Fewer mines are found together on the same side of the mid-vein than separated, on opposite sides of the mid-vein. Larvae do not normally cross the mid-vein but create small blotch-like mines along subsidiary veins. Investigations of the usual mortality factors acting on these leaf-miner populations, including competition, parasitism, and predation revealed no significant differences in these factors between mines separated by the mid-vein and those on the same side of the leaf. However, early leaf abscission, which kills the larvae present in the leaf, occurs significantly more frequently in cases where larvae are clustered on one leaf side. The reasons for this differential leaf abscission are not yet clear.

    Notes: Development of porcine conceptuses included transitions of five stages: blastocysts of spherical, ovoid, and tubular forms containing an embryonic disc and trophoblast, extremely elongated filamentous blastocysts, and stages of embryogenesis between days 9 and 18 after mating. Embryonic survival was reduced by 17% during this period. In this litter-bearing species, intense alteration in distribution patterns occurred at days 11 and 12, when blastocysts rapidly elongated to filamentous forms. Increased embryo mortality did not result from rapid changes in distribution patterns of conceptuses within the same uterine horn at this time. Filamentous blastocysts quickly reached lengths often exceeding 60 cm, and these conceptuses became regularly spaced with no overlap of tubular membranes from other embryos in that horn. The number of concep-tuses within a uterine horn ranged from 1 to 13. Protein in individual concep-tuses was used as an indicator of growth and denoted exponential increase, but at a lower rate for blastocysts of spherical, ovoid, and tubular forms as compared with that found in filamentous blastocysts and embryogenesis stage conceptuses. Growth of a conceptus, based upon its protein content, was independent of the developmental stage or potential loss of those neighbors nearest that conceptus.

    Notes: To determine the origin of oocyte maturation inhibitor (OMI), cumulus-enclosed porcine oocytes from medium follicles were cultured for two days in Medium 199 alone, the low molecular weight (〈 2,000 daltons) fraction of porcine follicular fluid (pFFL), or extracts of granulosa cells from small (1-2 mm), medium (3-5 mm), and large (6-12mm) antral follicles. Additionally, the cumulus-oocyte complexes were grown in the presence of the low molecular weight fraction of “conditioned” medium from suspension cultures of medium follicle granulosa cells. The percent maturation in cultures with pFFL was significantly (P 〈 .001) less than control cultures. Similarly, addition of the granulosa cell extracts at a 1/20 dilution resulted in a significant reduction in the percent oocyte maturation as compared with controls. The percent maturation after addition of conditioned medium was similarly reduced (P 〈 .001). These results suggest that the granulosa cells probably synthesize and secrete OMI which inhibits oocyte maturation in vitro. Additionally, it appears that the content of OMI in the granulosa cells decreases as the follicle matures.

    Notes: Poly(ADP-ribose) polymerase activity was determined at various times during the in vitro life span of two human diploid fibroblast-like cell lines of different donor ages. The cell lines differed in their ability to transfer ADP-ribose, with cells from an embryonic donor exhibiting 2 to 3 times the activity found in cells obtained from a newborn donor. The activity in both cell lines decreased by 30-60% as the cells moved through their in vitro life spans. The decline could not be attributed to increases in glycohydrolase or the leakage of polymerase from older cell preparations. Enzyme activation with DNase I indicated that similar levels of enzyme were present in both cell lines at all in vitro ages. These results indicate that although poly(ADP-ribosyl)ation is inversely related to donor age as well as in vitro age the decrease is in response to other factors which change with increasing age.

    Notes: Chronic exposure of animals to 60-Hz electric fields is known to affect the nervous system in a variety of subtle ways. The mechanism whereby these effects are produced remains unknown. One hypothesis is that the effects are a result of direct interaction between neuronal membranes and induced currents. Alternatively, the effects could be produced indirectly, as a result of sensory stimulation and the resulting low-level stress. To test these hypotheses, a system was developed to expose the surface of an anesthetized cat's paw to surface electric fields up to 600 kV/m while simultaneously measuring, in dorsal root fibers, afferent nerve impulses originating from various receptor types in the exposed paw. Of the 245 receptor units tested, comprising ten cutaneous receptor types, ten responded to the electric field with an increase in firing rate. The most sensitive receptor type was the rapidly adapting field receptor (RAF); eight of 20 (40%) were sensitive to the electric field, with thresholds as low as 160 kV/m. One of 35 rapidly adapting high-frequency receptors and one of 22 type T hair-follicle receptors were also sensitive to the electric field. Follow-up tests on the RAF receptors showed that hair removal reduced but did not eliminate the electric field sensitivity, suggesting that at least one other mechanism was involved in addition to stimulation via hair movement. The most likely mechanism is field-induced vibrations of the skin, since a further reduction in firing rate occurred following application of mineral oil to the depilated paw. Direct interaction with neuronal membranes is not supported by our evidence.

    Notes: Mature female rats and their subsequent litters were exposed either to 112- or to 150-kV/m, 60-Hz electric fields or sham-exposed for 19 h daily through pre-breeding, breeding, and rearing periods of experimentation. Exposed females mated in equal percentages and reared litters of equal numbers, and mean body masses of pups were the same as those of sham-exposed animals. Thus, experiments to investigate electric-field effects on reproduction and development in rats are feasible at effective field strengths of 112 and 150 k V/m.

    Notes: Rats exposed for 3 weeks to uniform 60-Hz electric fields of 39 kV/m (effective field strength) failed to show normal pineal gland circadian rhythms in serotonin N-acetyl transferase activity and melatonin concentrations. The time required for recovery of the melatonin rhythm after cessation of field exposure was determined to be less than 3 days. The rapid recovery suggests that the overall metabolic competence of the pineal is not permanently compromised by electric-field exposure, and that the circadian rhythm effect may be neuronally mediated.

    Notes: Thirty-two male rats were tested in two replicates of an experiment to determine whether body currents induced by 60-Hz magnetic fields might lead to avoidance behavior comparable to that which results from exposure to strong 60-Hz electric fields. The test apparatus was a two-compartment Plexiglas® shuttlebox enclosed in a sound-attenuating plywood chamber, which in turn was encompassed by two copper bus bars that, when energized, served as a source of 60-Hz magnetic fields. Location of the rat, and traverse activity in the shuttlebox were monitored by nine infra-red photo detectors equally spaced along the length of the apparatus. Rats were divided into 2 groups: 1 group of rats (n = 8 per group per replicate) was sham exposed while rats in the other group (n = 8 per group per replicate) were exposed to a 3.03 mT (30.3 G), 60-Hz magnetic field whenever they traversed to or were located on the side (L or R) predetermined as the exposed side. To control artifact incident to side preference, the side exposed (L or R) was alternated over the exposed rats. Each rat was tested individually in a 1-h session. A 2-factor ANOVA (exposed vs. control, replicate 1 vs. replicate 2) failed to reveal any significant effects due to either factor or to an interaction between factors. These data demonstrate that rats do not avoid exposure to 60-Hz magnetic fields at a flux density of 3.03 mT and further imply that the avoidance by rats of high level 60-Hz electric fields is mediated by something other than the internal body currents induced by the exposure. 1992 Wiley-Liss, Inc.
