    Abstract: Genetic variants in the fat mass- and obesity-associated gene Fto are linked to the onset of obesity in humans. The causal role of the FTO protein in obesity is supported by evidence obtained from transgenic mice; however, the underlying molecular pathways pertaining to the role of FTO in obesity have yet to be established. In this study, we investigate the Fto gene in mouse brown adipose tissue and in the browning process of white adipose tissue. We analyze distinct structural and molecular factors in brown and white fat depots of Fto-deficient mice under normal and obesogenic conditions. We report significant alterations in the morphology of adipose tissue depots and the expression of mRNA and microRNA related to brown adipogenesis and metabolism in Fto-deficient mice. Furthermore, we show that high-fat feeding does not attenuate the browning process of Fto-deficient white adipose tissue as observed in wild-type tissue, suggesting a triggering effect of the FTO pathways by the dietary environment.

    Abstract: In the last 30 years the adipose cell has been object of several studies, turning its reputation from an inert cell into the main character involved in the pathophysiology of multiple diseases, including the ongoing COVID-19 pandemic, which has changed the clinical scenario of the last two years. Composed by two types of tissue (white and brown), with opposite roles, the adipose organ is now classified as a real endocrine organ whose dysfunction is involved in different diseases, mainly obesity and type 2 diabetes. In this mini-review we aim to retrace the adipose organ history from physiology to physiopathology, to provide therapeutic perspectives for the prevention and treatment of its two main related diseases (obesity and type 2 diabetes) and to summarize the most recent discoveries linking adipose tissue to COVID-19.

    Abstract: Several evidences suggest that the functional β cell defects anticipate the onset of hyperglycemia and the reduction of mass. To assess whether the β-cell deterioration corresponds to a loss of β-cell mass, we performed glucose oral tolerance test (OGTT) and hyperglycemic clamp (HC) followed by arginine stimulation, in 16 nondiabetic patients who underwent pancreatoduodenectomy (PD), pre and post-surgery. Based on the post-operative OGTT, the subjects were divided into 3 glucose tolerance groups: normal (NGT, n = 5), altered (IGT, n = 4) or diabetes (DM, n = 7) (8F/8M, 51 ± 15 years). To evaluate β cell function, βcell glucose sensitivity (GS) was calculated during HC, as a ratio between insulin secretion and glucose increase. On surgical pancreas samples, the% of intraparenchymal white adipose tissue (WATi score) was measured. Before surgery, insulin secretion after arginine (AIS) was similar between groups, while GS was lower in IGT and DM compared to NGT subjects (62.9 ± 23.1 and 45.5 ± 11.2 vs. 90.6 ± 18.7 pmol • min-1 • m-2, respectively). After 50% PD, the GS decreased in all patients (p & lt;0.01 for all groups), but the reduction was greater in DM than in patients with IGT and NGT (ΔGS: NGT -0.20 ± 0, 19 vs. IGT -0.27 ± 0.11 vs. DM 0.37 ± 0.08, p & lt;0.003). A similar reduction was observed in ΔAIS (NGT -0.38 ± 0.13 vs. IGT -0.76 ± 0.06 vs. DM -0.90 ± 0.04; p & lt;0.01) and in the second phase of insulin secretion. WAT score was significantly higher in IGT and DM than in NGT (WATi score IGT 37.75 ± 11.07%, DM 44.46 ± 31.83%, NGT 5.8 ± 2.35%, p & lt;0.01 and p & lt;0.05 respectively). The loss of GS is the most sensitive predictive parameter of diabetes onset after a 50% reduction in mass.     Abstract: β cells dedifferentiation has been recently introduced as the main mechanism responsible for the functional “disappearance” of β cells from the islands of diabetic subjects. Starting from results obtained from a mouse model of type 2 (T2) diabetes (in whose a significant increase in density of noradrenergic fibers was detected compared to nondiabetic mice), we asked if the result could be replicated in humans and if it could be related to the process of dedifferentiation. Human pancreas of 8 healthy donors and 9 with T2 diabetes were analyzed by immunohistochemistry (M / F 4/4 vs. 5/4 NS; BMI (kg / m2) 24.8 ± 2.8 vs. 25.6 ± 4.1; NS). The dedifferentiation score was calculated as % of cells synaptophysin positive but negative for the four major pancreatic hormones. Tyrosine hydroxylase (TH) was used as a marker for the evaluation of noradrenergic fiber expression. The islands of diabetic subjects were about 3 times more innervated than controls (0.42 ± 0.51 vs. 1.66 ± 2.2 n.fibersTH +/island; p = 0.02); the increase of these fibers correlated positively with the dedifferentiation score (p & lt;0.001; r = 0.69). In conclusion, our data show an increase in the number of sympathetic nerve fibers potentially able to transmit inhibitory signals on insulin secretion in the islands of diabetic subjects. The important correlation with the dedifferentiation score suggests a significant role of noradrenergic fibers in the pathogenesis of the dedifferentiation process, introducing new strategies for the preservation of cellular mass/secretion and, therefore, for the prevention and treatment of T2 diabetes.     Abstract: Background: Lynch syndrome (LS) is a genetic disease with increased risk of colorectal cancer and other malignancies. There are few reported cases of thyroid cancer in LS patients. The aim of this study is to investigate the presence of thyroid nodules in LS patients and to explore their association with the genetic features of the disease. Methods: A retrospective and descriptive analysis was conducted to include all LS patients followed at the CEMAD (Centro Malattie Apparato Digerente) of Fondazione Policlinico Universitario A. Gemelli IRCCS. The characteristics of LS disease, gene mutations, and previous history of thyroid disease were evaluated. Majority of patients underwent thyroid ultrasound (US), and nodule cytology was performed when needed. Results: Of a total of 139 patients with LS, 110 patients were included in the study. A total of 103 patients (74%) underwent thyroid ultrasound examinations, and 7 patients (5%) had a previous history of thyroid disease (cancer or multinodular goiter). The mean age was 51.9 years. Thyroid nodules were found in 62 patients (60%) who underwent US, and 9 of them (14%) had suspicious features of malignancy, inducing a fine-needle aspiration biopsy. A cytologic analysis classified 7 of 9 cases (78%) as TIR2 and 2 (22%) as TIR3a. Between patients with nodular thyroid disease (single nodule, multinodular goiter, and cancer), most of them (25 patients, 36% of total) were carriers of the MSH6 mutation, while 22 (32%), 17 (24%), and 5 (7%) had MSH2, MLH1, and PMS2 mutations, respectively. Conclusions: A high prevalence of thyroid nodules was found in patients with LS, especially in MSH6-carrying patients. Performing at least one thyroid ultrasound examination is suggested for the detection of nodular thyroid disease in LS patients. Systematic investigations are needed to estimate their prevalence, features, and risk of malignant transformation.

    Abstract: In this work, the consequences of a local gorgonian coral mortality on the whole coralligenous assemblage were studied. A Before/After-Control/Impact sampling design was used: the structure of the coralligenous assemblage was compared before and after the gorgonian mortality event at the mortality site and two control sites. At the mortality site, a relevant decrease in alpha and beta diversity occurred, with a shift from a stratified assemblage characterized by gorgonians and other invertebrates to an assemblage dominated by algal turfs; conversely, neither significant variations of the structure nor decrease in biodiversity were observed at the control sites. The assemblage shift involved the main taxa in different times: in autumn 2018, a large proportion of the plexaurid coral Paramuricea clavata died, but no significant changes were observed in the structure of the remaining assemblage. Then, in autumn 2019, algal turfs increased significantly and, one year later, the abundance of the gorgonian Eunicella cavolini and bryozoans collapsed. Although the mechanisms of the assemblage shift following gorgonian loss will remain uncertain and a cause-effect relationship cannot be derived, results suggest the need for detecting signs of gorgonian forests stress in monitoring programs, which should be considered early indicators of their condition. in the coralligenous monitoring programs for detecting any sign of gorgonian forests stress which should be considered an early indicator of the assemblage condition.

    Abstract: Seagrass planting techniques have shown to be an effective tool for restoring degraded meadows and ecosystem function. In the Mediterranean Sea, most restoration efforts have been addressed to the endemic seagrass Posidonia oceanica, but cost-benefit analyses have shown unpromising results. This study aimed at evaluating the effectiveness of environmental engineering techniques generally employed in terrestrial systems to restore the P. oceanica meadows: two different restoration efforts were considered, either exploring non-degradable mats or, for the first time, degradable mats. Both of them provided encouraging results, as the loss of transplanting plots was null or very low and the survival of cuttings stabilized to about 50%. Data collected are to be considered positive as the survived cuttings are enough to allow the future spread of the patches. The utilized techniques provided a cost-effective restoration tool likely affordable for large-scale projects, as the methods allowed to set up a wide bottom surface to restore in a relatively short time without any particular expensive device. Moreover, the mats, comparing with other anchoring methods, enhanced the colonization of other organisms such as macroalgae and sessile invertebrates, contributing to generate a natural habitat.

    Abstract: Adherence to vaccination recommendations is a challenge for national immunization programs. We quantified adherence to COVID-19 vaccination recommendations in people with substance use disorders (SUDs) attending an outpatient addiction center in Rome, Italy; we investigated the determinants of adherence, and also analyzed patient risk perception and compliance with preventive measures. A multivariable logistic regression model identified predictors of adherence to vaccination recommendations, with statistical validity tested by estimating adjusted odds ratios (aORs) and 95% confidence intervals (CIs). From December 2021 to January 2022, 200 SUD patients completed a questionnaire, 80% of whom reported being vaccinated against SARS-CoV-2 (minimum one dose). Negative predictors of vaccine uptake included being non-Italian (aOR: 0.36, 95% CI: 0.13–0.97), having coexisting comorbidities (aOR: 0.35, 95% CI: 0.13–0.95), and previous use of heroin (aOR: 0.24, 95% CI: 0.08–0.71). No difference was found for cocaine use, demographic characteristics, previous COVID-19 infection, methadone therapy, or compliance with preventive measures. Major reasons for non-adherence to vaccination recommendations were fear of side effects, insufficient recognition of the importance of vaccination, bureaucratic issues, and lack of trust in the authorities. Given their vulnerability, additional efforts are needed to facilitate access to vaccination for people with SUDs, and to limit disinformation around vaccines..

    Abstract: Interactions between fishing and dolphins can be detrimental, since on one hand dolphins can be lethally entangled by nets and trawls, and on the other dolphins can predate fish caught by nets. For dolphins, this interaction can be dangerous as they can be wounded or accidentally killed; for fishers, the predation of their catch results in economic losses due to reduced quantity and/or quality of catches and damage to fishing gear. During July and November 2020, we surveyed the “dolphin–fisheries conflict” through compiling 209 fisher interviews from nine locations in Italy and Croatia. Fishers mentioned the common bottlenose dolphin (Tursiops truncatus) as the species primarily interacting with fishing, with the major issue being catch damage by predation. The interaction probability varied among gears and seasons, with some fishing activities (e.g., passive nets) more affected than others (e.g., bottom trawls), especially in terms of economic loss (1000–10,000 €/year on average). More than 70% of the fishers claimed that dolphin populations have increased over the last 10 years, in different degrees and based on different areas. Dolphin bycatch rates are generally low; however, 34.6% of respondents reported having captured at least one dolphin during their career. The fishers’ attitude towards acoustic deterrents (“pingers”) as a mitigation measure revealed that few of them were aware of these devices or were using them.
