    Abstract: A continuous floc image analyser (C-FIA) was developed for tracking the dynamics of floc particle formation, growth and settling during conventional drinking water treatment processes, comprising coagulation, flocculation and settling. For this, a script was developed to (1) extract the video frames and sync these with reaction time ( t ), (2) transform the video frames to an equivalent matrix of pixel RGB data, (3) optically filter the video frames for background effects, (4) calculate the filtered (corrected for background) flocculation index (C-FIA FI t ) for the frame belonging to each process time through RGB data and (5) correct the C-FIA FI t for baseline RGB signal (raw water frame, C-FIA FI 0 ). The video frames collected by C-FIA have potential to provide direct information on floc size and concentration. The C-FIA FI data were acquired from jar tests using a range of alum doses performed on three natural surface water samples with highly contrasting water quality. The C-FIA FI data acquired, displayed lower variance in comparison with available data of photometric dispersive analysis (PDA). The indices previously developed for PDA analysis were utilized in this study for C-FIA FI data, including plateau index. In experiments conducted, this index showed an inverse relationship with residual turbidity, UVA 254 and colour following coagulation. This indicates a potential for C-FIA to be applied for coagulant dose control at conventional drinking water treatment plants. On the other hand, from the experimental work performed, impacts of light intensity and light radiation angle to the photography cell on the resulting FI value were found. These can impact on the pixels' red/green/blue (RGB) values of the video frames/images, and consequently can impact on the resultant C-FIA FI data.

    Abstract: In this study, an enhanced electro-coagulation (En-EC) technique is described for removal of natural dissolved organic matter (DOM) from surface drinking water sources. Assessment of the En-EC technique included investigating the impacts of operational factors on DOM removal efficiency, DOM characteristic variations, and energy consumption ( C E ). DOM removal efficiencies by En-EC were compared with removals by enhanced chemical coagulation (EnC) along with residual metal levels. The removal of DOM was assessed by tracking the % reduction ( R %) of dissolved organic carbon (DOC) and its surrogate parameters ( i.e. UV absorbance at 254 nm ( A 254 ), colour, fluorophore DOM fluorescence signal (fDOM s )) following the En-EC process. Experiments were conducted using samples collected from three surface drinking water sources, i.e. , Murray River, Myponga River and Middle River located in South Australia, Australia. An electrocoagulation cell was assembled and equipped with capability for measuring consumed C E and electrical charge ( Q ). The impacts of process parameters ( i.e. I EC , the process pH [pH EC ], and the aluminium coagulant dose [Al EC ]) on the R % and C E were also studied. The DOM characteristic variations following the process were studied using UV-vis spectroscopy, fluorescence excitation–emission (ex/em) (matrix) spectroscopy and high performance size exclusion chromatography equipped with UV and fluorescence ex/em spectrophotometer detectors. An optimum pH EC of 6 was obtained experimentally for the En-Ec process. A comparable DOM removal efficiency was observed for either Al EC in En-EC or alum in EnC. A maximum % A 254 removal of 74% to 92% was observed for the studied water sources, depending on the DOM characteristics. A comparable DOM removal performance was observed for En-EC and EnC. This was a basis for adopting a previously developed decision support system for alum dosing in the EnC process, for Al EC dosing control in the En-EC process.

    Abstract: Blood cells derive from hematopoietic stem cells through stepwise fating events. To characterize gene expression programs driving lineage choice, we sequenced RNA from eight primary human hematopoietic progenitor populations representing the major myeloid commitment stages and the main lymphoid stage. We identified extensive cell type–specific expression changes: 6711 genes and 10,724 transcripts, enriched in non–protein-coding elements at early stages of differentiation. In addition, we found 7881 novel splice junctions and 2301 differentially used alternative splicing events, enriched in genes involved in regulatory processes. We demonstrated experimentally cell-specific isoform usage, identifying nuclear factor I/B (NFIB) as a regulator of megakaryocyte maturation—the platelet precursor. Our data highlight the complexity of fating events in closely related progenitor populations, the understanding of which is essential for the advancement of transplantation and regenerative medicine.

    Abstract: Speech-language pathologists (SLPs) encounter many challenges in the school environment. We sought to identify school-based SLPs' needs in the areas of training and research to support school-based practice. Method We collected surveys from 145 school-based SLPs practicing in Wisconsin. Respondents were asked to describe aspects of preservice training that most and least prepared them for school-based practice. Respondents next described training needs for early-career SLPs and desired topics for general professional development. Respondents then described research topics that require further study to advance school-based practice. After completing this series of open-ended questions, the respondents completed a series of preferential-ranking questions about general needs for training and research. We employed mixed methods to analyze the data, using grounded theory for the qualitative analysis. Results During preservice training, respondents most valued the applied aspects of their training. Respondents noted that early-career SLPs require significant support, particularly with the professional responsibilities of school-based practice (e.g., individualized education programs, interprofessional practice, and scheduling). When describing general professional development and research needs, the respondents placed high value on practical topics that have clear implications for school-based practice. Conclusions Understanding the perceived needs of school-based SLPs can assist in designing training supports and ensuring that the developing evidence base is relevant to SLPs practicing in the schools. This exploration-stage study established a foundation for future testing of implementation initiatives to fill gaps in school-based practice.
