    Notes: Abstract: Membranes of the secretory vesicles from bovine adrenal medulla were investigated for the presence of the endogenous protein phosphorylation activity. Seven phosphoprotein bands in the molecular weight range of 250,000 to 30,000 were observed by means of the sodium dodecyl sulphate electrophoresis and autoradiography. On the basis of the criteria of molecular weight, selective stimulation of the phosphorylation by cyclic AMP (as compared with cyclic GMP) and immunoprecipitation by specific antibodies, band 5 (molecular weight 60,300) was found to represent the phosphorylated form of the secretory vesicle-bound tyrosine hydroxylase. The electrophoretic mobility, the stimulatory and inhibitory effects of cyclic AMP in presence of Mg2+ and Zn,2+ respectively, and immunoreactivity toward antibodies showed band 6 to contain two forms of the regulatory subunits of the type II cyclic AMP-dependent protein kinase, distinguishable by their molecular weights (56,000 and 52,000, respectively). Phosphorylation of band 7 (molecular weight 29,800) was stimulated about 2 to 3 times by Ca2+ and calmodulin in the concentration range of both agents believed to occur in the secretory tissues under physiological conditions.

    Notes: Zuxammenfassung Die Zwischenwirbelscheiben sind nicht selten Sitz sowohl hämatogener wie exogener, dann in der Regel iatrogener eitriger Infektionen nichttuberkulöser Art. Bei längerem Bestehen der Infektion werden die angrenzenden Wirbelkörperflächen mitbeteiligt. Klinisch sowie therapeutisch führend ist nicht die Wirbelosteomyelitis, sondern die Bandscheibeninfektion, die das Krankheitsbild und den Verlauf bestimmt. Die Bandscheibeninfektionen sind vor den erst spät, oft zu spät auftretenden unsicheren Röntgenveränderungen diagnostizierbar durch eine klassische Symptomenverkoppelung: Eine immer hole Senkungsbeschleuninung um 100 im 2-Stundenwert und das ausdrucksvolle klinische Bild der “chronischen akuten Lumbago”. Durch operative Ausräumung des erkrankten Zwischenwirbelraums werden die bei konservativer Behandlung 5–6 Monate andauernden qualvollen akuten Symptome in 14 Tagen beseitigt und mit Frühaufstehen unter Verzicht auf jede fixierende Behandlung die Behandlungszeit durch schnelle fibröse oder knöcherne Verödung und Verlötung des Zwischenwirbelraums außerordentlich abgekürzt und weit sicherer als bei konservativer Behandlung endgültige Schmerzfreiheit und frühzeitige volle Belastbarkeit in der Arbeit erreicht.

    Notes: Abstract. Identification of pancreatic cancer in patients presenting with an enlarged pancreatic mass is a major diagnostic problem. Positron emission tomography (PET) using the radiolabeled glucose analogue 18F-fluorodeoxyglucose (FDG) has been suggested to provide excellent accuracy for noninvasive determination of suspicious pancreatic masses. We conducted a prospective study to verify these results. Forty-two patients admitted for pancreatic surgery underwent PET scanning. Image analysis was based on visual film evaluation and quantification of regional tracer uptake. PET imaging was visually analyzed by three observers blinded for the results of other diagnostic tests; they qualitatively graded the scans using a five-point scale (I = low to V = high) for the presence and intensity of focal FDG uptake. Diagnosis was proven by histology (n= 38) or follow-up (n= 4). Furthermore, the results of PET were compared with helical computed tomography (CT) and conventional ultrasonography (US), done during the routine diagnostic workup before pancreatic cancer surgery. Regarding only the results with scores of IV and V as positive for representing definite malignancy yielded a sensitivity of 71% and a specificity of 64% for film reading. Quantification of regional tracer uptake contributed no significant diagnostic advantage for differentiation between benign and malignant tumors. Helical CT revealed a sensitivity of 74% and a specificity of 45.5% and abdominal US 56% and 50%, respectively. We concluded that PET imaging provides only fair diagnostic accuracy (69%) for characterizing enlarged pancreatic masses. PET does not allow exclusion of malignant tumors. In doubtful cases, the method must be combined with other imaging modalities, such as helical CT. The results indicate that the number of invasive procedures is not significantly reduced by PET imaging.

    Description / Table of Contents: Zusammenfassung Bei serologischen Abstammungsbegutachtungen wurden 9 Familien gefunden, in denen isolierte, entgegengesetzte Phänotypen zwischen einem Elter und dem Kinde im System der sauren Erythrocytenphosphatase bestand. In einer weiteren Familie bestand keine konträre Situation. Nach quantitativer Elektrophorese im Polyacrylamidgel waren die elektrophoretischen Muster gegenüber reinerbigen Kontrollmustern deutlich abgeschwächt. Die Enzymaktivität war bei den kritischen Probanden etwa auf die Hälfte reduziert, im Vergleich zu den Werten von zwei Kontrollgruppen. Die Streuung der Einzelwerte in den Kontrollgruppen war relativ gering (Standarddeviation 10 bis 13%). Hierdurch konnte die Diagnose eines Enzymdefekts bei den entsprechenden Individuen mit forensischer Sicherheit gestellt werden — vermutet wird die Existenz des stummen Allels P0. Chromosomenanalysen sowie hämatologische Untersuchungen zeigten keine wesentlichen Veränderungen mit Ausnahme einer fraglichen „milden“ Anämie bei einigen der Probanden. Die Häufigkeit von P0 wird auf ca. 1–2mal unter 2000 Individuen geschätzt, es wird jedoch vermutet, daß in entsprechenden forensischen Fällen die Häufigkeit wesentlich höher ist — z. B. ca. 2–6%.
    Notes: Summary Serological paternity tests covering 15–20 different markers of the blood, revealed singular exclusions of the mother or of the disputed father in 9 families, exclusively due to apparently opposite homozygosity in the red cell acid phosphatase types. Another family showed identical phenotypes of the disputed father and the child. The electrophoretic patterns obtained by quantitative electrophoresis in polyacrylamide showed remarkably weaker isozyme intensities than simultaneous controls. The enzymatic activity was reduced to 50% of the normal values, when the “critical” individuals were compared with appropriate controls. The variation of the enzyme activities of the control groups was rather low, the standard deviation ranging from 10–13%, thus allowing the reliable detection of enzyme deficiency in the excluded individuals, leading to the presumed existence of the silent allele P0. Chromosome analysis and some haematological investigations performed in only some of the individuals showed no particular irregularities except for tentative suspicion of very mild anaemia in some of the propositi. The incidence of P0 is roughly estimated to range from 1–2 in 2000 individuals, but it is suspected to be much higher in relevant forensic cases, e.g. 2–6%.

    Notes: Summary In an experimental study in dogs, four different techniques of microsurgical anastomosis using Vicryl 8-0, have been compared: 1. Interrupted submucosal sutures; 2. Continuous submucosal sutures; 3. Full thickness interrupted sutures; 4. Full thickness continuous sutures. Intravenous urography and electron microscopic studies three months post operatively showed that ureteric stricture had occurred in only 2 cases out of 24. Microsurgical techniques are recommended for ureteric anastomoses.

    Notes: Summary High concentrations of polyamines have been found in the normal human prostate. The profile of these amines appeared significantly changes in benign hypertrophy of the prostate. An increase of spermine and a fall of putrescine were always found in patients with a hypertrophied prostate weighing more than 30 g. Alterations of plasmin in these tissues seemed to reflect changes in the matrix; abnormalities of thyroid and pancreatic function documented by changes in the serum levels of TSH and c-peptide which are thought to be further evidence of a mesenchymal-epithelial interaction in the pathogenesis of benign prostate hypertrophy.

    Notes: Abstract  Cells from the inner and outer granulosa cell layers of the ovarian follicles differ in function, probably because of their different origins from the surface epithelium and from the rete. This suggestion has not so far been thoroughly investigated in the human ovary. We examined fetal ovaries from the early, middle and late gestational periods, ovaries from fertile women, and preovulatory follicular cells obtained from patients under in vitro fertilization therapy (IVF). Indirect immunohistology and immunocytology were used to detect the presence of cytokeratin (CK)-positive epithelial cells. In fetal ovaries from the early gestational period, prominent rete tubules (sometimes with oocytes) appeared to be fused with the sex cords and primordial follicles. Both showed CK-positively, detected with the pan-CK antibody Lu-5. Cytokeratin 19 was clearly expressed in the fusion area. In the fetal and adult ovaries, CK-positive follicular or granulosa cells were noted in the primordial and primary follicles as well as the preovulatory follicles. Cytokeratin was not detected in the granulosa cells of growing follicles, CK-positive and -negative luteal cells were identified in the developing corpus luteum. We conclude for the human ovary: (1) the heterogeneous morphology of granulosa cells may be explained by their twofold origin from the surface epithelium and the rete, (2) the rete tubules appear to be involved in folliculogenesis, (3) the transient absence of CK expression in growing follicles compared to resting and mature follicles or to the developing corpus luteum indicates a particular role of CK-positive cells at the periovulatory period.

    Description / Table of Contents: Zusammenfassung Streptomyces glaucescens, Stamm Tü 49=ETH 22794, produziert neben Hydroxystreptomycin die Tetracenomycine, ein Gemisch lipophiler Antibiotica, von denen das mengenmäßig vorherrschende und zugleich das aktivste das Tetracenomycin C ist. Das blaßgelbe, kristallisierte Tetracenomycin C hat die Summenformel C23H20O11 und ähnelt chemisch den Tetracyclinen und den Anthracyclinonen. Tetracenomycin C ist wirksam gegen einige Grampositive Bakterien und vor allem gegen Streptomyceten; Gram-negative Bakterien werden nicht gehemmt. Aufgrund der Unterschiede bei den funktionellen Gruppen, des Wirkungsspektrums und den bislang bekannten Daten zur Wirkungsweise kann Tetracenomycin C weder zu den Tetracyclinen noch zu den Anthracyclinonen gerechnet werden. Der Erfolg der Suche nach neuen Antibiotica hängt in starkem Maße von dem verwendeten Screening-Modell ab. Ein erfolgversprechendes Modell sollte unter anderen zwei Bedingungen erfüllen: Erstens sollen ungewöhnliche, bisher nicht erfaßte Aktivitäten sichtbar werden und zweitens sollen bekannte Substanzen weitgehend ausgeschlossen werden. Ein Screening-Modell, das solche Ansprüche erfüllt, ist die Suche nach Engspektrum-Antibiotica (Zähner, 1974, 1977), die nur wenige Testkeime hemmen. Wie bei einigen anderen “nichtklassischen” Screening-Methoden wird hier auf eine Korrelation zwischen erstem Test und späterer Anwendung verzichtet; der erste Test dient nur als Leitschiene für die Auffindung und Isolierung eines neuen Metaboliten. Erst auf einer zweiten Stufe wird die Frage der Einsatzbarkeit des neuen Metaboliten geprüft. Neuartige Screeningmethoden werden erst seit relativ kurzer Zeit eingesetzt. Es schien uns deshalb lohnend, auch ältere, gut charakterisierte Antibioticaproduzenten auf die Bildung bisher übersehener Sekundärmetaboliten anzusehen. Streptomyces glaucescens (Stamm Tü 49=ETH 22794) ist schon seit längerer Zeit als Produzent von Hydroxy-Streptomycin bekannt (Hütter, 1967). Das Hauptinteresse an diesem Stamm gilt jedoch seiner Genetik, insbesondere den genetischen Grundlagen der Melaninbildung (Lerch u. Ettlinger, 1972; Baumann et al., 1974; Baumann u. Kocher, 1976). Bei der Prüfung dieses Stammes auf Bildung von Engspektrum-Antibiotica fanden wir die Tetracenomycine, ein Gemisch lipophiler Antibiotica. Hauptkomponente ist Tetracenomycin C, das nachstehend beschrieben werden soll.
    Notes: Abstract Streptomyces glaucescens, strain Tü 49=ETH 22794, produces hydroxystreptomycin as well as the tetracenomycins, a mixture of several lipophilic antibiotics. The main component and the most active one is tetracenomycin C. Tetracenomycin C has a molecular formula C23H20O11 and is chemically related to tetracyclines and anthracyclinones. The pale yellow antibiotic is active against some gram-positive bacteria, especially against streptomycetes. Gram-negative bacteria and fungi are not inhibited. In considering the differences of biological activity and the functional groups of the molecule, tetracenomycin C is not a member of the tetracycline or anthracyclinone group of antibiotics.

Based on a general discussion of orthogonalization effects, two new one-electron orthogonalization corrections are derived to improve existing semiempirical models at the neglect of diatomic differential overlap level. The first one accounts for valence-shell orthogonalization effects on the resonance integrals, while the second one includes the dominant repulsive core–valence interactions through an effective core potential. The corrections for the resonance integrals consist of three-center terms which incorporate stereodiscriminating properties into the two-center matrix elements of the core Hamiltonian. They provide a better description of conformational properties, which is rationalized qualitatively and demonstrated through numerical calculations on small model systems. The proposed corrections are part of a new general-purpose semiempirical method which will be described elsewhere.
