    Description / Table of Contents: Ein neuartiger Polyelektrolyt-Komplex (PEC) wurde aus den beiden natürlich vorkommenden Polyelektrolytliganden Chitosan (CS) and Pektin (PTn) hergestellt. Die PEC-Zusammensetzung sowie deren Einfluß auf das Quellungsverhalten der PEC wurden untersucht. Die Ergebnisse zeigen, daß die Quellungseigenschaften des Polyelektrolyt-Komplexes von dessen Zusamensetzung sowie von dem Deacetylierungsgrad von CS und dem Methoxylierungsgrad von PTn abhängen.
    Notes: This paper deals with a novel polyelectrolyte complex (PEC) of two natural polyelectrolyte ligands, i.e. chitosan (CS) and pectin (PTn). The composition and its effect on the swelling properties of this CS-PTn PEC were studied. The results reveal that the swelling of the polyelectrolyte complex is connected with its composition and is also affected by the degree of deacetylation of CS and the methoxy degree of PTn.

    Notes: In an ongoing effort to understand the thermodynamic properties of proteins, solid-state heat capacities of poly(amino acid)s of all 20 naturally occurring amino acids and 4 copoly(amino acid)s have been previously reported on and were analyzed using our Advanced THermal Analysis System (ATHAS). We extend the heat capacities of poly(L-methionine) (PLMFT) and poly(L-phenylalanine) (PLPHEA) with new low temperature measurements from 10 to 340 K. In addition, analyses were performed on literature data of a first protein, zinc bovine insulin dimer C508H752O150N130S12Zn, using both the ATHAS empirical addition scheme and computation with an approximate vibrational spectrum for the protein. For the solid state, agreement with the measurement could be accomplished to ±1.6% for PLMET, ±3.5% for PLPHEA, and ±3.2% for insulin, linking the macroscopic heat capacity to its microscopic cause, the group and skeletal vibrational motion. For each polymer, one set of parameters, Θ1 and Θ3, of the Tarasov function representing the skeletal vibrational contribution to the heat capacity are obtained from a new optimization procedure [PLMET: 542 K and 83 K (number of skeletal vibrations Ns = 15); PLPHEA: 396 K and 67 K (Ns = 11); and insulin monomer: 599 K and 79 K (Ns = 628), respectively]. Enthalpy, entropy, and Gibbs free energy have been derived for the solid state. © 1995 John Wiley & Sons, Inc.

    Notes: In the early Xenopus embryo, a quadrant of endodermal cells that have descended from the vegetal dorsal localization in the zygote produces signals that pass into the animal hemisphere and induce dorsal mesoderm from the marginal zone. From the remaining three quadrants of the bordering endoderm, signals pass into the animal hemisphere and induce ventral mesoderm in the marginal region. There is evidence that suggests that these same mesoderm-inducing signals continue through the plane of the tissue of the animal hemisphere where they may at least begin the processes of neural and epidermal induction by changing the competence of the prospective ectodermal cells, and possibly influencing the early regional biasing of later expression of at least some gene products, such as Epi-1 whose expression in the future epidermal domain seems specified before gastrulation. We hypothesized that the interaction of the ventral and dorsal signals within the plane of the tissue of the animal hemisphere may position the border of the neural plate. If this is so, then transplantation into the animal pole of cells that signal induction of ventral mesoderm should drive the neural plate boundary back toward the blastopore and shorten the anterior-posterior axis. Removal of cells that induce ventral mesoderm should result in an axis that is longer than normal. Results of our experiments support these predictions. Also, by late pregastrula stage 9, increasing the ventral signals has no effect. Thus the evidence suggests that the position of the anterior neural plate boundary is established before gastrulation begins by the interaction of the signals that induce the ventral and dorsal mesoderm. © 1993 wiley-Liss, Inc.

    Notes: The polyacrylic acid derivative, the copolymer (PABAM) of acrylic acid (AA) and p-benzoic acid acrylamide (BAM) were prepared by polymeric reaction. The coordinate structure of the polymer-Eu3+ complexes was characterized by X-ray photoelectron spectroscopy. The fluorescence intensity of the PABAM-Eu3+ complex was enhanced because of introduction of a BAM unit in the polyacrylic acid and reached a maximum when the mole ratio of AA to BAM was about 1 : 1.The ternary complexes of Eu3+-polymer-1,10-phenanthroline (PRL), or 2,2-bipyridine (BPD) were synthesized. The polymers used were PABAM and polyacrylic acid (PAA). The fluorescence intensity of the polymer-Eu3+ complexes was increased by 5∼21 times owing to the introduction of PRL and BPD.

    Description: Purpose: To investigate the feasibility of T 1 -weighted dynamic contrast-enhanced (DCE) MRI for the measurement of brain perfusion. Materials and Methods: Dynamic imaging was performed on a 3.0 Tesla (T) MR scanner by using a rapid spoiled-GRE protocol. T 1 measurement with driven equilibrium single pulse observation of T 1 (DESPOT1) was used to convert the MR signal to tracer concentration. Cerebral perfusion maps were obtained by using an improved gamma-variate model in 10 subjects and compared with those with arterial spin label (ASL) approach. Results: The cerebral blood volume (CBV) values were calculated as 4.74 ± 1.09 and 2.29 ± 0.58 mL/100 g in gray matter (GM) and whiter matter (WM), respectively. Mean transit time (MTT) values were 6.15 ± 0.59 s in GM and 6.96 ± 0.79 s in WM. The DCE values for GM/WM cerebral blood flow (CBF) were measured as 53.41 ± 9.23 / 25.78 ± 8.91 mL/100 g/min, versus ASL values of 49.05 ± 10.81 / 23.00 ± 5.89 mL/100 g/min for GW/WM. Bland-Altman plot revealed a small difference of CBF between two approaches (mean bias = 3.83 mL/100 g/min, SD = 11.29). There were 6 pairs of samples (5%, 6/120) beyond the 95% limits of agreement. The correlation plots showed that the slop of Y (CBF_ DCE ) versus X intercept (CBF_ ASL ) is 0.95 with the intercept of 4.53 mL/100 g/min ( r = 0.74; P 〈 0.05). Conclusion: It is feasible to evaluate the cerebral perfusion by using T 1 -weighted DCE-MRI with the improved kinetic model. J. Magn. Reson. Imaging 2012;. © 2012 Wiley Periodicals, Inc.

    Description: Patched homolog 1 (PTCH1) is a known tumor suppressor that regulates the Hedgehog (Hh) pathway and has been implicated in tumorigenesis. The role of PTCH1 in colon carcinogenesis, however, is controversial. The aim of the present study was to investigate epigenetic modifications of PTCH1 in aberrant crypt foci (ACF), the earliest precursor lesion of colorectal cancer (CRC). Using laser-capture microdissection (LCM), a pure population of ACF epithelial cells was isolated and studied. The inherent protein expression levels of SHH, PTCH1, SMO, and GLI1 were assessed by immunohistochemistry for 405 ACF, including 54 dysplastic ACF (d-ACF) and 351 non-dysplastic ACF (n-ACF). The mRNA levels and methylation status of PTCH1 were also determined in 54 d-ACF and 96 n-ACF. Our data showed the expression of SHH, SMO, and GLI1 were significantly up-regulated in d-ACF, compared to n-ACF. Also, the mRNA and protein levels of PTCH1 were lower in d-ACF than n-ACF. Using MSP or MS-HRM, PTCH1 methylation was present in 64.8% (35/54) or 63.3% (34/54), respectively, of d-ACF and 19.8% (19/96) or 22.9% (11/48), respectively, of n-ACF. PTCH1 methylation was more frequent in d-ACF than n-ACF ( p 〈 0.001) and was associated with PTCH1 mRNA levels (r = 0.358, p 〈0.01). There was a statistically significant correlation between PTCH1 methylation status and the prevalence of colorectal neoplasms. In conclusion, this study suggests that aberrant methylation of the PTCH1 promoter may be an early, initiating event of colon carcinogenesis. © 2012 Wiley Periodicals, Inc.

    Description: Tributyltin (TBT), a highly toxic environmental contaminant, has been shown to induce mitochondrial-dependent apoptosis in several mammalian cells. However, the upstream signal transduction pathways involved in TBT-induced apoptosis are still not fully elucidated. In this study, the protein phosphatase (PP) 2A, microtubule organization, and mitogen-activated protein kinases (MAPKs), including JNK, p38 and their downstream transcription factors, c-Jun and ATF-2, respectively, were investigated in human amnionic cells treated by TBT. Furthermore, the activation of procaspase-3 after blocking either one of these MAPK pathways was also observed. The results showed that TBT effectively induced apoptosis characterized by caspase-3 activation. In apoptotic cells, the inhibition of PP2A activity and microtubule depolymerization was detected. Additionally, JNK and p38, as well as their downstream targets, c-Jun and ATF-2, were activated. Moreover, a JNK inhibitor, but not p38 inhibitor, significantly reduced caspase-3 activation. It can be concluded that the inhibition of PP2A may (1) play as a role in the activation of JNK and c-Jun and the concomitant promotion of microtubule depolymerization and (2) lead to the activation of caspase-3 in TBT-induced apoptotic cells. The results of this study suggest a critical role of PP2A in the TBT toxicity mechanism. © 2011 Wiley Periodicals, Inc. Environ Toxicol, 2011.
