Abstract:
Die Stimulation der Phosphorinosit‐Hydrolyse (PI) durch Norepinephrin (NE) Carbachol (Carb) und exzitatorische Aminosäuren (EAA) wurden in Dünnschnitten gemessen, die von Neokortex nach resektiver Epilepsiechirurgie angefertigt wurden. NE und Carb verstärkten den PI‐Umsatz beträchtlich. (EC 50 :NE, 12 μ M und Carb, 661 μ M ) als Ausdruck einer ( 3 H) Jnositol‐Monophosphatanháufung (IP 1 ) in Gewebeschnitten, die vorher ( 3 H‐Myoinositol) markiert worden waren. Dieser Effekt war dosisabhängig bis zur Sättigung und 5–6‐fach über der bas en IP 1 ‐Ansammlung. Eine schwáchere Stimulation (über 2‐fach) wurde mit Quisqualat (QUIS, EC 50 1.1 μ M ) beobachtet, ebenso wie mit Glutamat (GLU, EC 50 1 μ M ), während bei Kainat (KA) und N‐Methyl‐D‐Aspartat (NMDA) nur eine geringe oder gar keine Stimulation gesehen wurde. Der Agonisten‐stimulierte PI‐Umsatz war signifinkant verringert bei Proben aus aktivem spike‐entladendum epileptischen Neokortex gegenuber spikefreien Regione entsprechend dem ECoG (NE ‐23%, p 0.05; Carb ‐ 44%, P 0.01). Vorherige Inkubation der Diinnschnitte mit verschiedenen EAA verhinderte die Carb induzierte IP 1 ‐ Bildung. Das maximale Ausmaβ der Inhibition (1 mM) war sowohl Arninosäuren‐(NMDA KA QUIS GLU) und auch konzentrationsabhängig (IC 5 O: NMDA, 5 μ M ; KA, 3.3 μ M ; QLJIS, 47 μ M ; und GLU I m M ). Diese Ergebnisse lassen vermuten, daβ die epileptische Aktivität den PI‐Metabolismus moduliert und die Rezeptor‐Effektor‐Kopplung verändert. Als wichtige Mediatoren der Epileptogenese könnten die EAA mit der Wirkung dieses zweiten übertragungssystems interferiern.
