    Notes: The epididymis in the male bird consists of the rete testis, ductuli efferentia, and epididymal tubules. The rete testis is lined with cuboidal epithelium not resting on a basement membrane. When distended with sperm the epithelium appears squamous-like. The tubuli efferentia are lined for the most part with pseudostratied ciliated columnar epithelium resting on a basement membrane. The epithelium is thrown into permanent longitudinal ridges. The epididymal tubules are lined with ciliated columnar epithelium resting on a bastment membrane.The three types of genital ducts are intermingled to some extent, but maintain a definite sequence in their connection with one another. The rete testis drains into the tubuli efferentia, and the latter into the epididymal tubules. These drain into the vas deferens.

    Notes: Subtilisin Carlsberg was covalently attached to five macroporous acrylic supports of varying aquaphilicity (a measure of hydrophilicity). Kinetic parameters of the transesterification of S and R enantiomers of secphenethyl alcohol with vinyl butyrate, catalyzed by various immobilized subtilisins, were determined in anhydrous dioxane and acetonitrile. Enzyme enantioselectivity in acetonitrile, but not in dioxane, correlated with the aquaphilicity of the support; a mechanistic rationale for this phenomenon was proposed. Although the catalytic activity of immobilized subtilisin in anhydrous solvents strongly depended on enzyme pretreatment, the enantioselectivity was essential conserved. © 1994 John Wiley & Sons, Inc.

    Notes: The diffusivities and the vapor-liquid equilibrium coefficients of two organic components, 1-hexanol and 2-butanone, were measured in concentrated maltodextrin solutions using newly developed techniques. Accurate determination of these quantities required complete material balances of the two phases and control of the water concentration in the experimental cell. The diffusion coefficients of these solutes correlated with solution viscosity to the 1/2 power over a range of temperatures and concentrations. Other literature data for diffusion coefficients in concentrated malto-dextrin and sugar solutions are shown to correlate with viscosity.

    Notes: The rate of loss of the antioxidant 2-tertiary-butyl-4-methoxy phenol (BHA) from high-density polyethylene (HDPE) was determined using high pressure liquid chromatography (HPLC). The rate of loss was found to follow a first-order expression. Almost all of the antioxidant was lost (〉95 percent) within 1 day at 50°C, within 3 days at 40°C, and within 7 days at 30°C. The controlling parameter for mass transfer of the antioxidant was found to be volatilization rather than diffusion.

    Notes: We report measurements of the incorporation of radioactive molecules during short labeling periods, as a function of cell-cycle stage, using a cell-sorter-based technique that does not require cell synchronization. We have determined: (1) tritiated thymidine (3H-TdR) incorporation throughout S-phase in Lewis lung tumor cells in vitro both before and after treatment with cytosine arabinoside; (2) 3H-TdR incorporation throughout S-phase in KHT tumor cells in vitro and in vivo; (3) 3H-TdR incorporation throughout S-phase in Chinese hamster ovary cells and compared it with DNA synthesis throughout S-phase; (4) a mathematical expression describing 3H-TdR incorporation throughout S-phase in Chinese hamster M3-1 cells; and (5) the simultaneous incorporation of 3H-TdR and 35S-methionine as they are related to cell size and DNA content in S49 mouse lymphoma cells. In asynchronously growing cells in vitro and in vivo, 3HH-TdR incorporation was generally low in early and late S-phase and highest in mid-S-phase. However, in Lewis lung tumor cells treated with cytosine arabinoside 3H-TdR incorporation was highest in early and late S-phase and lowest in mid-S-phase. Incorporation of 35S-methionine increased continuously with cell size and DNA content. Incorporation of 3H-TdR in CHO cells was proportional to DNA synthesis.

    Notes: The thermal properties of some organotin complexes of general structure (4-ZC6H4)2SnCl2.L2 (Z=Me, CF3, F, Cl, OMe and H; L2 = 2,2′ -bipyridyl and 4,4′ -dimethyl-2,2′ -bipyridyl) are reported. The thermal data obtained by Differential Thermal Analysis (DTA) for these complexes is reported. Thermal decomposition experiments for some of the complexes are described and indicate a disproportionation of the complexes into the aryltin trichloride complex and the triaryltin chloride. The controlled decomposition provides a possible alternative preparative route to some aryltin trichloride complexes.

    Notes: Processing and intracellular transport of RNA transcripts are essential for gene expression. Translational regulation of gene expression may occur by several mechanisms, including control of transcript movement from sites of synthesis to site(s) of translation. We describe temporal analysis of the intracellular translocation of c-myc transcripts from the site of synthesis in the nucleus to sites of translation in the cytoplasm. Fluorescence in situ hybridization (FISH) and quantitative fluorescence microscopy were used to measure intracellular traffic of c-myc transcripts in individual recombinant cells following activation of c-myc sequences linked to a heat shock promoter. C-myc nuclear transcripts are visible in the nucleus within minutes of heat exposure. Transcripts remain in the nucleus for at least 4 hr after gene activation. Transport of c-myc transcripts to the cytoplasm begins ∼ 1 hr after cells are returned to 37°C. These data demonstrate the feasibility of measuring intracellular transcript transport following gene activation and provide a description of the kinetics of intracellular traffic of inducible c-myc transcripts in heated cells in situ. © 1994 Wiley-Liss, Inc.

    Notes: The relationship between gene copy number and expression and cellular consequences of elevated levels of c-myc protein has been investigated using recombinant Chinese hamster ovary cell lines transfected with DNA coding for the murine c-myc gene. HC-8 and LC-5 recombinant cells carry approximately 800 and 50 copies of c-myc sequences, respectively, under control of an inducible heat shock promoter. Multivariate flow cytometric analysis and clonogenic assays were used to measure the relationship among c-myc expression, rate of DNA synthesis, and cell survival. Following heat exposure, maximally induced HC-8 cells produced approximately tenfold more c-myc protein than heated LC-5 cells, suggesting a close relationship between gene copy number and level of expression. However, considerable heterogeneity in the level and time of c-myc expression was observed following heat induction, even though the amounts of genomic c-myc were reatively constant. Heterogeneity in gene expression was not attributable to variation in heat induction methodologies and/or cell cycle phase distributions. The presence of high levels of recombinant c-myc protein was associated with a decreased rate of bromodeoxyuridine incorporation into DNA. High levels of c-myc protein in HC-8 cells were inversely correlated with cell survival postheating, suggesting that high levels of c-myc protein are incompatible with cell survival.
