    Abstract: A widespread and diversified mesophotic system of carbonate bioconstructions along the Southern Adriatic and Ionian Italian coasts is described, providing new data on the distribution, structure and associated megabenthic assemblages of mesophotic Mediterranean bioconstructions. The bioconstructions were detected at six different sites off the coasts of Apulia, in presence of marked morphological escarpments, developing on a basal substrate consisting of meso-Cenozoic carbonate rocks. Two biogenic structure types were observed, one mainly built by the nonsymbiotic scleractinians Phyllangia americana mouchezii and Polycyathus muellerae , at depths between approximately 35 and 55 m, and the other by the oyster Neopycnodonte cochlear , at depths from approximately 40 to 70 m. A total of 52 taxa of megabenthic invertebrates, belonging to 6 phyla, were found on the surface of the mesophotic bioconstructions, thus confirming the role of biodiversity hotspots of these carbonate structures. Megabenthic assemblages showed a remarkable heterogeneity both in pattern of species and abundance, probably depending on both the morphological differences of the seabed and the life traits of the single species. Primary bioconstructors seemed to influence the associated community pattern. This peculiar system deserves sound conservation measures in the light of the holistic ecosystem approach for the management of coastal marine areas.

    Abstract: Seahorses are considered a flagship species for conservation efforts and due to their conservation status, improving knowledge on their dietary composition while applying a non-invasive approach, could be useful. Using Hippocampus guttulatus as a case study, the present study represents pioneering research into investigating the diet of seahorses by NGS-based DNA metabarcoding of fecal samples. The study developed and tested the protocol for fecal DNA metabarcoding during the feeding trials where captive seahorses were fed on a diet of known composition; the process was subsequently applied on fecal samples collected from wild individuals. The analysis of samples collected during the feeding trials indicated the reliability of the applied molecular approach by allowing the characterization of the effectively ingested prey. In the field study, among detected prey species, results revealed that the majority of the seahorse samples contained taxa such as Amphipoda, Decapoda, Isopoda, and Calanoida, while less common prey taxa were Gastropoda and Polyplacophora. As only a small amount of starting fecal material is needed and the sampling procedure is neither invasive nor lethal. The present study indicates DNA metabarcoding as useful for investigating seahorse diet and could help define management and conservation actions.

    Abstract: The present paper provides a multidisciplinary fine-scale description of a Mediterranean mesophotic new habitat dominated by the bivalve Neopycnodonte cochlear (Poli, 1795), building large and thick pinnacles on vertical cliffs at two study areas along the southern Italian coast. The pinnacles, constituted by a multilayered aggregation of living and dead specimens of N. cochlear , were interconnected with each other to form a framework of high structural complexity, never observed before for this species. The bioconstruction, considerably extended, resulted very complex and diversified in the associated community of structuring organisms. This latter included 165 taxa attributable to different ecological groups occurring in different microhabitats of the bioconstruction. Among the secondary structuring taxa there were scleractinians, serpulids and bryozoans, all contributing to the deposition of calcium carbonate, and poriferans, helping to bind shells together or eroding carbonate by boring species. In comparison with coralligenous sensu stricto and the recently described Mediterranean mesophotic coral reef, the Neopycnodonte bioconstruction showed peculiar features, since it lacked the major contribution of encrusting coralline algae and scleractinians as reef builders, respectively.

    Abstract: The seahorse Hippocampus guttulatus reaches its highest abundance in confined environments, where it has unique biological and ecological traits that suggest significant genetic differentiation among populations. In the present study, we aimed to reveal the genetic structure of this species by analysing eight microsatellite loci and a mitochondrial DNA region (cytochrome b) of eight populations from the Central–Western Mediterranean Sea, including lagoon sites. Levels of genetic diversity, as measured by the total number of alleles, number of private alleles, allelic richness and heterozygosity, ranged from low to moderate. The overall value of inbreeding was high, indicating a deficiency in heterozygotes. The haplotype network had a star-like construction, with the most common haplotype present in all populations. Data from the two molecular markers congruently displayed a similar pattern and revealed low genetic differentiation, notwithstanding predictions based on species traits. The observed genetic structure is probably the result of both historical population demographic events and current gene flow. The investigated lagoons, however, revealed a unique genetic profile, which is especially highlighted by the Taranto population. At this site, the results also showed altered values of observed/expected heterozygosity and allelic richness, a characteristic of marginal populations. Our study suggests that lagoon populations should be managed as distinct genetic units.

    Abstract: Population abundance, distribution and habitat preference of the M editerranean sympatric seahorses H ippocampus guttulatus and H ippocampus hippocampus were investigated in a semi‐enclosed sea system ( A pulian coast, I onian S ea). A total of 242 individuals of seahorses were sighted in the 11 transects surveyed in summer 2011. H ippocampus guttulatus (n = 225) were 14 times more abundant than H . hippocampus (17). The mean abundance of H . guttulatus for all the pooled sites was 0.018 m −2 ( SE  ± 0.003) ranging from a maximum of 0.035 ( SE  ± 0.007) to a minimum of 0.008 ( SE  ± 0.002). The size structure of long‐snouted seahorse shows a population ranging from 7 to 14 cm ( SL ) with a peak at 10 cm ( TL ). Juveniles (96.0 ± 8.0 mm) represent a significant fraction of the population, accounting more than 21% of the sighted individuals. In M ar P iccolo, H . guttulatus is able to shelter both in monotonous habitats, including the algal beds, and diversified ones, such as the rich filter‐feeder communities that colonize hard substrates. By contrast, H . hippocampus is mainly associated with habitats of low complexity. Today, the M ar P iccolo di T aranto is among the most heavily polluted water bodies in S outh I taly, with trace metals, hydrocarbons, pesticides and organic wastes affecting both biotic and abiotic matrices. However, despite the high level of degradation, the presence of a large mussel farm has avoided the impact of towed fishing gears, and eutrophication of water bodies has ensured a high trophic level that supports large crustacean populations, potential prey for seahorses.

    Abstract: We report the presence of the non-indigenous species (NIS) Celleporaria brunnea (Bryozoa, Ascophora) in the Central Mediterranean. It was found in the Gulf of Taranto (Ionian Sea, southern Italy), which represents one of the most important hotspots for NIS in the Mediterranean. Using PVC plates as species collectors, immersed in different times of the year, we observed a recruitment of C. brunnea during the warmer period of the year from July to October. This work highlights a range expansion of C. brunnea and provides information about its autoecology traits in the host environment.

    Abstract: Confined marine environments are particularly susceptible to climate change and anthropic pressures. Indeed, the long-term monitoring of benthic assemblages in these environments allows us to understand the direction of changes over time. The demosponge Geodia cydonium is a suitable study case, since it is widely represented in many Mediterranean environments, while being a long-living and important habitat-forming species. Here, we report the results of a descriptive study on temporal and spatial variations of this demosponge in three semi-enclosed environments along the Italian coast: Marsala Lagoon, Porto Cesareo Bay, and Mar Piccolo of Taranto. At Marsala and Porto Cesareo, the study compares the present data with those reported by the literature at the end of the 1990s. Caused by the modification of its preferential habitats, the data indicated the loss and a remarkable regression of this species at Porto Cesareo and Marsala, respectively. In addition, we hypothesized that the increase in severe weather phenomena in the small Bay of Porto Cesareo recorded during the last 20 years may have had a marked impact on water mass, thus affecting the sponge assemblages. At Taranto, despite a remarkable environmental degradation, G. cydonium has appeared stable and persistent in the last 45 years, thus representing one of the richest and most well-preserved populations in the Mediterranean Sea.

    Abstract: Bryozoan assemblages of three mesophotic habitats, recently found in the Adriatic and Ionian Seas, were investigated in terms of species composition and diversity, colonial growth forms, and species ecological affinity, with the purposes to analyze variations and similarities between the different bioconstructions and to compare the mesophotic assemblages with those of other benthic habitats. Bryozoans came from three sites off the Apulian coast: Monopoli, 30–55-m depth, where scleractinians are dominant; Otranto, 45–64-m depth; and Santa Maria di Leuca, 45–70-m depth, where the bivalve Neopycnodonte cochlear is the main bioconstructor. A total number of 50 species of bryozoans were recorded, accounting for about 10% of the total number of Mediterranean species. Only few species were shared between the three sites, whereas a considerable pool of species was characteristic of each site. The β-diversity and Sörensen similarity analyses highlighted the highest similarity between the two southern sites, Otranto and Santa Maria di Leuca, with respect to the northern site of Monopoli, in agreement with the hydrological pattern of the area. The encrusting zoarial type was the most abundant in each site, and few species with erect, celleporiform and petraliform colonies were found. The Sörensen similarity, based on data of zoarial forms composition, revealed values considerably higher than those based on species composition, thus highlighting the similar ecological role played by the bryozoans in all the sites. Moreover, significant differences between the mesophotic bryozoan assemblages and those of other benthic Mediterranean habitats were highlighted, showing higher affinity with the coralligenous and detritic sciaphilic habitats. The relevant ecological role of bryozoans as binders in the bioconstructions of the Mediterranean mesophotic zone has been exhibited.
