    Description: In mitochondria, the sirtuin SIRT5 is an NAD+-dependent protein deacylase that controls several metabolic pathways. Although a wide range of SIRT5 targets have been identified, the overall function of SIRT5 in organismal metabolic homeostasis remains unclear. Given that SIRT5 expression is highest in the heart and that sirtuins are commonly stress-response proteins, we used an established model of pressure overload–induced heart muscle hypertrophy caused by transverse aortic constriction (TAC) to determine SIRT5's role in cardiac stress responses. Remarkably, SIRT5KO mice had reduced survival upon TAC compared with wild-type mice but exhibited no mortality when undergoing a sham control operation. The increased mortality with TAC was associated with increased pathological hypertrophy and with key abnormalities in both cardiac performance and ventricular compliance. By combining high-resolution MS-based metabolomic and proteomic analyses of cardiac tissues from wild-type and SIRT5KO mice, we found several biochemical abnormalities exacerbated in the SIRT5KO mice, including apparent decreases in fatty acid oxidation and glucose oxidation as well as an overall decrease in mitochondrial NAD+/NADH. Together, these abnormalities suggest that SIRT5 deacylates protein substrates involved in cellular oxidative metabolism to maintain mitochondrial energy production. Overall, the functional and metabolic results presented here suggest an accelerated development of cardiac dysfunction in SIRT5KO mice in response to TAC, explaining increased mortality upon cardiac stress. Our findings reveal a key role for SIRT5 in maintaining cardiac oxidative metabolism under pressure overload to ensure survival.

AtPAP2 was previously shown to localize to both chloroplasts and mitochondria and to modulate carbon metabolism in Arabidopsis. Over-expression of AtPAP2 resulted in faster growth and increased biomass in several plant species, indicating its great potential for crop improvement of phosphate use and yield. Here, we studied the localization of AtPAP2 by transient expression in tobacco leaves. The results showed AtPAP2 was localized to the plasma membrane through the secretory pathway, which is different from previous studies. We also found that AtPAP2 had a close relationship with fungal PAP2-like proteins based on phylogenetic analysis. In addition, the C-terminal transmembrane domain conserved in land plants is unique among other AtPAPs except AtPAP9, which is a close homolog of AtPAP2. Taken together, our results provide information for further study of AtPAP2 in understanding its special function in crop improvement.

    Description: Adhyperforin is a novel constituent of Hypericum perforatum L., but its antidepressant-like activity remains unclear. To explore that, several well-validated animal models of depression as well as neurotransmitter reuptake and transporter binding assays were conducted. The results showed adhyperforin could reduce the immobility time of mice in the forced swimming test and tail suspension assay, antagonize the behaviors induced by reserpine, and have no effect on locomotor activity. Furthermore, following establishment of a chronic unpredictable mild stress model, adhyperforin increased the number of crossings and rearings in rats in the open field test and increased the sucrose consumption. Finally, adhyperforin inhibited uptake of serotonin, norepinephrine, and dopamine, and displayed robust binding affinities for the serotonin and norepinephrine transporters. Overall, the current study provides the first evidence that adhyperforin is a novel, active ingredient of Hypericum perforatum L. with robust antidepressant-like activity.     Description: The urban heat island (UHI) effect resulting from rapid urbanization generally has a negative impact on urban residents. Shijiazhuang, the capital of Hebei Province in China, was selected to assess surface thermal patterns and its correlation with Land Cover Types (LCTs). This study was conducted using Landsat TM images on the mesoscale level and airborne hyperspectral thermal images on the microscale level. Land surface temperature (LST) was retrieved from four scenes of Landsat TM data in the summer days to analyze the thermal spatial patterns and intensity of surface UHI (SUHI). Surface thermal characteristics were further examined by relating LST to percentage of imperious surface area (ISA%) and four remote sensing indices (RSIs), the Normalized Difference Vegetation Index (NDVI), Universal Pattern Decomposition method (VIUPD), Normalized Difference Built-up Index (NDBI) and Biophysical Composition Index (BCI). On the other hand, fives scenes of airborne TASI (Thermal Airborne Spectrographic Imager sensor) images were utilized to describe more detailed urban thermal characteristics of the downtown of Shijiazhuang city. Our results show that an obvious surface heat island effect existed in the study area during summer days, with a SUHI intensity of 2–4 °C. The analyses reveal that ISA% can provide an additional metric for the study of SUHI, yet its association with LST is not straightforward and this should a focus in future work. It was also found that two physically based indices, VIUPD and BCI, have the potential to account for the variation in urban LST. The results concerning on TASI indicate that diversity of impervious surfaces (rooftops, concrete, and mixed asphalt) contribute most to the SUHI, among all of the land cover features. Moreover, the effect of impervious surfaces on LST is complicated, and the composition and arrangement of land cover features may play an important role in determining the magnitude and intensity of SUHI. Overall, the analysis of urban thermal signatures at two spatial scales complement each other and the use of airborne imagery data with higher spatial resolution is helpful in revealing more details for understanding urban thermal environments.

    Description: Plant phenolics can have applications in pharmaceutical and other industries. To identify and quantify the phenolic compounds in Helianthus tuberosus leaves, qualitative analysis was performed by a reversed phase high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS) and quantitative analysis by HPLC. Ten chlorogenic acids (CGAs) were identified (3-o-caffeoylquinic acid, two isomers of caffeoylquinic acid, caffeic acid, p-coumaroyl-quinic acid, feruloylquinic acid, 3,4-dicaffeoyquinic acid, 3,5-dicaffeoylquinic acid, 1,5-dicaffeoylquinic acid, and 4,5-dicaffeoylquinic acid) by comparing their retention times, UV-Vis absorption spectra, and MS/MS spectra with standards. In addition, four other phenolic compounds, including caffeoyl glucopyranose, isorhamnetin glucoside, kaempferol glucuronide, and kaempferol-3-o-glucoside, were tentatively identified in Helianthus tuberosus leaves for the first time. The 3-o-caffeoylquinic acid (7.752 mg/g DW), 4,5-dicaffeoylquinic acid (5.633 mg/g DW), and 3,5-dicaffeoylquinic acid (4.900 mg/g DW) were the major phenolic compounds in leaves of Helianthus tuberosus cultivar NanYu in maturity. The variations in phenolic concentrations and proportions in Helianthus tuberosus leaves were influenced by genotype and plant growth stage. Cultivar NanYu had the highest concentration of phenolic compounds, in particular 3-o-caffeoylquinic acid and 4,5-dicaffeoylquinic acid compared with the other genotypes (wild accession and QingYu). Considering various growth stages, the concentration of total phenolics in cultivar NanYu was higher at flowering stage (5.270 mg/g DW) than at budding and tuber swelling stages. Cultivar NanYu of Helianthus tuberosus is a potential source of natural phenolics that may play an important role in the development of pharmaceuticals.

    Description: To estimate the surface air temperature by remote sensing, the advection-energy balance for the surface air temperature (ADEBAT) model is developed which assumes the surface air temperature is driven by the local driving force and the advective driving force. The local driving force produces a local surface air temperature whereas the advective driving force changes it by adding an exotic air temperature. An advection factor is defined to measure the quantity of the exotic air brought by the advection. Since the is determined by the advection, this paper improves it to a regional scale by using the Inverse Distance Weighting (IDW) method whereas the original ADEBAT model uses a constant of for a block of area. Results retrieved by the improved ADEBAT (IADEBAT) model are evaluated and comparison was made with the in situ measurements, with an (correlation coefficient) of 0.77, an RMSE (Root Mean Square Error) of 0.31 K, and a MAE (Mean Absolute Error) of 0.24 K. The evaluation shows that the IADEBAT model has higher accuracy than the original ADEBAT model. Evaluations together with a -test of the MAD (Mean Absolute Deviation) reveal that the IADEBAT model has a significant improvement.

    Description: Quantifying vegetation fractional cover (VFC) and assessing its role in heat fluxes modeling using medium resolution remotely sensed data has received less attention than it deserves in heterogeneous urban regions. This study examined two approaches (Normalized Difference Vegetation Index (NDVI)-derived and Multiple Endmember Spectral Mixture Analysis (MESMA)-derived methods) that are commonly used to map VFC based on Landsat imagery, in modeling surface heat fluxes in urban landscape. For this purpose, two different heat flux models, Two-source energy balance (TSEB) model and Pixel Component Arranging and Comparing Algorithm (PCACA) model, were adopted for model evaluation and analysis. A comparative analysis of the NDVI-derived and MESMA-derived VFCs showed that the latter achieved more accurate estimates in complex urban regions. When the two sources of VFCs were used as inputs to both TSEB and PCACA models, MESMA-derived urban VFC produced more accurate urban heat fluxes (Bowen ratio and latent heat flux) relative to NDVI-derived urban VFC. Moreover, our study demonstrated that Landsat imagery-retrieved VFC exhibited greater uncertainty in obtaining urban heat fluxes for the TSEB model than for the PCACA model.
