    Abstract: Pyroptosis is a type of inflammatory cell death and is related to pulpitis and apical periodontitis. In this study, the aim was to investigate how periodontal ligament fibroblasts (PDLFs) and dental pulp cells (DPCs) respond to pyroptotic stimuli and explore whether dimethyl fumarate (DMF) could block pyroptosis in PDLFs and DPCs. Methodology Three methods (stimulation with lipopolysaccharide [LPS] plus nigericin, poly(dA:dT) transfection and LPS transfection) were used to induce pyroptosis in PDLFs and DPCs, two types of fibroblasts related to pulpitis and apical periodontitis. THP‐1 cell was used as a positive control. Afterwards, PDLFs and DPCs were treated with or without DMF before inducing pyroptosis to examine the inhibitory effect of DMF. Pyroptotic cell death was measured by lactic dehydrogenase (LDH) release assays, cell viability assays, propidium iodide (PI) staining and flow cytometry. The expression levels of cleaved gasdermin D N‐terminal (GSDMD NT), caspase‐1 p20, caspase‐4 p31 and cleaved PARP were examined by immunoblotting. Immunofluorescence analysis was used to detect the cellular distribution of GSDMD NT. Results Periodontal ligament fibroblasts and DPCs were more sensitive to cytoplasmic LPS‐induced noncanonical pyroptosis than to canonical pyroptosis induced by stimulation with LPS priming plus nigericin or by poly(dA:dT) transfection. In addition, treatment with DMF attenuated cytoplasmic LPS‐induced pyroptotic cell death in PDLFs and DPCs. Mechanistically, it was shown that the expression and plasma membrane translocation of GSDMD NT were inhibited in DMF‐treated PDLFs and DPCs. Conclusions This study indicates that PDLFs and DPCs are more sensitive to cytoplasmic LPS‐induced noncanonical pyroptosis and that DMF treatment blocks pyroptosis in LPS‐transfected PDLFs and DPCs by targeting GSDMD, suggesting DMF might be a promising drug for the management of pulpitis and apical periodontitis.

    Abstract: The aim of this article is to describe the innovations in the School of Stomatology at Wuhan University (WHUSS) that are likely to shape the future of dental education in the People's Republic of China. There are forty dental schools in China; the five most well known are located in Beijing, Chengdu, Shanghai, Xi'an, and Wuhan. Although patient‐centered, comprehensive care has been recommended as the future of dental education, traditional dental education in China still faces many challenges to accomplish this goal. WHUSS, one of the more progressive dental schools in China, has implemented several educational innovations to traditional dental education by restructuring the curriculum through the introduction of problem‐based learning (PBL) and other strategies in clinical training. Although implementation of educational innovations is still at an early stage throughout China, it is reasonable to speculate that many schools will develop similar strategies as those developed at Wuhan to improve dental education during the next several years. However, additional research is necessary to evaluate the efficacy of such educational strategies and to determine the appropriate implementation of a contemporary dental curriculum and pedagogic methodology. The curriculum modifications that have been achieved to date as well as the existing challenges are discussed to provide the reader with an understanding of contemporary dental education in China.

    Abstract: A municipal wastewater treatment plant plays an important role in treating urban sewage and reducing the quantity of pollutants discharged into rivers. However, the energy consumption of the municipal wastewater treatment industry is large. High energy consumption indirectly produces ecological damage, accelerates the energy crisis, and increases carbon emissions. For energy conservation and emission reduction in wastewater treatment plants, it is first necessary to identify the main factors influencing energy consumption. Electricity consumption accounts for more than 80% of the energy consumption of wastewater treatment plants. Wastewater quantity and wastewater quality have become the key influencing factors of energy conservation and consumption reduction in wastewater treatment plants. In this study, a municipal wastewater treatment plant in Northeast China was selected as the research object, and the measured data, such as air temperature, wastewater quantity, wastewater quality, and electricity consumption of the plant from 2017 to 2020 were statistically analyzed to explore the influences of temperature and wastewater quantity and wastewater quality indicators of influent and effluent on energy consumption. Firstly, the range of influent quantity in the wastewater treatment plant was large. The influent quantity in summer was high because some rainwater entered the sewage treatment plant. In winter, average daily electricity consumption (ADEC) was higher than that in summer. The relationship between ADEC and the wastewater quantity showed a positive correlation, and ADEC slowly increased with the increase in wastewater quantity. Electricity consumption per unit of wastewater (UEC) was negatively correlated with the wastewater quantity, but the correction coefficient in winter was larger than that in summer. Secondly, the ranges of chemical oxygen demand (CODCr) and ammonia nitrogen in influent were large, and the ranges of CODCr and ammonia nitrogen in effluent were small. Influent CODCr concentration was negatively correlated with influent ammonia nitrogen concentration. ADEC increased slightly with the increase in influent CODCr concentration. In winter, the increasing trend of ADEC with the influent CODCr concentration was higher than that in the summer. The increasing trend of UEC with the increase in influent COD concentration in summer was more significant than that in winter. Thirdly, influent CODCr in 11.6% of the samples exceeded the corresponding designed value, and influent ammonia nitrogen concentration in 41.4% of the samples exceeded the corresponding designed value. Effluent CODCr in 10.6% of the samples exceeded the First Level Class B standard in “Discharge Standard of Pollutants for Municipal Wastewater Treatment Plants (GB18918-2002)”, and unqualified CODCr in 94% of the effluent samples was ascribed to the unqualified ammonia nitrogen concentration in the influent samples. The electricity consumption level under abnormal conditions was higher than that under normal conditions. Fourthly, ADEC was positively correlated with the average daily CODCr reduction. The correction coefficient of ADEC with average daily CODCr reduction was greater in winter than that in summer. Fifthly, the average electricity consumption per unit of wastewater was close to the national average energy consumption, displaying the characteristics of high energy consumption in winter and low energy consumption in summer. The correlation analysis results of unit electricity consumption and temperature showed that when it was below 0 °C, the lower the temperature, the higher the electricity consumption. In Northeast China, the influences of seasons and temperatures on the electricity consumption of sewage plants were obvious. Accordingly, it is necessary to implement the diversion of rainwater and sewage, reduce the discharge of unqualified wastewater from enterprises, and take thermal insulation measures in winter. In addition, activated sludge microorganisms suitable for a low temperature area and the optimal scheduling of sewage pipe networks can also improve the operation and management of sewage treatment plants.

    Abstract: MG53 is transcriptionally activated by the IRS‐1/PI3K/AKT signal pathway, which is closely related with oncogenesis of several tumors. Here, the role of MG53 in the tumorigenesis of tongue cancer is analyzed in vitro and in vivo. The stable MG53 overexpression/knockdown SCC9 and SCC25 cells are constructed through retrovirus infection. Then a PLGA cylinder is used to provide a 3D culture environment for cell growth. Cell counting results suggest that overexpression of MG53 inhibits the cell proliferation and colony formation of SCC9 and SCC25 cells. While knockdown of MG53 has the opposite effect. Furthermore, knockdown of MG53 significantly promotes the invasion of SCC9 and SCC25 cells. Western blotting data confirm that MG53 affects the expression of the AKT signaling pathway. In a xenograft assay, knockdown of MG53 promotes the growth of xenograft which is induced by SCC25 cells in nude mice. The findings demonstrate that MG53 affects the biological behavior of human tongue cancer SCC9 and SCC25 cells.

    Abstract: The aim of current study is to investigate the in vitro and in vivo behavior of dental pulp stem cells (DPSCs) seeded on electrospun poly(ε‐caprolactone) (PCL)/gelatin scaffolds with or without the addition of nano‐hydroxyapatite (nHA). For the in vitro evaluation, DNA content, alkaline phosphatase (ALP) activity and osteocalcin (OC) measurement showed that the scaffolds supported DPSC adhesion, proliferation, and odontoblastic differentiation. Moreover, the presence of nHA upregulated ALP activity and promoted OC expression. Real‐time PCR data confirmed these results. SEM micrographs qualitatively confirmed the proliferation and mineralization characteristics of DPSCs on both scaffolds. Subsequently, both scaffolds seeded with DPSCs were subcutaneously implanted into immunocompromised nude mice. Scaffolds with nHA but without cells were implanted as control. Histological evaluation revealed that all implants were surrounded by a thin fibrous tissue capsule without any adverse effects. The cell/scaffold composites showed obvious in vivo hard tissue formation, but there was no sign of tissue ingrowth. Further, the combination of nHA in scaffolds did upregulate the expression of specific odontogenic genes. In conclusion, the incorporation of nHA in nanofibers indeed enhanced DPSCs differentiation towards an odontoblast‐like phenotype in vitro and in vivo . © 2009 Wiley Periodicals, Inc.
