    Notes: Abstract  The action of vanadate on intact human erythrocytes was studied by 1H spin echo and 51V NMR spectroscopy as a model for the behaviour of vanadium(V) complexes in experimental diabetes. Vanadate is reduced by the intact erythrocyte at the expense of intracellular glutathione which rapidly depletes from the intracellular volume. Using the blocking agent 4,4′-diisothio-cyanatostilbene-2,2′-disulfonic acid (DIDS), which specifically blocks the anion transporter, vanadate reduction could be inhibited and glutathione depletion arrested. Thus, for the reaction with the intact cell to occur, vanadium(V) must cross the cell wall, possibly via the anion transporter. Nitrofurantoin was used to inhibit glutathione reductase in the erythrocyte suspensions. Under these conditions, treatment of the cells with vanadate induced glutathione oxidation prior to depletion. A study of the reaction of vanadate with haemolysate indicates that, without the influence of the membrane, rapid oxidation of glutathione to glutathione disulfide by the vanadyl cation occurs with no glutathione depletion, and that under these conditions vanadate reduction is incomplete. This study generates a model for the behaviour of vanadium complexes in vivo, providing a basis for the rational design and synthesis of new vanadium-based agents as insulin mimics. In essence, vanadium is transported across the membrane as vanadate(V), is reduced in situ by glutathione, and becomes complexed to a wide range of intracellular binding sites. Exchange reactions between glutathione and sulfhydryl groups present on haemoglobin and membrane lead to the depletion of glutathione from the cytosol.

    Notes: Abstract Semi-quantitative and direct determination of labelled sulphydryl groups on the surface of intact erythrocytes has been accomplished for the first time with surface-enhanced resonance Raman scattering (SERRS). The method, which involves the use of citrate-reduced silver colloids, is sensitive and selective. A 10−8 M effective concentration of picomole quantities of sulphydryl groups was determined in the presence of the normally overwhelming signal from haemoglobin. This seminal study suggests that SERRS may be applied to other in situ, site-directed labelling experiments.

    Notes: A discussion is given of the behavior of a mathematical model chosen to approximate conditions during precipitation chromatographic fractionation of polymer (Baker Williams fractionation). With only one molecular weight species of polymer a complete solution of the mathematical model was obtained. With more than one species it was necessary to use a computer solution. Curves showing the movement of polymer containing one and two species of polymer are given for a variety of conditions of operation. It was found that the degree of fractionation of polymer approaches a limiting value with increasing length of column. Under some conditions the degree of fractionation may go through a maximum with length of column. The equilibrating distance (distance of travel of the liquid phase required to establish equilibrium of polymer between the liquid and the bed) was found to be very important in determining the fractionation.

    Notes: The theory of perturbation chromatography and isotopic tracers has been applied to the investigation of the oxygen exchange reaction between carbon monoxide and carbon dioxide in the presence of a copper-zinc oxide catalyst at 205°C and atmospheric pressure. By analyzing the retention times of carbon-14 monoxide and carbon-14 dioxide tracers in six binary carbon monoxide/carbon dioxide carrier gas streams, it has been possible to shed light on the mechanism of the exchange reaction. Two types of carbon monoxide adsorption and two types of carbon dioxide adsorption were identified. Based on a numerical simulation of the experimental data, it has been concluded that the exchange reaction proceeds via the dissociative adsorption of carbon dioxide. Furthermore, the surface oxygen species formed during the dissociative adsorption of carbon dioxide appears to be the active site for the exchange reaction.

A vapor pressure flask with a built-in condenser allows the mixture to be degassed without change in composition. The entire apparatus is assembled from simple parts.     Notes: A resolution index for gel-permeation chromatograph (GPC) columns is defined by the relation log (RI) = αW, where RI is the resolution index, α is the slope of the calibration curve of logarithm of molecular weight against peak position for narrow polymer fractions, and W is the width of the GPC curves for narrow fractions. This resolution index is calculated approximately for several GPC columns individually and in series combinations by using data obtained on narrow fractions of polystyrene. The index increases with number of columns and with flow rate over a limited range of flow rate, does not vary much with solvent, and does not correlate well with plate count obtained with low molecular weight molecules. It is believed that the resolution index provides an improved indication of the performance of GPC columns.

    Notes: Interlaboratory comparisons on several materials are being conducted among a half-dozen laboratories having high speed tensile testers of widely different mechanical design and instrumentation. This progress report gives results from the first two round robins, the first on two plastic films and the second on copper wire. These materials were tested a t several speeds in the range of 1.3 to 130 m./min. (50 to 5000 in./min.). Preliminary conclusions and plans for future work are offered.

    Notes: The study of strain patterns in deformed, transparent plastic placed between crossed Polaroids can shed considerable light on the responses of these plastics to stress. This article describes a study of the differing responses of three thermoplastics of varying degrees of toughness to the notched Izod impact test. The growth and waning of the strain patterns during impact are followed by means of high-speed cinematography; interpretation of the phenomena thus observed is aided by thin section microscopy of partially deformed specimens.
