    Notes: Summary Fractionation of the cat cerebellum has revealed that the parallel fibers of the cerebellar cortex break into two types of fragments which can be recognized in thin sections and in suspensions of negatively stained material. Fragmented material correlates well with electron micrographs of parallel fibers in situ. Parallel fiber axons and their bulbous presynaptic expansions sediment mainly into two subfractions. One unique layer consists of the intersynaptic tubular pieces of parallel fiber axons. Another layer is enriched in the synaptic portions which make contact with Purkinje spines. The possible contaminants of these fractions are discussed. These fractions may permit analysis of the chemical organization of conducting vis à vis synaptic membranes from a known anatomical source. This material was presented as a preliminary report at the Sixth Annual Meeting of the American Society for Cell Biology, November 1966, and is contained in J. Cell. Biol. 31, 52A, 1966.

    Notes: Summary Subfractionation of a standard “synaptosome” fraction from whole brain has yielded two lighter layers rich in axonal segments devoid of synaptosomes similar to those obtained from cerebellar homogenates. The heavier layers were found to contain both synaptosomes and axonal segments plus a significant population of synaptosomes with axons attached. Quantitative studies of the percent perimeter of external membrane contributed by the various elements within the subfractions showed that the major constituents in decreasing order were axonal segments, synaptosomes, membranes of undetermined origin and free mitochondria. These findings have revealed that “synaptosome” preparations are more heterogeneous than previously reported and at the same time have indicated the possibility of future studies on axonal preparations from whole brain. Correlated biochemical data strongly suggest chemical differences in fractions rich in axonal segments in contrast to those enriched in synaptosomes.

    Notes: Summary Three distinct phospholipid patterns occurred in the mitochondrial subcellular fractions: a) myelin, b) axonal-synaptosomal and c) mitochondrial. Axonal and synaptosomal fractions were found to be very similar in phospholipid and cholesterol composition. In all submitochondrial fractions cholesterol, phosphatidyl serine and sphingomyelin occurred in the relatively constant molar ratio of 1.0∶0.18∶0.11. The phospholipid patterns reported in this paper correspond well to others published for synaptosomes and “synaptic membranes” from mammalian sources as well as axonal membranes from invertebrate sources. However, morphological analysis has indicated that axonal as well as synaptosomal fractions contained significant amounts of microsomes and abundant membranes of undetermined origin which must also be considered when attempting morphological-biochemical correlations. Attention was given to the possible functional and chemical similarity of axonal and synaptosomal membranes other than that portion presumably specialized for chemical transmission, and to the difficulty of establishing the lipid pattern of the “true synaptic membrane” in presently available preparations.

    Notes: Summary Mild homogenization of the cat cerebellum and centrifugation of the resultant particulate matter yielded a crude mitochondrial fraction which was further fractionated by sucrose-density gradient centrifugation. The fractions obtained were examined under the electron microscope. Fraction P2A contained large numbers of myelin fragments. Fraction P2B was enriched in segments of unmyelinated axons. Fraction P2C was enriched in axons with their synaptic enlargements. Fractions P2D and P2E were composed predominately of free mitochondria, although axonal segments with bulbar enlargements corresponding to synaptosomes were also seen. Fractions enriched in axons were characterized by a high concentration of gangliosides and acetylcholinesterase (per mg protein). Fractions that contain axons with synaptic enlargements were similarly enriched in these constituents. Glycoprotein-NANA was rather widely distributed, but the concentration of glycoprotein-NANA, per mg protein, was somewhat higher in those fractions that contain axons with attached synaptic enlargements. The distribution of gangliosides and glycoproteins differed so that the ratio of glycoprotein-NANA to ganglioside-NANA increased with increasing particle size or density.

    Notes: Summary Previous studies have indicated that fractions of unmyelinated axons can be obtained from cerebellum, whole brain or forebrain. It seemed likely that unmyelinated axons break into segments upon homogenization and that any circumscribed region of brain containing abundant unmyelinated axons could be found to yield axonal fractions. Caudate-putamen material was used to test this hypothesis. Fractions analogous to cerebellum were obtained from sucrose gradients and were found to yield abundant axonal profiles which could be correlated with the appearance of unmyelinated axons after sucrose immersion in situ. We conclude that fractions of unmyelinated axons can likely be obtained from any anatomical region rich in them and can be used upon purification for obtaining chemical data from different axonal types.

    Notes: Abstract We describe the brood duration and marsupial development of three temperate coastal mysid species, Anisomysis mixta australis (Zimmer), Tenagomysis tasmaniae Fenton and Paramesopodopsis rufa Fenton, found commonly in Tasmanian waters. Larvae cultured in vitro had brood durations at 13 °C (17 °C) of 22 (15), 23 (15), and 28 (20) d, respectively. Development through seven larval stages, and brood durations for these three species are similar to those reported for coastal mysids from other temperate areas throughout the world.

    Notes: Abstract A novel schedule of 5-fluorouracil administration has been developed for biochemical modulation studies. In combination with the pyrimidine synthesis inhibitor PALA, 5-fluorouracil has been given as a 24-hour infusion, repeated weekly: a dose of 2600 mg/m2 is well tolerated. To identify a suitable dose of 5-fluorouracil as a single agent on this schedule, we treated 26 patients at doses ranging from 2800 to 3400 mg/m2 per week. Two-thirds of the patients had failed previous therapy, and most were symptomatic from their disease. Over half of the patients had metastatic colorectal cancer. The dose-limiting toxicity was diarrhea: Grade 3 or 4 toxicity occurred at every level tested. Twenty-two of the 26 patients required therapy interruption because of toxicity. The severity of this toxicity indicated that escalation of 5-fluorouracil on this schedule beyond the 2600 mg/m2 known to be tolerated in the PALA-containing regimen, would be impractical. Two patients, both with previously untreated colorectal cancer, had partial remissions lasting three and five months respectively. This dose-intense schedule of 5-fluorouracil administration will be explored further in large-scale randomized trials.

    Notes: As part of an ongoing program to prepare and evaluate thermally-stable, processable materials, 18 aromatic, silicon containing polyamides (structures shown below) have been synthesized via interfacial polymerization of six silicon-containing diacid chlorides with 3,3′-diaminodiphenyl-nethane, 3,3′-diaminobenzophenone, and l-(3′-amino-benzy)-4-(3″aminobenzoyl)benzene. All polyamides were soluble in m-cresol and N,N-dimethylacetamide and had glass transition temperatures between 178 and 254°C, Thermogravimetric analyses conducted in static air on film specimens were-1 used to show 5 and 10 percent weight losses between 331-400°C and 354-440°C, respectively. 3,3′- and 4,4′-silane isomersR = R′ = Me; R = Me; R′ = Ph; R = R′ = Ph
