    Description: Glycerol monoalkyl glycerol tetraethers (GMGTs) are a group of membrane spanning lipids produced by some species of archaea and bacteria. They differ from the more commonly studied glycerol dialkyl glycerol tetraethers (GDGTs) in having an additional covalent carbon-carbon bond connecting the two alkyl chain. The relative abundance and distribution of bacterial branched GMGTs (brGMGTs) in surface sediments from a set of East African lakes were studied. The abundance of brGMGTs relative to the brGDGTs is positively correlated to measured mean annual air temperature (MAAT), although with a significant amount of scatter. BrGMGT abundance was not correlated to lake water pH. Seven major brGMGTs that vary in degree of methylation were identified, with m/z 1020, 1034 and 1048. Further, the mass chromatograms of the m/z 1020 and 1034 brGMGTs show an interesting distribution of peaks, which likely relates to the occurrence of distinct brGMGT isomers. This structural complexity is higher than previously observed in peats and marine sediments. Principal component analysis of the fractional abundance of bacterial tetraether lipids revealed the brGMGTs behave similarly to one another but differently from both the 5- or 6-methyl brGDGTs. This suggests the brGMGTs are produced by a common source organism and are methylated at a different position. The distribution of the seven brGMGTs showed considerable correlation with MAAT. This variability was captured in a new proxy index (the brGMGTI), which showed a strong positive linear relationship with MAAT. Lacustrine brGMGTs show potential to be applied to ancient settings to provide information about paleoclimate.

    Description: We investigated the occurrence and activity of anaerobic ammonia oxidation (anammox) bacteria in sandy and muddy sand sediments of the southern North Sea. The presence of anammox bacteria was established through the detection of specific phosphocholine-monoether ladderane lipids, 16S rRNA gene, and hydrazine synthase (hzsA) genes. Anammox activity was measured in intact sediment cores (in situ rate) and in sediment slurries (potential rate) as the rate of N2 evolution from 15N-labeled substrates and compared to the transcriptional activity of genes of anammox bacteria. The contribution of anammox to N2 production ranged between 0% and 29%. The potential rate of anammox agreed well with the abundance of anammox bacteria 16S rRNA and hzsA gene copies and the transcriptional activity of the anammox bacteria 16S rRNA gene. We found a higher abundance and activity of anammox bacteria in sediments with higher organic carbon content and also higher activity in summer than in winter. The abundance of anammox bacteria and their potential anammox rates were similar to those reported for other marine coastal sediments, suggesting that potentially they are important contributors to the nitrogen cycle in sandy sediments of shallow continental shelf areas.

    Description: Biomass burning on the African continent is widespread and interactions with climate, vegetation dynamics and biogeochemical cycling are complex. To obtain a better understanding of these complex relationships, African fire history has been widely studied, although mostly on relatively short time-scales (i.e. yrs to kyrs) and less commonly on long-term scales. Here, we present a 192-kyr, continuous biomass-burning record from sub-Saharan Northwest Africa based on the fire biomarker levoglucosan in a marine sediment core offshore Guinea. Notable features of our record include an increase in levoglucosan accumulation at 80 ka and two peaks at 50-60 ka. The event at 80 ka is likely related to an overall increase in sedimentation rates rather than an increase in biomass burning in the Northwest African savanna region. Our record indicates that glacial/interglacial changes in regional climate and vegetation composition (C3 vs. C4 plants) were not a major influence on biomass burning over the last 192 kyrs. However, we suggest that the burning events at 50-60 ka might be caused by increased occurrence of C3 vegetation and human settlement in this region. At this time, the savanna region became wetter and fuel loads likely increased. Therefore, the region was more hospitable for humans, who likely used fire for hunting activities. Collectively, we hypothesize that on longer (glacial/interglacial) timescales, biomass burning, regional climate and African vegetation are not necessarily coupled, while around 50-60 ka, higher fuel loads and human fire-use may have influenced fire occurrence in sub-Saharan Northwest Africa.

    Description: Lipid extraction of biomass prior to stable isotope analysis is known to cause variable changes in the stable nitrogen isotopic composition (d15N) of residual biomass. However, the underlying factors causing these changes are not yet clear. Here we address this issue by comparing the d15N of bulk and residual biomass of several marine animal tissues (fish, crab, cockle, oyster, and polychaete), as well as the d15N of the extracted lipids. As observed previously, lipid extraction led to a variable offset in d15N of biomass (differences ranging from -2.3 to +1.8 per mil). Importantly, the total lipid extract (TLE) was highly depleted in 15N compared to bulk biomass, and also highly variable (differences ranging from -14 to +0.7 per mil). The TLE consisted mainly of phosphatidylcholines, a group of lipids with one nitrogen atom in the headgroup. To elucidate the cause for the 15N-depletion in the TLE, the d15N of amino acids was determined, including serine because it is one of the main sources of nitrogen to N-containing lipids. Serine d15N values differed by -7 to +2 per mil from bulk biomass d15N, and correlated well with the 15N depletion in TLEs. On average, serine was less depleted (-3 per mil) than the TLE (-7 per mil), possibly due to fractionation during biosynthesis of N-containing headgroups, or that other nitrogen-containing compounds, such as urea and choline, or recycled nitrogen contribute to the nitrogen isotopic composition of the TLE. The depletion in 15N of the TLE relative to biomass increased with the trophic level of the organisms.
