    Notes: We have investigated the Ca2+-dependent interactions of villin, a protein of the intestinal microvillar core, with actin by monitoring resonance energy tranfer between fluorescently labeled actin subunits. In the presence of elevated free Ca2+(∼20 μM), villin affects both the nucleation and the elongation phases of actin polymerization. Consistent with previous reports, villin stimulates the nucleation process and will form stable nuclei under depolymerization conditions. Compared to the control, the net rate of polymerization is slightly inhibited at low con-centrations of villin (villin/actin ∼ 1:400) but is stimulated at higher concentrations (villin/actin 〉 1:100). Villin also significantly increases the critical concentration of actin polymerization. Addition of either villin or villin-actin complexes induces depolymerization of preassembled actin filaments. This villin-induced depolymerization is reversible upon removal of free Ca2+ or upon the addition of phalloidin. The exchange of actin subunits at steady state is inhibited at low concentrations of villin (villin/actin ∼ 1:200) but is stimulated at higher concentrations (villin/actin ∼ 1:50). None of the above effects is observed at 〈 10-8 M free [Ca2+].

    Notes: The 21 S complex of enzymes for DNA synthesis in the combined low salt nuclear extract-post microsomal supernatant from HeLa cells [Malkas et al. (1990) Biochemistry 29:6362-6374] was purified by poly (ethylene glycol) precipitation, Q-Sepharose chromatography, Mono Q Fast Protein Liquid Chromatography (FPLC), and velocity gradient centrifugation. The procedure gives purified enzyme complex at a yield of 45%. The 21 S enzyme complex remains intact and functional in the replication of simian virus 40 DNA throughout the purification. Sedimentation analysis showed that the 21 S enzyme complex exists in the crude HeLa cell extract and that simian virus 40 in vitro DNA replication activity in the cell extract resides exclusively with the 21 S complex. The results of enzyme and immunological analysis indicate that DNA polymerase α-primase, a 3′,5′ exonuclease, DNA ligase I, RNase H, and topoisomerase I are associated with the purified enzyme complex. Denaturing polyacrylamide gel electrophoresis of the purified enzyme complex showed the presence of about 30 polypeptides in the size range of 300 to 15 kDa. Immunofluorescent imaging analysis, with antibodies to DNA polymerase α,β and DNA ligase I, showed that polymerase α and DNA ligase I are localized to granular-like foci within the nucleus during S-phase. In contrast, DNA polymerase β, which is not associated with the 21 S complex, is diffusely distributed throughout the nucleoplasm.

    Description: This study describes the first general optimization model for complex systems with uncertain parameters and decision variables represented as intervals in CDM forestry projects. We work through a specific example of the optimization method developed for a Clean Development Mechanism (CDM) forestry project in Inner Mongolia, China. This model is designed to optimize the carbon sink capacity of the new forests, and can deal with uncertainties in the carbon sink capacity, average annual rainfall, ecological parameters, and biological characteristics of tree species. The uncertain inputs are presented in the form of intervals, as are several of the optimized output variables. Compared with the project’s originally recommended scheme, the optimized model will absorb and fix between 1,142 and 885,762 tonnes of extra carbon dioxide. Moreover, the ecological and environmental benefits of the project are also raised to various extents.

    Description: Serials of Ga doping on Sn sites as heterovalent substitution in Cu2CdSnSe4 are prepared by the melting method and the spark plasma sintering (SPS) technique to form Cu2CdSn1-xGaxSe4 (x = 0, 0.025, 0.05, 0.075, 0.01, and 0.125). Massive atomic vacancies are found at x = 0.10 by the heterovalent substitution, which contributes significantly to the increase of electrical conductivity and the decrease of lattice thermal conductivity. The electrical conductivity is increased by about ten times at 300 K after Ga doping. Moreover, the seebeck coefficient only decreases slightly from 310 to 226 μV/K at 723 K, and a significant increase of the power factor is obtained. As a result, a maxium value of 0.27 for the figure of merit (ZT) is obtained at x = 0.10 and at 723 K. Through an ab initio study of the Ga doping effect, we find that the Fermi level of Cu2CdSnSe4 is shifted downward to the valence band, thus improving the hole concentration and enhancing the electrical conductivity at low doping levels. Our experimental and theoretical studies show that a moderate Ga doping on Sn sites is an effective method to improve the thermoelectric performance of Cu2CdSnSe4.     Description: Increased calcium influx secondary to glutamate induced excitotoxicity initiates and potentiates devastating pathological changes following ischemic stroke. Pertussis toxin (PTx), a G-protein blocker, is known to suppress intracellular calcium accumulation. We hypothesize that PTx can protect against stroke by blocking calcium influx. In a permanent middle cerebral artery occlusion model, PTx (1000 ng) was given intraperitoneally 30 minutes after inducing stroke. Magnetic Resonance Imaging of perfusion and T2-weighted brain scans were obtained to evaluate cerebral blood flow (CBF) and infarct volume. Primary neuronal culture was used to test glutamate induced excitotoxicity and calcium influx. We established a nonlinear exponential curve model to minimize variations in animal cerebrovasculature. A reduction of 40%-60% in relative CBF was a critical window where infarct volume started to increase as rCBF reduced. PTx showed maximal effects in reducing infarct volume at this window. In vitro studies further demonstrated PTx increased neuronal cell survival by decreasing glutamate-induced calcium influx into neurons and preventing neurons from apoptosis. PTx salvages the ischemic penumbra by blocking calcium influx. This provides us a new mechanism upon which experimental therapies can be explored to treat ischemic stroke.     Description: During past decades, many automated software faults diagnosis techniques including Spectrum-Based Fault Localization (SBFL) have been proposed to improve the efficiency of software debugging activity. In the field of SBFL, suspiciousness calculation is closely related to the number of failed and passed test cases. Studies have shown that the ratio of the number of failed and passed test case has more significant impact on the accuracy of SBFL than the total number of test cases, and a balanced test suite is more beneficial to improving the accuracy of SBFL. Based on theoretical analysis, we proposed an PNF (Passed test cases, Not execute Faulty statement) strategy to reduce test suite and build up a more balanced one for SBFL, which can be used in regression testing. We evaluated the strategy making experiments using the Siemens program and Space program. Experiments indicated that our PNF strategy can be used to construct a new test suite effectively. Compared with the original test suite, the new one has smaller size (average 90% test case was reduced in experiments) and more balanced ratio of failed test cases to passed test cases, while it has the same statement coverage and fault localization accuracy.

    Description: Water, Vol. 9, Pages 573: An Optimization Model for Waste Load Allocation under Water Carrying Capacity Improvement Management, A Case Study of the Yitong River, Northeast China Water doi: 10.3390/w9080573 Authors: Chong Meng Xiaolei Wang Yu Li In this study, a two-stage stochastic programming (TSP) model was developed for supporting regional waste load(chemical oxygen demand (COD)and NH3-N) allocation in four main pollution departments (industry, municipal, livestock breeding, and agriculture), constrained by the water carrying capacity, which can be improved by ecological restoration engineering, in the nine function zones of the Yitong River. A number of scenarios corresponding to different stream flow levels were examined. The results revealed that the carrying capacity of COD and NH3-N has a similar tendency with a positive correlation to stream flow levels. The allocation amount of each pollutant for the four departments was obtained differently in each zone, and ecological restoration engineering solutions were obtained for different zones to improve the carrying capacity of the pollutants in order to meet the permitted emission allocation and water qualities. The results are helpful in establishing a rational discharge permit system of each pollution unit under water quality targets, and provide a basis for production plans of these pollution units.
