Description / Table of Contents: Summary Of the distinct syndromes and associations featuring overgrowth, some still await further characterization. Major distinguishing marks between these overgrowth disorders include craniofacial abnormalities, statomotoric and mental retardation, and muscular hypotonia. We describe a 3-year-old girl with tall stature, facial dysmorphy, severe mental retardation, and an interstitial deletion of the long arm of chromosome 22/del(22)(q13.1;q13.33). The chromosome 22q13 deletion syndrome was only recently reported by Nesslinger et al. [1] as a new nosologic entity. Discussion: It is assumed that there is a growth gene at the chromosome 22q deletion.

Notes: Abstract Local intra-arterial fibrinolysis (LIF) is an effective treatment for selected patients in acute thromboembolic occlusion of the middle cerebral artery, vertebrobasilar system or ophthalmic artery. However, the risk/benefit ratio of thrombolysis in patient subgroups requires classification. Advanced age has been regarded as a prognostic factor for poor clinical outcome. We report our experience with LIF in seven patients with a mean age of 79 years (range 76–83 years) who represented thromboembolic occlusion of the basilar, middle cerebral or pericallosal arteries. We discuss why advanced age need not contraindicate LIF.

Notes: Summary In photoreaction with the pyrimidine bases (thymine, cytosine, uracil) as well as with nucleic acids (DNA, RNA) a C4-cycloaddition of furocoumarins to the 5.6-double bond of pyrimidine bases takes place. The simple photoadduct furocoumarin-pyrimidine base can be split by reirradiation at wavelengths shorter than 334 nm. Reactivation of bacterial cells photodamaged by psoralen (365 nm) was tried experimentally. However, reirradiation at shorter wavelengths and with visible light of the psoralen-inactivated bacterial cells was without any effect. The inability of the shorter wavelengths to repair this photodamage was probably due to a filter effect of DNA for such wavelengths, as shown by experiments on a DNA-psoralen combination. On the other hand the observed ability of psoralen to form inter-strand cross-linkages in the photoreaction with DNA may be significant for explaining the absence of photoreactivation when the inactivated bacterial cells are irradiated with visible light.

The oxidation of the new compounds with Br2 at -78°C, in dichloromethane, leads to the corresponding complex salts of pentavalent antimony (R4E)2[Br3Cl2Sb]2CH2.

Description: Mast cell (MC) activation through the high-affinity IgE receptor FcRI leads to the release of mediators involved in immediate-type allergic reactions. Although Abs against the tetraspanins CD63 and CD81 inhibit FcRI-induced MC degranulation, the intrinsic role of these molecules in FcRI-induced MC activation is unknown. In MCs, CD63 is expressed at the cell surface and in lysosomes (particularly secretory lysosomes that contain allergic mediators). In this study, we investigated the role of CD63 in MC using a CD63 knockout mouse model. CD63-deficiency did not affect in vivo MC numbers and tissue distribution. Bone marrow–derived MC developed normally in the absence of CD63 protein. However, CD63-deficient bone marrow–derived MC showed a significant decrease in FcRI-mediated degranulation, but not PMA/ionomycin-induced degranulation, as shown by β-hexosaminidase release assays. The secretion of TNF-α, which is both released from granules and synthesized de novo upon MC activation, was also decreased. IL-6 secretion and production of the lipid mediator leukotriene C 4 were unaffected. There were no ultrastructural differences in granule content and morphology, late endosomal/lysosomal marker expression, FcRI-induced global tyrosine phosphorylation, and Akt phosphorylation. Finally, local reconstitution in genetically MC-deficient Kit w/w-v mice was unaffected by the absence of CD63. However, the sites reconstituted with CD63-deficient MC developed significantly attenuated cutaneous anaphylactic reactions. These findings demonstrate that the absence of CD63 results in a significant decrease of MC degranulation, which translates into a reduction of acute allergic reactions in vivo, thus identifying CD63 as an important component of allergic inflammation.
