    Description: Two new alkaloids, mappine A ( 1 ) and mapposidic acid ( 2 ), together with eleven known compounds, were isolated from the stems of Mappianthus iodoides Hand.-Mazz. Their structures were elucidated by means of spectroscopic and mass-spectrometric analyses, particularly 1D- and 2D-NMR spectroscopy. The cytotoxic activities of the two new alkaloids were also evaluated.

    Description: A new prenylated isoflavonoid, flemiphilippinin G ( 1 ), and a new flavonol glycoside, flemiphilippininside ( 2 ), along with eleven known isoflavonoids were obtained from the roots of Flemingia philippinensis ( Merr. et Rolfe) Li . Their structures were elucidated on the basis of spectroscopic data. The in vitro cytotoxicities of compounds 1 – 13 against MCF-7, A549, and Hep-G2 cell lines were determined by using the MTT (=3-(4,5-dimethylthazol-2-yl)-2,5-diphenyl-2 H -tetrazolium bromide) colorimetric assay, and their antioxidant activities were evaluated by ferric-reducing antioxidant power (FRAP) method. Compound 1 exhibited significant cytotoxicity against all the tested cell lines with IC 50 values of 4.8–7.3 μ M , and compound 2 was found to be inactive. Both compounds 1 and 2 showed weak antioxidant activities with FRAP values of 110±15 and 124±16 μmol/g, respectively.

    Description: We performed simulations with a global model of ocean biogeochemistry forced with orbitally driven anomalies of oceanic conditions for the mid-Holocene, known as Holocene climate optimum, to investigate natural variability in the Eastern Equatorial Pacific oxygen minimum zone (EEP OMZ). While the global mean temperature during the mid-Holocene was likely slightly higher than the 1961-90 mean, the SST in the EEP was slightly lower. Mid-Holocene oxygen concentrations in the EEP OMZ are generally increased, locally by up to 50% and the EEP OMZ volume was, depending on definition of the OMZ threshold, at least 6% lower. These higher oxygen levels are the combined result of competing physical and biogeochemical processes. Our results imply that mechanisms for past changes in the EEP OMZ intensity and extension can differ from the global warming driven decline in oxygen levels observed for the recent decades and predicted for the future.

    Description: ABSTRACT Optimal resolution by stimulated emission depletion (STED) microscopy requires precise alignment of the donut-shaped depletion focus to the excitation focus. In this article, we demonstrate that fluorescence lifetime distribution can be implemented to align the STED system. Different from the traditional aligning methods in which a scattering imaging module is often equipped, the lifetime-based method is free from probable mismatches between the scattering mode and the fluorescent mode, drift errors caused by separate imaging and complex fitting methods.     Description: ABSTRACT Fat-specific protein 27 (Fsp27) is a lipid droplet-associated protein that promotes lipid droplet (LD) growth and triglyceride (TG) storage in white adipocytes. Fsp27 is also highly expressed in the steatotic liver and contributes to TG accumulation. In this study, we discovered that the liver produces Fsp27β, an alternative Fsp27 isoform, which contains 10 additional amino acids at the N-terminus of the original Fsp27 (Fsp27α). White adipose tissue (WAT) and the liver specifically expressed Fsp27α and Fsp27β transcripts, respectively, which were driven by distinct promoters. The Fsp27β promoter was activated by the liver-enriched transcription factor cyclic-AMP-responsive-element-binding protein H (CREBH) but not by peroxisome proliferator-activated receptor gamma (PPARγ) which activated the Fsp27α promoter. Enforced expression of the constitutively active CREBH strongly induced Fsp27β and the human ortholog CIDEC2 in mouse hepatocytes and HepG2 cells, respectively. In contrast, loss of CREBH decreased hepatic Fsp27β in fasted mice, suggesting that CREBH plays a critical role in Fsp27β expression in the liver. Similar to Fsp27α, Fsp27β localized on the surface of lipid droplets and suppressed lipolysis. Consequently, enforced expression of Fsp27β or CREBH promoted lipid droplet enlargement and TG accumulation in the liver. Our study demonstrated that the CREBH-Fsp27β axis is important for regulating lipid droplet dynamics and TG storage in the liver. (H epatology 2014;)

    Description: A new naphthoquinone, 6-hydroxy- α -dunnione ( 1 ) and a new binaphthoquinone, methyl 1,1′,4,4′-tetrahydro-3-hydroxy-1,1′,4,4′-tetraoxo[2,2′-binaphthalene]-3′-carboxylate ( 2 ), along with ten known compounds, including naphthoquinones, anthraquinones, and phenylethanoid glucosides, were isolated from the roots of Didymocarpus hedyotideus Chun . Their structures were identified by spectroscopic analyses, particularly 1D- and 2D-NMR spectroscopy. The cytotoxic activities of the two new naphthoquinones were also evaluated.

    Description: The 2-carboxy-6-hydroxyoctahydroindole moiety is an essential residue for the antithrombotic activity of aeruginosins, which are a class of cyanobacteria-derived bioactive linear tetrapeptides. The biosynthetic pathway of the 2-carboxy-6-hydroxyoctahydroindole moiety has not yet been resolved. AerF was indicated to be involved in the biosynthesis of the 2-carboxy-6-hydroxyoctahydroindole moiety. This study reports the cloning, expression, purification, crystallization and preliminary X-ray diffraction analysis of AerF from Microcystis aeruginosa with a C-terminal His 6 tag. The crystal diffracted to a maximum resolution of 1.38 Å and belonged to the tetragonal space group P 4 3 22, with unit-cell parameters a = b = 101.581, c = 116.094 Å. The calculated Matthews coefficient and solvent content of the crystal were 2.47 Å 3  Da −1 and 50.32%, respectively. The initial model of the structure was obtained by the molecular-replacement method and refinement of the structure is in progress.

    Description: Pou4f2 acts as a key node in the comprehensive and step-wise gene regulatory network (GRN) and regulates the development of retinal ganglion cells (RGCs). Accordingly, deletion of Pou4f2 results in RGC axon defects and apoptosis. To investigate the GRN involved in RGC regeneration, we generated a mouse line with a POU4F2-green fluorescent protein (GFP) fusion protein expressed in RGCs. Co-localization of POU4F2 and GFP in the retina and brain of Pou4f2-GFP/+ heterozygote mice was confirmed using immunofluorescence analysis. Compared with those in wild-type mice, the expression patterns of POU4F2 and POU4F1 and the co-expression patterns of ISL1 and POU4F2 were unaffected in Pou4f2-GFP/GFP homozygote mice. Moreover, the quantification of RGCs showed no significant difference between Pou4f2-GFP/GFP homozygote and wild-type mice. These results demonstrated that the development of RGCs in Pou4f2-GFP/GFP homozygote mice was the same as in wild-type mice. Thus, the present Pou4f2-GFP knock-in mouse line is a useful tool for further studies on the differentiation and regeneration of RGCs.

    Description: Upper Triassic sandstones in the Ordos Basin, northern-central China, comprise tight oil reservoirs. Using a combination of thin sections, SEM, BSE, EDS, XRD, and fluid inclusion analyses, 24 core samples from 13 wells were collected to study the petrology, paragenesis, and diagenetic processes and implications for reservoir quality. Quartz cement usually occurs as overgrowths or euhedral quartz. Extensive dissolution and albitization of K-feldspar can be observed. Five types of carbonate cements, ferrocalcite, ankerite, dolomite, calcite, and siderite, occur during different diagenetic stages. Two main types of illite and 5 main habits of chlorite are observed in this study. Kaolinite mainly occurs as booklets and vermicular aggregates. Diagenetic illite, chlorite, biotite, mixed-layer illite/smectite (I/S), and other minor minerals are also observed. The diagenetic processes include compaction, alteration of volcanic materials and mica, clay mineral transformation, cementation (silica, aluminosilicate, and carbonate), and dissolution of feldspars and rock fragments. Compaction was a significant porosity-reducing agent, and the presence of carbonate cement exerts a dominant impact on the reduction of porosity. Quartz cement and authigenic clays are less important; however, it is worth mentioning that pore-lining clays are conducive to porosity preservation. In this study, most of the porosity variation is caused by a combination of compaction, carbonate cements, quartz cement, and authigenic clays. This study gives insights into diagenetic alterations within tight sandstones and has implications for reservoir quality prediction in similar settings.
