    Abstract: Patatin‐like phospholipase domain‐containing protein 3 ( PNPLA3 ) I148M (rs738409) genotype influences clinical/biochemical characteristics in patients with nonalcoholic fatty liver disease (NAFLD), but whether PNPLA3 ‐I148M (rs738409) genotype also influences the diagnostic performance of noninvasive diagnostic tests for NAFLD is uncertain. Our aim was to investigate the differences in diagnostic performance of noninvasive diagnostic tests for NAFLD according to PNPLA3 ‐I148M (rs738409) genotype. Methods Fifty‐eight healthy controls and 349 patients with biopsy‐proven NAFLD were included. Areas under the receiver operating characteristic curve (AUROCs) were calculated to predict hepatic steatosis (fatty liver index and hepatic steatosis index), nonalcoholic steatohepatitis (cytokeratin‐18 M30 and M65), and significant fibrosis (≥F2 fibrosis) (fibrosis‐4 and BARD), stratifying by rs738409 genotypes (CC and CG + GG groups). Results Fatty liver index and hepatic steatosis index showed good diagnostic performance for diagnosing steatosis only in the CG + GG group with AUROCs ranging from 0.819 to 0.832. Cytokeratin‐18 M30 (AUROC = 0.688) and M65 (AUROC = 0.678) had suboptimal performance for diagnosing nonalcoholic steatohepatitis in the CG + GG group, whereas both had good performance (AUROC = 0.814 and 0.813, respectively) in the CC group. BARD score showed good performance in the CG + GG group compared with the CC group (AUROC = 0.805 and 0.532, respectively). Fibrosis‐4 had suboptimal performance in the CG + GG group and good performance in the CC group (AUROC = 0.662 and 0.801, respectively). Conclusions Diagnostic performance of noninvasive tests for NAFLD varied markedly according to PNPLA3 genotypes. Clinicians should be aware that PNPLA3 genotype limits the clinical utility of noninvasive diagnostic tests for diagnosing NAFLD.

    Abstract: A simple protocol for the UV light irradiated preparation of heteroaryl trifluoromethyl thioethers from disulfides and inexpensive, environmentally sodium triflinate (CF 3 SO 2 Na) has been developed. The features of simple and clean reaction conditions, in moderate to good yields, and broad substrate scope render this new approach synthetically attractive for the preparation of potentially pharmaceutically active compounds.

    Abstract: The major histocompatibility complex ( MHC ) is most closely associated with nasopharyngeal carcinoma ( NPC ), but the complexity of its genome structure has proven challenging for the discovery of causal MHC loci or genes. We conducted a targeted MHC sequencing in 40 Cantonese NPC patients followed by a two‐stage replication in 1065 NPC cases and 2137 controls of Southern Chinese descendent. Quantitative RT ‐ PCR analysis ( qRT ‐ PCR ) was used to detect gene expression status in 108 NPC and 43 noncancerous nasopharyngeal ( NP ) samples. Luciferase reporter assay and chromatin immunoprecipitation (Ch IP ) were used to assess the transcription factor binding site. We discovered that a novel SNP rs117565607_A at TRIM 26 displayed the strongest association ( OR  = 1.909, Pcombined = 2.750 × 10 −19 ). We also observed that TRIM 26 was significantly downregulated in NPC tissue samples with genotype AA / AT than TT . Immunohistochemistry ( IHC ) test also found the TRIM 26 protein expression in NPC tissue samples with the genotype AA / AT was lower than TT . According to computational prediction, rs117565607 locus was a binding site for the transcription factor Yin Yang 1 ( YY 1). We observed that the luciferase activity of YY 1 which is binding to the A allele of rs117565607 was suppressed. Ch IP data showed that YY 1 was binding with T not A allele. Significance analysis of microarray suggested that TRIM 26 downregulation was related to low immune response in NPC . We have identified a novel gene TRIM 26 and a novel SNP rs117565607_A associated with NPC risk by regulating transcriptional process and established a new functional link between TRIM 26 downregulation and low immune response in NPC .

    Abstract: Tourette syndrome ( TS ) is a common tic disorder in children and adolescents. There is preliminary evidence that herbal medicine may possess the potential to treat tics. The purpose of this study was to formally evaluate the efficacy and safety of 5‐Ling Granule (5‐ LG r), a proprietary polyherbal product, for the treatment of patients with TS in comparison with tiapride and placebo. Methods In this multisite, double‐blind, double‐dummy, randomized, placebo‐controlled trial, 603 patients with TS aged 5–18 years were randomly assigned to treatment with placebo ( n  = 117), tiapride ( n  = 123, 200–400 mg/day) or 5‐ LG r ( n  = 363, 15–22.5 g/day) for 8 weeks. The primary outcome was measured using the Yale Global Tic Severity Scale ( YGTSS ) and its subscales, total tic Score ( TTS ) and tic‐related impairment. Incidence of adverse events was compared among the three groups. Results While tics of all patients were reduced over time, 5‐ LG r and tiapride treatment produced significantly greater improvement on the YGTSS overall scale and subscale for TTS and impairment at endpoint than the placebo. Seventy‐four percentage of patients in the 5‐ LG r arm and 68.3% in the tiapride arm had clinical response and these rates of response were significantly higher than those on placebo (44.0%, p  〈   .001). The incidence of overall adverse events was significantly fewer for patients on placebo and 5‐ LG r compared to tiapride (11.2% and 13.8% vs. 26.0%, p  =   .002); in particular physical tiredness, dizziness and sleep disturbance. Conclusions The clinical efficacy of 5‐ LG r is comparable to tiapride in reducing tics. Its safety profile is better than tiapride. 5‐ LG r can be considered a safe and effective therapy for TS (Trial registration: www.clinicaltrials.gov : NCT 01501695).

    Abstract: Both exposure to heavy metals and alcohol intake have been related to the risk of type 2 diabetes (T2D). In this study, we aimed to assess the potential interactions between metal exposure and alcohol intake on the risk of T2D and prediabetes in a cohort of Chinese male workers. Methods We conducted a cross‐sectional analysis of 26,008 Chinese male workers in an occupational cohort study from 2011 to 2013. We assessed metal exposure and alcohol consumption at baseline in these workers who were aged ≥20 years. Based on occupations which were categorized according to measured urine metal levels, multiple logistic regression analyses were used to evaluate the independent and joint effects of metal and alcohol exposure on the risk of T2D and prediabetes. Results Risks of T2D ( P trend  = 0.001) and prediabetes ( P trend  = 0.001) were significantly elevated with increasing number of standard drinks per week, years of drinking, and lifetime alcohol consumption. An adjusted odds ratio ( OR ) of 6.1 (95% confidence interval [ CI ]: 4.8–7.8) was observed for the smelting/refining workers (highest metal exposure levels) who had the highest lifetime alcohol consumption ( 〉 873 kg) ( P interaction  = 0.018), whereas no statistically significant joint effect was found for prediabetes ( P interaction = 0.515). Conclusions Both exposures to metal and heavy alcohol intake were associated with the risk of diabetes in this large cohort of male workers. There was a strong interaction between these two exposures in affecting diabetes risk that needs to be confirmed in future studies.

    Abstract: This retrospective study was undertaken to analyze the effectiveness and safety of surgery combined with postoperative 125 I seed brachytherapy in the treatment of mucoepidermoid carcinoma (MEC) of the parotid gland with risk factors in pediatric patients. Procedure From September 2002 to January 2012, 24 patients, ages 5–16 years (mean, 13.2 years; median, 12.3 years), with MEC of the parotid gland were included. Patients with high risk factors received 125 I seed brachytherapy (median actuarial D90, 97 Gy) within 4 weeks following surgery. Radioactivity was 18.5–33.3 MBq per seed and the prescription dose was 60–120 Gy. Overall and disease‐free survival rates, local control rate, and distant metastasis were recorded. Radiation‐associated late side effects, including dermatitis, hearing loss, thyroid nodules, and secondary malignancy, were also evaluated. Results During the follow‐up period of 5–13.4 years (median, 7.2 years), the overall and disease‐free survival rates were all 100%. No patients developed local recurrence, regional/distant metastasis, and no severe radiation‐associated complications including the second malignancy were noted. Conclusion Surgery combined with postoperative 125 I seed brachytherapy is effective and safe in the treatment of MEC of the parotid gland in pediatric patients, with no evidence of severe late radiation‐related complications. More patients and longer follow‐up data are still needed to prove the efficacy of 125 I brachytherapy.

    Abstract: The purpose of this study was to assess the association of blood pressure (BP) measurements with the risk of cardiovascular disease (CVD) and examine whether central systolic BP (CSBP) predicts CVD better than brachial BP measurements (SBP and pulse pressure [PP]). Based on a cross‐sectional study conducted in 2009‐2010 with follow‐up in 2016‐2017 among 35‐ to 64‐year‐old subjects in China, we evaluated the performance of non‐invasively predicted CSBP over brachial BP measurements on the first CVD events. Each BP measurement, individually and jointly with another BP measurement, was entered into the multivariate Cox proportional‐hazards models, to examine the predictability of central and brachial BP measurements. Mean age of participants (n = 8710) was 50.1 years at baseline. After a median follow‐up of 6.36 years, 187 CVD events occurred. CSBP was a stronger predictor for CVD than brachial BP measurements (CSBP, 1‐standard deviation increment HR = 1.49, 95%CI: 1.31‐1.70). With CSBP and SBP entering into models jointly, the HR for CSBP and SBP was 1.28 (1.04‐1.58) and 1.22 (0.98‐1.50), respectively. With CSBP and PP entering into models jointly, the HR for CSBP and PP was 1.51 (1.28‐1.78) and 0.98 (0.83‐1.15), respectively. For subgroup analysis, the association of CSBP with CVD was stronger than brachial BP measurements in women, those with hypertension and obesity. In the middle‐aged Chinese population, noninvasively estimated CSBP may offer advantages over brachial BP measurements to predict CVD events, especially for participants with higher risk. These findings suggest prospective assessment of CSBP as a prevention and treatment target in further trials.

    Abstract: Hierarchically porous HKUST‐1 has been successfully fabricated by using organic silane (3‐Aminopropyltrimethoxysilane, γ‐APS) as template under hydrothermal conditions. The amount of template affected on the porosity and morphology of the as‐synthesized samples, indicating the adjustable pore structure. The density functional theory (DFT) calculation was employed to investigate the synthetic mechanism, demonstrating that the amino group in γ‐APS may play an electrophilic role thus the MOFs could construct under the direction of template γ‐APS. The hierarchically porous structure of the as‐synthesized MOFs will improve the mass transfer efficiency compared to the conventional MOFs, demonstrating great significance for the development of MOFs.

    Abstract: Defect engineering plays an important role in property modification for piezoelectric materials. In this work, we pay much attention to the effect of Nb nonstoichiometry on structure and properties of typical 0.95(K 0.45 Na 0.55 )Nb 1+ x O 3 –0.05Bi 0.5 Na 0.5 HfO 3 ceramics. Large piezoelectric constant ( d 33  ~ 425 pC/N and ~482 pm/V) together with high Curie temperature ( T C  ~ 315°C) have been achieved in the ceramics with excess Nb content ( x  = 0.01). However, the ceramics with deficient Nb element have seriously suppressed cryogenic ε r ‐ T curves and deteriorated electrical properties. Multi‐scale characterizations including phase structure, microstructure, defect structure and domain structure have been adopted to explain the corresponding phenomenon. Defect complex caused by deficient Nb induces clamped domain wall motion, leading to blocked polarization vector and poor electrical properties. On the contrary, the enhanced properties for the ceramics with excess Nb are attributed to easier domain switching due to the suppressed vacancies. We believe that defect engineering, for example nonstoichiometry, cannot only modulate electrical properties but also help us to understand some fundamental and critical problems about KNN‐based ceramics.
