    Notes: Rates of reaction for the alkaline hydrolysis of various hydroxamic acids in the presence of cetyltrimethylammonium bromide have been determined. Empirical reaction orders of zero, one-half, and one were found for the hydroxamic acids depending upon reaction conditions and substrate structure. N-methylhydroxamic acids exhibited only first-order kinetics. The results are consistent with the Michaelis-Menten rate equation.

    Notes: Although the blood supply and the microcirculation of flexor tendons have been studied and defined extensively using qualitative methods, the quantitative assessment of blood flow has been lacking because of the limitations of the available experimental techniques. We studied the regional blood supply to the flexor tendons of dogs by the technique of radionuclide-labeled microspheres. Seven adult mongrel dogs were used. Microsphere injection and tissue-counting techniques previously used for other tissues were applied. Samples of proximal, isthmus, and distal portions of the profundus and superficialis flexor tendons were harvested from each digital unit of available limbs from each dog. Mean (± SE) flows (ml/100 g dry tissue/min) were proximal profundus 1.78 ± 0.60 and superficialis 7.10 ± 1.50. The differences were significant (p 〈 0.01). The study suggests that regional variation in blood flow to canine digital flexor tendons exists, so that a single value for blood flow to these tendons is not relevant. Furthermore, the study supports the concept of dual (vascular and synovial) nutrition to the digital flexor tendons in dogs. These observations may have implications regarding tendon repair techniques.

    Notes: In mature dogs with comparable levels of bone remodeling, we produced either increased (with adenosine triphosphate) or decreased (with epinephrine) blood flow to one hindlimb. In 13 dogs (five control, four with increased flow, and four with decreased flow), we compared uptake, at 3 h after injection of radiolabeled diphosphonate in the mid-tibia, with blood flow as determined by microspheres. Blood flow was determined with 85Sr-labeled microspheres, and determination of uptake of 99mTc methylene diphosphonate (99mTc-MDP) was by a gamma detector. There was a linear relationship between changes in diphosphonate uptake and changes in blood flow at decreased and normal flows; however, at high flows the relationship was nonproportional, indicating a disproportionately slower increase in 99mTc-MDP uptake with increasing blood flow. In six dogs an initial 1-h uptake curve of 99mTc-MDP was determined in both control and experimental limbs under states of increased and decreased blood flow. The 30-min uptake value, 60-min uptake value, area under the curve, and the slope of the curve were related to flow as determined by microspheres. The data are consistent with the hypothesis that deposition of bone-concentrating isotopes such as 99mTc-MDP is partly controlled by blood flow; at subnormal and normal flows tracer uptake is closely related to blood flow, but at supranormal flow rates it is not and appears to be diffusion limited.

    Notes: Eleven patients with free vascularized fibula graft transplants to the upper extremity were studied for donor-side morbidity effects. Based on their surgical follow-up times, the patients were divided into either group 1 (5 ± 2 months) or group 2 (21 ± 8 months) to examine the effects of surgical trauma more closely. Gait evaluation was performed using two walkways containing different ground conditions. Knee and ankle-foot motion was measured using a three-dimensional electrogoniometer. Basic temporodistance factors and ground reactions were obtained from foot switches, instrumented floor mats, and a force plate. A modified Cybex II isokinetic dynamometer was used to quantitate ankle-foot strengths. Moderate knee and ankle-foot motion changes were seen while walking on the level, sideslope, and ramp in patients evaluated less than 10 months after their surgical procedure. Minimal joint motion effects were discovered in patients with follow-up times longer than 10 months. Muscle strengths were significantly impaired in both groups, especially foot inversion and eversion. An inverse relationship existed between the length of resected fibula and ankle evertor muscle strength. Donor side morbidity does not appear to be caused by surgical soft-tissue trauma exclusively. These patients need to be studied for several more years to determine the long-term functional effects of free vascularized fibular graft transfer.

    Description: A one-dimensional diffusion model was used to investigate the effects of dense nonaqueous phase liquid (DNAPL) source zone dissolution and remediation on the storage and release of contaminants from aquitards. Source zone dissolution was represented by a power law source depletion model, which served as a time variable boundary condition to the diffusion equation used to describe mass transport in the aquitard. Two key variables were used to assess source zone dissolution behavior on aquitard mass storage and release: the power law exponential term Γ, which reflects the influence of the source zone architecture, and a new variable defined herein as the source to aquitard mass transfer coefficient β, which reflects the influences of both the source characteristics and the aquitard media properties. As Γ increased or as β increased because of more rapid source dissolution, the aquitard concentrations, depth of penetration, and long-term back-diffusion flux decreased. However, when β increased because of increased sorption, concentrations and back diffusion increased but penetration decreased. The duration of aquitard mass loading was found to be significantly less than the duration of mass release. Moreover, the mass per unit area stored in the aquitard was 3 or more orders of magnitude less than the initial DNAPL source zone mass per unit area, and the back diffusion flux from the aquitard was typically 4 or more orders of magnitude less than the initial source zone flux. Additionally, the effects of partial source zone remediation were investigated, and the results suggest that source remediation can have a favorable effect on long-term back-diffusion risk.

    Description: The proinflammatory cytokine interleukin-1β (IL-1β), whose levels are elevated in the brain in Alzheimer's and other neurodegenerative diseases, has been shown to have both detrimental and beneficial effects on disease progression. In this paper, we demonstrate that incubation of mouse primary cortical neurons (mPCNs) with IL-1β increases the expression of the P2Y 2 nucleotide receptor (P2Y 2 R) and that activation of the upregulated receptor with UTP, a relatively selective agonist of the P2Y 2 R, increases neurite outgrowth. Consistent with the accepted role of cofilin in the regulation of neurite extension, results indicate that incubation of IL-1β-treated mPCNs with UTP increases the phosphorylation of cofilin, a response absent in PCNs isolated from P2Y 2 R -/- mice. Other findings indicate that function blocking anti-α v β 3/5 integrin antibodies prevent UTP-induced cofilin activation in IL-1β-treated mPCNs, suggesting that established P2Y 2 R/α v β 3/5 interactions that promote G 12 -dependent Rho activation lead to cofilin phosphorylation involved in neurite extension. Cofilin phosphorylation induced by UTP in IL-1β-treated mPCNs is also decreased by inhibitors of Ca 2+ /calmodulin-dependent protein kinase II (CaMKII), suggesting a role for P2Y 2 R-mediated and G q -dependent calcium mobilization in neurite outgrowth. Taken together, these studies indicate that upregulation of P2Y 2 Rs in mPCNs under proinflammatory conditions can promote cofilin-dependent neurite outgrowth, a neuroprotective response that may be a novel pharmacological target in the treatment of neurodegenerative diseases. © 2013 International Society for Neurochemistry, J.
