    Abstract: This study characterized an acidic polysaccharide ( OHC ‐LDPA) isolated from the medicinal and edible homologous plant Onosma hookeri Clarke var. longiforum Duthie. The structure of OHC ‐LDPA was elucidated based on the analysis of infrared, one‑/two‐dimensional nuclear magnetic resonance, and gas chromatography–mass spectrometry data. The immunostimulatory effects of OHC ‐LDPA were identified by both in vitro and in vivo models. RESULTS The structure of OHC ‐LDPA was elucidated as a typical pectin polysaccharide, consisting of galacturonic acid, galactose, arabinose, and rhamnose as the primary sugars, with linear galacturonic acid as the main chain and arabinogalacturonic acid as the main branched components. OHC ‐LDPA could significantly stimulate the proliferation and phagocytosis of RAW264.7 macrophages and the release of nitric oxide in vitro . Also, it could accelerate the recovery of spleen and thymus indexes, enhance the splenic lymphocyte proliferation responses, and restore the levels of interleukin‐2, interleukin‐10, interferon‐ γ , and immunoglobulin G in the serum in a cyclophosphamide‐induced immunosuppressed‐mice model. In addition, OHC ‐LDPA could restore the intestinal mucosal immunity and reduce the inflammatory damage. CONCLUSION OHC‐ LDPA could improve the immunity both in vitro and in vivo and could be used as a potential immunostimulant agent. © 2022 Society of Chemical Industry.

    Abstract: In this study, besides isovaleryl shikonin, another shikonin derivative, tigloylshikonin, was also isolated from the roots of Onosma hookeri Clarke . var. longiforum Duthie as a main naphthoquinone constituent for the first time. Then optimization of the ultrasonic‐assisted extraction was done by Box–Behnken design–response surface methodology on the basis of single‐factor experiments. The optimized conditions were 72% (v/v) ethanol and the material to solution ratio was 1:37(g/mL) at 52 °C for 77 min. Under these conditions, the extraction yield of ethanol extract was 36.74 ± 0.32%, the contents of isovaleryl shikonin and tigloylshikonin reached 0.094 ± 0.003% and 0.223 ± 0.006%, respectively. Notably, in that optimized condition, the yield of isovaleryl shikonin increased by approximately 7.64‐fold than the previous report. In the in vitro antioxidant activity assay, the optimal ethanol extract exhibited similar 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) scavenging activity as butylated hydroxy toluene (BHT), but slightly weaker 2,2'‐azino‐bis‐3‐ethylbenzthiazoline‐6‐sulphonic acid (ABTS) scavenging activity and total antioxidant capacity than that of BHT. However, the active polar fraction, the ethyl acetate fraction, which is enriched with naphthoquinone constituents, performs as a better antioxidant agent than BHT. Therefore, both of them could be considered as a naturally sourced antioxidants compared to commercially available synthetic drugs. Practical Application Onosma hookeri Clarke . var. longiforum Duthie, a traditional Chinese medicine and food item, has been in use since a long time. A systematic determination of the main naphthoquinones, and antioxidant capacity of the naphthoquinones‐enriched ethanol extract and different polar fractions, was carried out in the present study. The results may provide theoretical basis for the claim that naphthoquinones‐enriched ethanol extract and ethyl acetate fraction from the roots of Onosma hookeri Clarke . var. longiforum Duthie could be used as potential natural antioxidants in the pharmaceutical, food, and cosmetic industries.

    Abstract: In this study, the optimal naphthoquinones‐enriched ethanol extract from the roots of Onosma hookeri Clarke. var. longiforum Duthie ( OHC ‐LD) was obtained under an optimal condition (69% ethanol, material to solution ratio of 27:1 at 60℃ for 59 min) by the ultrasound‐assisted extraction, according to four‐variable three‐level Box–Behnken design‐response surface methodology. The experimental yield of ethanol extract was 42.08 ± 0.65%, and the contents of naphthoquinones reached to 1.07 ± 0.004%. The optimal extract exhibited similar scavenging activity against ABTS (2,2'‐azino‐bis‐3‐ethylbenzthiazoline‐6‐sulfonic acid) radical as BHT(butylated hydroxytoluene) at 1,250 µg/ml, and better DPPH (2,2‐diphenyl‐1‐picrylhydrazyl) scavenging activity than BHT at 250 µg/ml. However, the optimal ethanol extract was not sensitive to MCF‐7 cell line ( IC 50 of 321.849 µg/ml). The results revealed the naphthoquinones‐enriched ethanol extract from the roots of OHC ‐LD had could be used as a potential natural antioxidant.

    Abstract: We examined the genetic background of a Chinese Han family in which some members presented with complex arrhythmias including sick sinus syndrome, progressive conduction block, atrial fibrillation, atrial standstill and Brugada syndrome. The possible underlying mechanism associated with the genetic mutation was explored. Methods Targeted capture sequencing was conducted in the probands in the coding and splicing regions of genes implicated in inherited arrhythmias. Stable cell lines overexpressing wild type (WT) or mutant SCN5A were generated in HEK293T cells. Whole‐cell recording was performed to evaluate the functional changes in sodium channels. Results The rare heterozygous linkage mutations, SCN5A R965C and R1309H, were found in these patients with complex familial arrhythmias. Compared to WT, R965C or R1309H, the peak current of sodium channel was dramatically reduced in HEK293T cell with linkage R965C‐R1309H mutation when testing potentials ranging from −45 to 15 mV. Notably, the maximum peak current of sodium channels with R1309H and linkage R965C‐R1309H displayed significant decreases of 31.5% and 73.34%, respectively, compared to WT. Additionally, compared to R965C or R1309H alone, the linkage mutation R965C‐R1309H demonstrated not only a more obvious depolarisation‐shifted activation and hyperpolarisation‐shifted inactivation, but also a more significant alteration in the time constant, V 1/2 and the slope factor of activation and inactivation. Conclusions The linkage mutation SCN5A R965C‐R1309H led to a more dramatically reduced current density, as well as more significant depolarisation‐shifted activation and hyperpolarisation‐shifted inactivation in sodium channels than R965C or R1309H alone, which potentially explain this complex familial arrhythmia syndrome.

    Abstract: The objective of this study was to investigate the hypoglycemic and hypolipidemic effects of phlorizin on sweet tea in rats with diabetes. Diabetic rat model was established by feeding with HFD (high‐fat diet) and then treating with intraperitoneal injection of STZ (streptozocin). The experiments were divided into therapeutic and preventive experiments. In both experiments, rats were divided into normal, diabetic control, positive control, and phlorizin groups. Symptoms of diabetes, fasting blood glucose (FBG) levels, serum lipid parameters, and pathological changes in the pancreas and liver were evaluated. It was found that the symptoms of diabetes were improved by phlorizin treatment. In addition, phlorizin could decrease FBG, improve serum lipid levels, protect against damaged pancreas islet, and decrease fat deposition in hepatic cells. These effects of phlorizin can be shown only attain to a certain dosage. It can be concluded that phlorizin has the therapeutic and preventive effects on hyperlipidemia and hyperglycemia in diabetes rats.

    Abstract: Cyprinid herpesvirus 2 (CyHV‐2) is the causative agent of herpesviral hematopoietic necrosis (HVHN), an infectious disease with high morbidity and mortality worldwide. Current research indicates that CyHV‐2 only infects fish in the genus Carassius . Outbreaks of fish viral diseases are highly temperature‐dependent, and HVHN outbreaks usually occur at water temperatures of 15–25°C. In November 2018, an infectious disease occurred on cultured gibel carp farms in ZiGong, Sichuan Province, China. The water temperature at the time was approximately 10°C. The clinical signs of the diseased fish included lethargy, inappetence, severe anoxia, hemorrhage on the body surface, and pale gills. Clinical and histopathological examinations, transmission electron microscopy (TEM), and molecular analyses were used to determine the etiology of this disease. Pathological observation revealed severe necrosis of the hematopoietic organs, particularly the gills, kidney, and spleen. Spherical herpesvirus‐like virion particles were observed with TEM in the kidney and spleen tissues from moribund fish. An experimental infection demonstrated a cumulative mortality rate of 50% (10/20) at a water temperature of 13°C, whereas the mortality of the positive control group was 75% (15/20) at a water temperature of 23°C. A specific fragment of the helicase gene of CyHV‐2 was detected with polymerase chain reaction. In a phylogenetic analysis of the helicase gene, our isolate clustered with CyHV‐2 isolates in the database. Together, these results show that the putative pathogen involved in this outbreak was CyHV‐2. To the best of our knowledge, this is the first report of a gibel carp disease caused by CyHV‐2 at low temperature. These findings are extremely important in reducing the risk of HVHN outbreaks in gibel carp aquaculture.
