    Abstract: A variety of industries employ synthetic azo dyes. However, the biosphere is being damaged by the unused/leftover azo dyes, which pose a danger to all living things. Therefore, treating them to shield the environment from the potential harm of azo dyes is crucial. Bio-sorption is a cheap and effective mode for eliminating toxic dyes in the environment. The current work focused on synthesizing magnesium oxide (MgO) nanoparticles using an aqueous leaf extract of neem ( Azadirachta indica ). The XRD and SEM analyses of MgO nanoparticles indicated the crystalline nature of MgO nanoparticles with a cubic structure, and the size was around 90–100 nm. FTIR analysis showed the presence of a stretching frequency peak at 550 cm −1 , confirming the Mg–O bond. The surface analysis revealed the cluster form of the synthesized nanoparticles. The UV–visible absorption peak for MgO nanoparticles was found at 294 nm and band gap of 4.52 eV. In order to eliminate the Reactive Red 195 dye, MgO nanoparticles were used. At pH 4, 40 °C, 0.02% dye concentration, and 0.003 g/L catalyst amount, the highest degree of decolorization (91%) was seen. Decreased total organic carbon (TOC) and the chemical oxygen demand (COD) percent were 84.33% and 81.3%, respectively. The proposed mechanism of target dye degradation was also investigated. MgO NPs were found to be effective in their catalytic behavior toward the degradation of Reactive Red 195 dye up to five cycles with almost no change in their catalytic activity.

    Abstract: The regulated production of filamentous protein complexes is essential in many biological processes and provides a new paradigm in signal transmission. The mitochondrial antiviral signaling protein (MAVS) is a critical signaling hub in innate immunity that is activated when a receptor induces a shift in the globular caspase activation and recruitment domain of MAVS into helical superstructures (filaments). It is of interest whether adaptive evolution affects the proteins involved in innate immunity. Here, we explore and confer the role of selection and diversification on mitochondrial antiviral signaling protein in mammalian species. We obtined the MAVS proteins of mammalian species and examined their differences in evolutionary patterns. We discovered evidence for these proteins being subjected to substantial positive selection. We demonstrate that immune system proteins, particularly those encoding recognition proteins, develop under positive selection using codon-based probability methods. Positively chosen regions within recognition proteins cluster in domains involved in microorganism recognition, implying that molecular interactions between hosts and pathogens may promote adaptive evolution in the mammalian immune systems. These significant variations in MAVS development in mammalian species highlights the involvement of MAVS in innate immunity. Our findings highlight the significance of accounting for how non-synonymous alterations affect structure and function when employing sequence-level studies to determine and quantify positive selection.

    Abstract: Mushrooms have been accepted as nutraceutical foods because of their high nutritional and functional values. They have also gained interest due to their medicinal properties, economic importance, and organoleptic merit. In this study, wild Ganoderma lucidum and four commercial mushrooms, that is, Pleurotus ostreatus, Volvariella volvacea, Hericium erinaceus, and Lentinus edodes from Pakistan were screened for their biological activities such as anticancer, antityrosinase, anti‐α‐glucosidase, and antithrombotic activities from their methanol, ethanol, and water extracts. Enzyme inhibition assay showed that selected mushrooms are potent inhibitors with %age inhibition ranging from 19.00% to 80.91%, and 32.85% to 83.38% for tyrosinase and α‐glucosidase, respectively. The best tyrosinase inhibition was shown by P. ostreatus whereas L. edodes was found best as α‐glucosidase inhibitor. These mushrooms were tested against cancer cell lines including HT ‐29 colon and H‐1299 lungs carcinoma cell lines. G. lucidum showed 29% and 24% viability of cells against HT ‐29 and H‐1299 cell lines, respectively. This antiproliferative effect was in dose‐dependent manner, and the maximum inhibition was observed at 200 μg/ ml . Mushrooms extracts were also found effective against clot lysis. The percentage of clot lysis was in the range of 27%–29%. The research would provide knowledge to the people of Pakistan about the importance of locally available commercial mushrooms and wild mushrooms for health improvement and prevention against different kinds of diseases.

    Abstract: Acupoint application has been used in China to treat various illnesses for ages. In cough variant asthma (CVA), the main clinical sign is episodic night cough. Acupoint application therapy of traditional Chinese medicine is an effective procedure to treat cough variant asthma. The current study is designed to systematically assess the effectiveness of acupoint application therapy in traditional medicine for patients with cough variant asthma. The comprehensive computer retrieval related to comparison between acupoint application and nonacupoint application therapy for cough variant asthma was carried out in various databases ( n = 8 ) from database establishment until July 4, 2021. Both English and Chinese articles about original investigations in humans were searched. Two independent authors extracted the data, and disagreements were resolved by discussion. ReviewManager 5.3 software provided by Cochrane did a meta-analysis of selected randomized controlled trials (RCTs). Quality of experimentation and risk bias were analyzed by the Cochrane Handbook tool. A total of thirteen randomized controlled clinical articles along with 1237 patients were included in the study. Findings of meta-analysis showed that compared with nonacupoint application treatment, the total effective rate of acupoint application treatment is more effective ( RD = 0.13 , 95% CI (0.09, 0.17), Z = 6.70 , P 〈 0.00001 ). Besides, acupoint application can improve patients’ lung function, the lung function index FVC ( mean   difference = 0.55 , 95% confidence interval (0.42, 0.68), Z = 8.40 , P 〈 0.00001 ), FEV1 ( MD = 0.35 , 95% CI (0.23, 0.47), Z = 5.86 , P 〈 0.00001 ), FEV1/FVC (%) ( MD = 12.68 , 95% CI (4.32, 21.03), Z = 2.97 , P = 0.003 ), FEV1 (%) ( MD = 8.63 , 95% CI (8.01, 9.25), Z = 27.44 , P 〈 0.00001 ), and PEF (day) ( MD = 0.62 , 95% CI (0.52, 0.71), Z = 12.40 , P 〈 0.00001 ) of patients treated by acupoint application therapy were increased. Moreover, acupoint application might lower the level of immunoglobulin E ( MD = − 54.58 , 95% CI (−63.54, −45.61), Z = 11.93 , P 〈 0.00001 ) and EOS ( MD = − 0.21 , 95% CI (−0.35, −0.06), Z = 2.77 , P = 0.006 ). The LCQ (Leicester cough questionnaire) total score of CVA patients was also increased ( MD = 2.30 , 95% CI (1.55, 3.06), Z = 5.98 , P 〈 0.00001 ). Acupoint application therapy is effective in controlling symptoms of CVA. It also has a positive effect in improving lung function and life quality of patients. It can reduce the eosinophil levels and peripheral blood IgE levels of patients as well.

    Abstract: Positive selection refers to the process by which certain genetic variations are more likely to be passed on to future generations because they confer some advantage in terms of survival or reproduction. Zinc finger proteins are a type of transcription factor that plays a role in regulating gene expression, particularly in undifferentiated stem cells. Suppose it has been found that certain zinc finger proteins show genetic signatures of positive selection in mammals. In that case, it suggests that these proteins may have played a role in adaptive evolution in undifferentiated stem cells. This could mean that the specific genetic changes in these zinc finger proteins gave an advantage to the organisms that possessed them, helping them survive and reproduce more effectively. These genetic changes may have allowed the organisms to adapt to changing environments or to develop new abilities, such as increased resistance to disease or faster growth. Undifferentiated stem cells that underwent adaptive evolution during the evolution of mammals can be identified genetically by the outcomes of positive selection on zinc finger proteins. Because of selection pressures like environmental shifts or the introduction of novel pathogens, it is plausible that some zinc finger proteins have experienced fast evolution. The emergence of novel activities or higher expression levels of these proteins as a result of this quick evolution may have given the creatures that possessed them a survival edge. Another possible outcome of positive selection in zinc finger proteins is the emergence of new genetic variations that allow for increased diversity and plasticity in stem cells. This increased diversity and plasticity could have allowed for more efficient adaptation to changing environments and could have played a role in the evolution of new organisms or new characteristics in existing organisms. Overall, the results of positive selection in zinc finger proteins can provide insight into how adaptive evolution occurred in undifferentiated stem cells during the evolution of mammals and how this evolution may have contributed to the development of new organisms and new characteristics and adaptations to changing environments.

    Abstract: Background. Antibiotic supplementation in feed has been continued for the previous 60 years as therapeutic use. They can improve the growth performance and feed efficiency in the chicken flock. A favorable production scenario could favor intestinal microbiota interacting with antibiotic growth promoters and alter the gut bacterial composition. Antibiotic growth promoters did not show any beneficial effect on intestinal microbes. Scope and Approach. Suitable and direct influence of growth promoters are owed to antimicrobial activities that reduce the conflict between host and intestinal microbes. Unnecessary use of antibiotics leads to resistance in microbes, and moreover, the genes can relocate to microbes including Campylobacter and Salmonella, resulting in a great risk of food poisoning. Key Findings and Conclusions. This is a reason to find alternative dietary supplements that can facilitate production, growth performance, favorable pH, and modulate gut microbial function. Therefore, this review focus on different nutritional components and immune genes used in the poultry industry to replace antibiotics, their influence on the intestinal microbiota, and how to facilitate intestinal immunity to overcome antibiotic resistance in chicken.

    Abstract: Cytochrome (CYP) enzymes catalyze the metabolic reactions of endogenous and exogenous compounds. The superfamily of enzymes is found across many organisms, regardless of type, except for plants. Information was gathered about CYP2D enzymes through protein sequences of humans and other organisms. The secondary structure was predicted using the SOPMA. The structural and functional study of human CYP2D was conducted using ProtParam, SOPMA, Predotar 1.03, SignalP, TMHMM 2.0, and ExPASy. Most animals shared five central motifs according to motif analysis results. The tertiary structure of human CYP2D, as well as other animal species, was predicted by Phyre2. Human CYP2D proteins are heavily conserved across organisms, according to the findings. This indicates that they are descended from a single ancestor. They calculate the ratio of alpha-helices to extended strands to beta sheets to random coils. Most of the enzymes are alpha-helix, but small amounts of the random coil were also found. The data were obtained to provide us with a better understanding of mammalian proteins’ functions and evolutionary relationships.
