CHANCE (Clopidogrel in High‐risk patients with Acute Non‐disabling Cerebrovascular Events) trial. Methods and Results In total, 3044 patients were included. Patients were stratified into 4 groups: neither, METS only, DM only, or both. METS was defined using the Chinese Diabetes Society ( CDS ) and International Diabetes Foundation ( IDF ) definitions. The primary outcome was new stroke (including ischemic and hemorrhagic) at 90 days. A multivariable Cox regression model was used to assess the relationship of METS and DM status to the risk of recurrent stroke adjusted for potential covariates. Using the CDS criteria of METS , 53.2%, 17.2%, 19.8%, and 9.8% of patients were diagnosed as neither, METS only, DM only, and both, respectively. After 90 days of follow‐up, there were 299 new strokes (293 ischemic, 6 hemorrhagic). Patients with DM only (16.1% versus 6.8%; adjusted hazard ratio 2.50, 95% CI 1.89–3.39) and both (17.1% versus 6.8%; adjusted hazard ratio 2.76, 95% CI 1.98–3.86) had significantly increased rates of recurrent stroke. No interaction effect of antiplatelet therapy by different METS or DM status for the risk of recurrent stroke ( P =0.82 for interaction in the fully adjusted model of CDS ) was observed. Using the METS ( IDF ) criteria demonstrated similar results. Conclusions Concurrent METS and DM was associated with an increased risk of recurrent stroke in patients with minor stroke and transient ischemic attack.

    Abstract: Objective: Myocardial ischemia/reperfusion (I/R) injury is one of the causes of most cardiomyocyte injuries and deaths. Berberine (BBR) has been suggested a potential to exert protective effects against myocardial I/R injury. This systematic review aims to determine the intrinsic mechanisms of BBR's protective effects in myocardial I/R injury. Methods: Seven databases were searched for studies performed from inception to July 2020. Methodological quality was assessed by SYRCLE's-RoB tool. Results: Ten studies including a total of 270 animals were included in this study. The methodology quality scores of the included studies ranged from 5 to 7 points. The meta-analysis we conducted demonstrated that BBR significantly reduced myocardial infarct size and the incidence of ventricular arrhythmia, compared to control groups ( P & lt; 0.00001). Cardiac function of animals in the BBR treatment group was also markedly increased ( P & lt; 0.00001). The index of myocardial apoptosis and the levels of biomarkers of myocardial infarction (LDH and CK) were also decreased in the BBR treatment groups compared to the control groups ( P & lt; 0.00001). Conclusions: The pre-clinical evidence, according to our study, showed that BBR is a promising therapeutic agent for myocardial I/R injury. However, this conclusion should be further investigated in clinical studies.

    Abstract: The role of dual-antiplatelet therapy with clopidogrel plus aspirin has been demonstrated to substantially decrease the risk of recurrent stroke among patients with minor stroke and transient ischemic attack. We aimed to determine whether the efficacy of clopidogrel-aspirin therapy among patients with minor stroke / transient ischemic attack was influenced by the stratification of CYP2C19 genotype and body mass index (BMI). Methods— CYP2C19 loss-of-function allele (LoFA) carriers were defined as patients with either LoFA of *2 or *3. Low/normal weight and overweight/obesity was defined as BMI 〈 25 and ≥25 kg/m 2 , respectively. Primary outcome was defined as stroke recurrence at 3 months. Results— In a total of 2933 patients, there were 1726 (58.8%) LoFA carriers and 1275 (43.5%) patients with overweight/obesity (BMI ≥25 kg/m 2 ). Stratified analyses by LoFA carrying status and BMI, hazard ratios (hazard ratios 95% CIs) of the clopidogrel-aspirin therapy for stroke recurrence were 0.90 (0.60–1.36), 0.87 (0.56–1.35), 0.65 (0.39–1.09), and 0.40 (0.22–0.71) among subgroups of LoFA carriers with overweight/obesity, LoFA carriers with low/normal weight, LoFA noncarriers with overweight/obesity, and LoFA noncarriers with low/normal weight, respectively, with P =0.049 for interaction. Conclusions— Efficacy of clopidogrel-aspirin therapy in reducing the risk of stroke recurrence is not present in CYP2C19 LoFA noncarriers with overweight/obesity. Our study suggests that BMI significantly influences the correlation between CYP2C19 genotype and efficacy of clopidogrel-aspirin therapy.     Abstract: The uptake of proven stroke treatments varies widely. We aimed to determine the association of evidence‐based processes of care for acute ischemic stroke ( AIS ) and clinical outcome of patients who participated in the HEADPOST (Head Positioning in Acute Stroke Trial), a multicenter cluster crossover trial of lying flat versus sitting up, head positioning in acute stroke. Methods and Results Use of 8 AIS processes of care were considered: reperfusion therapy in eligible patients; acute stroke unit care; antihypertensive, antiplatelet, statin, and anticoagulation for atrial fibrillation; dysphagia assessment; and physiotherapist review. Hierarchical, mixed, logistic regression models were performed to determine associations with good outcome (modified Rankin Scale scores 0–2) at 90 days, adjusted for patient and hospital variables. Among 9485 patients with AIS, implementation of all processes of care in eligible patients, or “defect‐free” care, was associated with improved outcome (odds ratio, 1.40; 95% CI, 1.18–1.65) and better survival (odds ratio, 2.23; 95% CI , 1.62–3.09). Defect‐free stroke care was also significantly associated with excellent outcome (modified Rankin Scale score 0–1) (odds ratio, 1.22; 95% CI , 1.04–1.43). No hospital characteristic was independently predictive of outcome. Only 1445 (15%) of eligible patients with AIS received all processes of care, with significant regional variations in overall and individual rates. Conclusions Use of evidence‐based care is associated with improved clinical outcome in AIS . Strategies are required to address regional variation in the use of proven AIS treatments.     Abstract: Organostibine‐mediated controlled/living radical polymerization of MMA was successfully achieved in room temperature ionic liquids (RTILs) [bmim]Tf 2 N and [bmim]PF 6 . The ln([M] 0 /[M]) increased linearly with increasing reaction time. The number‐average molecular weights of PMMA increased linearly with conversion, and the polydispersity indices were less than 1.15 and narrower in RTILs than in bulk. The living characters of SBRP were further demonstrated by chain extension of PMMA and preparation of copolymer PMMA ‐b‐ PS in RTILs. Recycle and reuse of RTILs were efficient with well‐controlled PMMA produced in the recycled RTILs. SBRP of St in RTILs was also discussed. The polymerization could be controlled only at low St conversion due to the heterogeneous reaction. magnified image

    Abstract: Heterostructures are widely fabricated for promotion of photogenerated charge separation and solar cell/fuel production. (Oxy)nitrides are extremely promising for solar energy conversion, but the fabrication of heterostructures based on nitrogen‐containing semiconductors is still challenging. Here, a simple ammonia thermal synthesis of a heterostructure (denoted as Ta 3 N 5 /BTON) composed of 1D Ta 3 N 5 nanorods and BaTaO 2 N (BTON) nanoparticles (0D), which is demonstrated to result in a remarkable increase in photogenerated charge separation and solar hydrogen production from water, is introduced. As analyzed and discussed, the Ta 3 N 5 /BTON heterostructure is type II and tends to create intimate interfaces between the 1D nanorods and 0D nanoparticles. The 1D Ta 3 N 5 nanorods are demonstrated to transfer electrons along the rod orientation direction. Furthermore, the intimate interfaces of the heterostructure are believed to originate from the similar Ta‐based octahedron units of Ta 3 N 5 and BTON. All of the above features are expected to integrally endow increased photoinduced charge separation and one order of magnitude higher solar overall water splitting activity with respect to counterpart systems. These results may open a new avenue to fabricate heterostructures on the basis of nitrogen‐containing semiconductors that is extremely promising for solar energy conversion.

    Abstract: (Oxy)nitride‐based nanophotocatalysts have been extensively investigated for solar‐to‐chemical conversion, and not only allow wide spectral utilization to achieve high theoretical energy conversion efficiency but also exhibit suitable conduction and valence band positions for robust reduction and oxidation of water. During the past decades, a few reviews on the research progress in designing and synthesizing new visible‐light‐responsive semiconductors for various applications in solar‐to‐chemical conversion have been published. However, those on the effects of their bulk and composite (surface/interface) nanostructures on basic processes as well as solar water splitting performances to produce hydrogen are still limited. In this review, a brief introduction on the relationship between the nanostructure photocatalytic properties is included. Three main processes of solar water splitting are involved, allowing the elucidation of the correlation with the nanostructural properties of the photocatalyst such as surface/interface, size, morphology, and bulk structure. Subsequently, the development of methodologies and strategies for modulating the bulk and composite structures to improve the efficiencies of the basic processes, particularly charge separation, is summarized in detail. Finally, the prospects of (oxy)nitride‐based photocatalysts such as controlled synthesis, modulation of 1D/2D morphology, exposed facet regulation, heterostructure formation, theoretical simulation, and time‐ and space‐resolved spectroscopy are discussed.
