    Notes: Background  Some cases of dermatofibrosarcoma protuberans (DFSP) do not protrude above the skin.Objectives  To assess the prevalence of these DFSPs and further to describe their presentation and course.Methods  One hundred and forty-three patients were retrospectively collected. They were asked to complete a standardized questionnaire indicating the history and appearance of the DFSP from the first skin changes identified to the time of diagnosis.Results  Eighty-one DFSPs were described as protuberant ab initio, and 62 as initially nonprotuberant (npDFSP). The latter remained at this stage for a mean period of 7·6 years. Twenty-nine per cent of npDFSPs were ‘morphoea-like’, 19% were ‘atrophoderma-like’ and 42% were ‘angioma-like’. Age at diagnosis was similar for both initial presentations. npDFSPs were most often misdiagnosed by physicians.Conclusions  Nearly half the patients first identified their early DFSP-related skin changes as patches. Both this frequency and the long duration at this preprotuberant stage should prompt dermatologists to consider the diagnosis of DFSP earlier, in order to make surgical treatment easier.

    Description: The impact of ARGO trajectory assimilation on the quality of ocean analyses is studied by means of an operational oceanographic model implemented in the Mediterranean Sea and a 3D-var assimilation scheme. For the first time, both ARGO trajectories and vertical profiles together with satellite data are assimilated to produce analyses for short term forecasts. The study period covers three months during winter 2005 when four ARGO trajectories were present in the northwestern Mediterranean Sea. It is shown that their integration is consistent with the other components of the assimilation system, and it contributes to refine the model error structure with new information on horizontal pressure gradients. So the analysis benefits of a more accurate description of the boundary currents and their instabilities that drive the mesoscale activity of regional circulations. As a consequence, the trajectory assimilation remotely and significantly influences the basin scale circulation. Changes can be depicted by intermediate water mass redistributions, mesoscale eddy relocations or net transports modulations. These impacts are detailed and assessed considering historical and contemporary datasets. The obtained qualitative and quantitative agreements motivate the integration of ARGO trajectories in the operational Mediterranean Forecasting System.

Comparisons between analyses obtained with and without trajectory assimilation suggest that trajectory assimilation can impact on the description of boundary currents and their instabilities, as well as mesoscale activity at regional scales. These impacts are detailed and assessed considering historical and simultaneous in-situ datasets. The results motivate the integration of ARGO trajectories in the operational Mediterranean Forecasting System.

    Description: Cancer cells can undergo a metabolic reprogramming from oxidative phosphorylation to glycolysis that allows them to adapt to nutrient-poor microenvironments, thereby imposing a selection for aggressive variants. However, the mechanisms underlying this reprogramming are not fully understood. Using complementary approaches in validated cell lines and freshly obtained human specimens, we report here that mitochondrial respiration and oxidative phosphorylation are slowed in metastatic melanomas, even under normoxic conditions due to the persistence of a high nuclear expression of hypoxia-inducible factor-1α (HIF-1α). Pharmacologic or genetic blockades of the HIF-1α pathway decreased glycolysis and promoted mitochondrial respiration via specific reduction in the expression of pyruvate dehydrogenase kinase-3 (PDK3). Inhibiting PDK3 activity by dichloroacetate (DCA) or siRNA-mediated attenuation was sufficient to increase pyruvate dehydrogenase activity, oxidative phosphorylation, and mitochondrial reactive oxygen species generation. Notably, DCA potentiated the antitumor effects of elesclomol, a pro-oxidative drug currently in clinical development, both by limiting cell proliferation and promoting cell death. Interestingly, this combination was also effective against BRAF V600E-mutant melanoma cells that were resistant to the BRAF inhibitor vemurafenib. Cotreatment of melanomas with DCA and elesclomol in vivo achieved a more durable response than single agent alone. Our findings offer a preclinical validation of the HIF-1/PDK3 bioenergetic pathway as a new target for therapeutic intervention in metastatic melanoma, opening the door to innovative combinations that might eradicate this disease.     Description: Ultrahigh-quality (Q) factor microresonators have a lot of applications in the photonics domain ranging from low-threshold nonlinear optics to integrated optical sensors. Glass-based whispering gallery mode (WGM) microresonators are easy to produce by melting techniques, however they suffer from surface contamination which limits their long-term quality factor to a few 108. Here we show that an optical gain provided by erbium ions can compensate for residual losses. Moreover it is possible to control the coupling regime of an ultrahigh Q-factor three port microresonator from undercoupling to spectral selective amplification by changing the pumping rate. The optical characterization method is based on frequency-swept cavity-ring-down-spectroscopy. This method allows the transmission and dispersive properties of perfectly transparent microresonators and intrinsic finesses up to 4.0 × 107 to be measured. Finally we characterize a critically coupled fluoride glass WGM microresonator with a diameter of 220 μm and a loaded Q-factor of 5.3 × 109 is demonstrated.
