    Abstract: Polygonatum kingianum Collett et Hemsl. var. grandifolium D.M. Liu & amp; W.Z. Zeng (1981), which sprouts twice a year, once in spring and once in autumn, differs from Polygonatum kingianum in leaves, bracts, perianth and filaments. Morphological comparison and molecular phylogeny indicate that it is identical to the newly-published Polygonatum hunanense H.H. Liu & amp; B.Z. Wang (2021). Hence, we propose that P. kingianum var. grandifolium should be recognised as a new synonym of P. hunanense . In addition, phylogenetic analyses confirmed that P. hunanense is sister to Polygonatum sect. Polygonatum, rather than P. kingianum of Polygonatum sect. Verticillata.

    Abstract: Haemaphysalis longicornis is drawing attentions for its geographic invasion, extending population, and emerging disease threat. However, there are still substantial gaps in our knowledge of viral composition in relation to genetic diversity of H. longicornis and ecological factors, which are important for us to understand interactions between virus and vector, as well as between vector and ecological elements. Results We conducted the meta-transcriptomic sequencing of 136 pools of H. longicornis and identified 508 RNA viruses of 48 viral species, 22 of which have never been reported. Phylogenetic analysis of mitochondrion sequences divided the ticks into two genetic clades, each of which was geographically clustered and significantly associated with ecological factors, including altitude, precipitation, and normalized difference vegetation index. The two clades showed significant difference in virome diversity and shared about one fifth number of viral species that might have evolved to “generalists.” Notably, Bandavirus dabieense , the pathogen of severe fever with thrombocytopenia syndrome was only detected in ticks of clade 1, and half number of clade 2-specific viruses were aquatic-animal-associated. Conclusions These findings highlight that the virome diversity is shaped by internal genetic evolution and external ecological landscape of H. longicornis and provide the new foundation for promoting the studies on virus-vector-ecology interaction and eventually for evaluating the risk of H. longicornis for transmitting the viruses to humans and animals.

    Abstract: Theileria , a tick-borne intracellular protozoan, can cause infections of various livestock and wildlife around the world, posing a threat to veterinary health. Although more and more Theileria species have been identified, genomes have been available only from four Theileria species to date. Here, we assembled a whole genome of Theileria luwenshuni , an emerging Theileria , through next-generation sequencing of purified erythrocytes from the blood of a naturally infected goat. We designated it T. luwenshuni str. Cheeloo because its genome was assembled by the researchers at Cheeloo College of Medicine, Shandong University, China. The genome of T. lunwenshuni str. Cheeloo was the smallest in comparison with the other four Theileria species. T. luwenshuni str. Cheeloo possessed the fewest gene gains and gene family expansion. The protein count of each category was always comparable between T. luwenshuni str. Cheeloo and T. orientalis str. Shintoku in the Eukaryote Orthologs annotation, though there were remarkable differences in genome size. T. luwenshuni str. Cheeloo had lower counts than the other four Theileria species in most categories at level 3 of Gene Ontology annotation. Kyoto Encyclopedia of Genes and Genomes annotation revealed a loss of the c-Myb in T. luwenshuni str. Cheeloo. The infection rate of T. luwenshuni str. Cheeloo was up to 81.5% in a total of 54 goats from three flocks. The phylogenetic analyses based on both 18S rRNA and cox1 genes indicated that T. luwenshuni had relatively low diversity. The first characterization of the T. luwenshuni genome will promote better understanding of the emerging Theileria . IMPORTANCE Theileria has led to substantial economic losses in animal husbandry. Whole-genome sequencing data of the genus Theileria are currently limited, which has prohibited us from further understanding their molecular features. This work depicted whole-genome sequences of T. luwenshuni str. Cheeloo, an emerging Theileria species, and reported a high prevalence of T. luwenshuni str. Cheeloo infection in goats. The first assembly and characterization of T. luwenshuni genome will benefit exploring the infective and pathogenic mechanisms of the emerging Theileria to provide scientific basis for future control strategies of theileriosis.

    Abstract: Atherosclerosis treatments by gene regulation are garnering attention, yet delivery of gene cargoes to atherosclerotic plaques remains inefficient. Here, we demonstrate that assembly of therapeutic oligonucleotides into a three-dimensional spherical nucleic acid nanostructure improves their systemic delivery to the plaque and the treatment of atherosclerosis. This noncationic nanoparticle contains a shell of microRNA-146a oligonucleotides, which regulate the NF-κB pathway, for achieving transfection-free cellular entry. Upon an intravenous injection into apolipoprotein E knockout mice fed with a high-cholesterol diet, this nanoparticle naturally targets class A scavenger receptor on plaque macrophages and endothelial cells, contributing to elevated delivery to the plaques (∼1.2% of the injected dose). Repeated injections of the nanoparticle modulate genes related to immune response and vascular inflammation, leading to reduced and stabilized plaques but without inducing severe toxicity. Our nanoparticle offers a safe and effective treatment of atherosclerosis and reveals the promise of nucleic acid nanotechnology for cardiovascular disease.

    Abstract: Building anti-floating anchors have been increasingly used in recent years, but conventional steel anchors under service conditions are easily subjected to chemical erosion. Glass fiber reinforcement polymer (GFRP) is a promising solution to this problem. In this study, field pullout tests were conducted on three full instrumented GFRP anti-floating anchors in weathered granite. Specifically, the GFRP anchors during pultrusion were innovatively embedded with bare fiber Bragg grating (FBG) sensors to monitor the axial force distribution along depth. It was found that the embedded FBG could reliably monitor the axial force distribution of GFRP anchors. The ultimate pullout force of a GFRP anchor with diameter of 28 mm and anchorage length of 5 m was up to 400 kN. The GFRP anchor yielded at 0.8 m underground. Force distribution and field photos at failure indicated shear failure occurred at the anchor/bolt interface at the end of the tests. The feasibility of the GFRP anti-floating anchor was also verified in civil engineering. Finally, an elastic mechanical model and Mindlin’s displacement solution are used to get distribution functions of axial force and shear stress along the depth, and the results accord with the test results.

    Abstract: Neutron capture reaction is one of the neutron reactions and plays an important role in using reactor control rods and shell materials, designing nuclear device structures, and studying nuclear astrophysics S processes and element origins. The 4π BaF 〈 sub 〉 2 〈 /sub 〉 detection device has advantages such as high time resolution, low neutron sensitivity, and high detection efficiency, thus making it suitable for measuring neutron radiation capture reaction cross-section data. In order to fill the gap in our neutron capture reaction data in the keV energy range and improve their accuracy, the Key Laboratory of Nuclear Data at the Chinese Institute of Atomic Energy (CIAE) has established a Gamma Total Absorption Facility (GTAF), which consists of 28 hexagonal BaF 〈 sub 〉 2 〈 /sub 〉 crystals and 12 pentagonal BaF 〈 sub 〉 2 〈 /sub 〉 crystals to form a spherical shell with an external diameter of 25 cm and an internal diameter of 10 cm, covering 95.2% of the solid angles. The Back-n beam line of the Chinese Spallation Neutron Source (CSNS) is a back-streaming white beam line that covers neutron energy ranging from a few eV to several hundred MeV, making it suitable for measuring neutron capture cross-sections. The reaction cross-section data of 〈 sup 〉 197 〈 /sup 〉 Au is measured by using GTAF on the Back-n beam line. The measurement data are preliminarily background deducted through energy screening, PSD method, and crystal multiplicity screening. Subsequently, the background is analyzed and deducted based on the measurement data of 〈 sup 〉 nat 〈 /sup 〉 C and empty samples, and the yield of 〈 sup 〉 197 〈 /sup 〉 Au capture reaction is obtained. Resonance parameters are a set of parameters extracted from experimental data to describe the resonance curve, which can eliminate the influence of experimental conditions on resonance data and are more important than the cross-section obtained from experiments. The resonance energy, neutron resonance width, and gamma resonance width parameters of 〈 sup 〉 197 〈 /sup 〉 Au at 1–100 eV are fitted by using the SAMMY program. From the comparison between the resonance curves obtained from experimental measurements and the resonance parameters obtained from fitting with the ENDF/B-VIII.0 database, it can follow that the experimental measurement results are in good agreement with the database, nevertheless, there exist some differences in the resonance parameter, which may be due to the GTAF energy resolution, Back-n neutron spectrum measurement accuracy, and the experimental background deduction method. Our next work is to identify the sources of difference.

    Abstract: It’s a challenge for detecting the therapeutic targets of a polypharmacological drug from variations in the responsed networks in the differentiated populations with complex diseases, as stable coronary heart disease. Here, in an adaptive, 31-center, randomized, double-blind trial involving 920 patients with moderate symptomatic stable angina treated by 14-day Danhong injection(DHI), a kind of polypharmacological drug with high quality control, or placebo (0.9% saline), with 76-day following-up, we firstly confirmed that DHI could increase the proportion of patients with clinically significant changes on angina-frequency assessed by Seattle Angina Questionnaire (ΔSAQ-AF ≥ 20) (12.78% at Day 30, 95% confidence interval [CI] 5.86–19.71%, P  = 0.0003, 13.82% at Day 60, 95% CI 6.82–20.82%, P  = 0.0001 and 8.95% at Day 90, 95% CI 2.06–15.85%, P  = 0.01). We also found that there were no significant differences in new-onset major vascular events ( P  = 0.8502) and serious adverse events ( P  = 0.9105) between DHI and placebo. After performing the RNA sequencing in 62 selected patients, we developed a systemic modular approach to identify differentially expressed modules (DEMs) of DHI with the Z summary value less than 0 compared with the control group, calculated by weighted gene co-expression network analysis (WGCNA), and sketched out the basic framework on a modular map with 25 functional modules targeted by DHI. Furthermore, the effective therapeutic module (ETM), defined as the highest correlation value with the phenotype alteration (ΔSAQ-AF, the change in SAQ-AF at Day 30 from baseline) calculated by WGCNA, was identified in the population with the best effect (ΔSAQ-AF ≥ 40), which is related to anticoagulation and regulation of cholesterol metabolism. We assessed the modular flexibility of this ETM using the global topological D value based on Euclidean distance, which is correlated with phenotype alteration ( r 2 : 0.8204, P  = 0.019) by linear regression. Our study identified the anti-angina therapeutic module in the effective population treated by the multi-target drug. Modular methods facilitate the discovery of network pharmacological mechanisms and the advancement of precision medicine. (ClinicalTrials.gov identifier: NCT01681316).
