    Abstract: To evaluate the oocyte fertilization ability and embryo growth after cyclophosphamide ( CPA ) treatment in mice. Methods Mice were treated with CPA at different doses (0‐800 mg/kg body weight). The oocytes then were retrieved and evaluated for their in vitro fertilization efficiency. Results The average number of metaphase II ( MII ) oocytes significantly decreased by ≥400 mg/kg CPA administration. The fertilization rate also decreased in the group that was treated with ≥400 mg/kg CPA . However, after fertilization, the embryos demonstrated normal growth ability. Two weeks after CPA administration, the number of mice from which the oocytes could be retrieved markedly decreased, but the fertilization rate and development of morphological features in the embryos were similar to those of the controls. One month after CPA administration, the number of mice from which the oocytes could be retrieved, fertilization rate, and development of the morphological features in the embryos were similar to those of the controls. Conclusion The number of oocytes decreased as the CPA administration level increased; however, the oocytes' potential for fertilization and development to the blastocyst stage was not significantly affected. One month after CPA administration, the number of oocytes and the potential for development into blastocysts were recovered.

    Abstract: An 89‐year‐old woman with small papules on her face presented to our hospital complaining of progressive dyspnoea. Chest computed tomography (CT) showed bilateral multiple lung cysts, a nodular opacity in the right lower lobe, and bilateral pleural effusion. She was diagnosed with adenocarcinoma. Her son, a 65‐year‐old man, also had bilateral basally located lung cysts and a past medical history of spontaneous pneumothorax. He had multiple papules on the face and neck, which were pathologically diagnosed as fibrofolliculomas. We considered these cases to be Birt–Hogg–Dubé syndrome (BHDS). Folliculin ( FLCN ) gene mutations that may be tumour suppressive are suspected to be causative of this syndrome. FLCN dysfunction might lead to the development of various types of tumours other than renal tumours.

    Abstract: Benign tumors and tumor‐like conditions in the ampullary area are uncommon, and there are extremely rare cases of adenomyoma (AM) and adenomyomatous hyperplasia (AMH). Surgical treatment is necessary if these lesions cause biliary obstruction. In addition, the differential diagnosis of AM and AMH from carcinoma is often difficult by standard endoscopic biopsy and cytopathological analysis that may show differential findings, resulting in unnecessary surgeries sometimes being performed. Methods Immunohistochemical (IHC) analysis of periampullary AM and AMH was performed. Results For both types of lesions, epithelial glandular cells (EGCs) showed diffuse expression of MUC6 and focal expression of HIK1083, mainly in the inner region, and focal expression of MUC5AC, mainly at the surface. The EGCs showed no expression of MUC1 or MUC4, both of which were identified as malignant tumor markers in our previous series of mucin expression studies in pancreatobiliary tumors. The expression of CK7, which was diffusely positive in normal periampullary mucosa, was decreased in the EGCs of AM and AMH. Conclusions A combined evaluation of IHC findings may be effective in the detection of AM and AMH, and also in distinguishing benign periampullary lesions, such as AM and AMH, from ampulla of Vater adenocarcinoma, thus avoiding excessive surgery.

    Abstract: It has been reported that bisphenol A ( BPA ), a widespread xenoestrogen employed in the production of polycarbonate plastics, affects brain development in both humans and rodents. In the present study employing mice, we examined the effects of exposure to BPA (500 μg/kg/day) during fetal and lactational periods on the development of the locus coeruleus ( LC ) at the age of embryonic day 18 ( E 18), postnatal 3 weeks ( P3W ), P8W and P16W . The number of tyrosine hydroxylase‐immunoreactive cells ( TH‐IR cells) in females exposed to BPA was decreased, compared with the control females at P3W. At P8W , the number of TH‐IR cells in females exposed to BPA was significantly decreased, compared with the control females, whereas the number of TH‐IR cells in males exposed to BPA was significantly increased, compared with the control males, which resulted in reversed transient sexual differences in the numbers of TH‐IR cells observed in the controls at P8W . However, no significant changes were demonstrated at E18 or P16W . Next, we examined the density of the fibers containing norepinephrine transporter ( NET ) in the anterior cingulate cortex ( ACC ) and prefrontal cortex, at P3W , P8W and P16W , because NET would be beneficial in identifying the targets of the LC noradrenergic neurons. There were no significant differences shown in the density of the NET ‐positive fibers, between the control and the groups exposed to BPA . These results suggested that BPA might disrupt the development of physiological sexual differences in the LC ‐noradrenergic system in mice, although further studies are necessary to clarify the underlying mechanisms.

    Abstract: Plant ‘activators’, such as benzothiadiazole (BTH), protect plants from various diseases by priming the plant salicylic acid (SA) signalling pathway. We have reported previously that a transcription factor identified in rice, WRKY45 (OsWRKY45), plays a pivotal role in BTH‐induced disease resistance by mediating SA signalling. Here, we report further functional characterization of WRKY45. Different plant activators vary in their action points, either downstream (BTH and tiadinil) or upstream (probenazole) of SA. Rice resistance to Magnaporthe grisea , induced by both types of plant activator, was markedly reduced in WRKY45 ‐knockdown ( WRKY45 ‐kd) rice, indicating a universal role for WRKY45 in chemical‐induced resistance. Fungal invasion into rice cells was blocked at most attempted invasion sites (pre‐invasive defence) in WRKY45 ‐overexpressing ( WRKY45 ‐ox) rice. Hydrogen peroxide accumulated within the cell wall underneath invading fungus appressoria or between the cell wall and the cytoplasm, implying a possible role for H 2 O 2 in pre‐invasive defence. Moreover, a hypersensitive reaction‐like reaction was observed in rice cells, in which fungal growth was inhibited after invasion (post‐invasive defence). The two levels of defence mechanism appear to correspond to Type I and II nonhost resistances. The leaf blast resistance of WRKY45 ‐ox rice plants was much higher than that of other known blast‐resistant varieties. WRKY45 ‐ox plants also showed strong panicle blast resistance. BTH‐induced resistance to Xanthomonas oryzae pv. oryzae was compromised in WRKY45 ‐kd rice, whereas WRKY45 ‐ox plants were highly resistant to this pathogen. However, WRKY45 ‐ox plants were susceptible to Rhizoctonia solani . These results indicate the versatility and limitations of the application of this gene.

    Abstract: In this study, we established and analyzed a novel human myeloid leukemia cell line, AMU‐AML1, from a patient with acute myeloid leukemia with multilineage dysplasia before the initiation of chemotherapy. AMU‐AML1 cells were positive for CD13, CD33, CD117, and HLA‐DR by flow cytometry analysis and showed a single chromosomal abnormality, 46, XY, t(12;22)(p13;q11.2), by G‐banding and spectral karyotyping. Fluorescent in situ hybridization analysis indicated that the chromosomal breakpoint in band 12p13 was in the sequence from the 5′ untranslated region to intron 1 of TEL and that the chromosomal breakpoint in band 22q11 was in the 3′ untranslated region of MN1. The chimeric transcript and protein of MN1‐TEL could not be detected by reverse‐transcriptase polymerase chain reaction or Western blot analysis. However, the MN1 gene was amplified to three copies detected by array comparative genomic hybridization analysis, and the expression levels of the MN1 transcript and protein were high in AMU‐AML1 cells when compared with other cell lines with t(12;22)(p13;q11‐12). Our data showed that AMU‐AML1 cells contain t(12;22)(p13;q11.2) without chimeric fusion of MN1 and TEL . The AMU‐AML1 cells gained MN1 copies and had high expression levels of MN1. Thus, the AMU‐AML1 cell line is useful for studying the biological consequences of t(12;22)(p13;q11.2) lacking chimeric MN1‐TEL . © 2011 Wiley Periodicals, Inc.

    Abstract: To assess an embryo's ability to develop into a good‐quality blastocyst during the early‐cleavage stage using time‐lapse imaging and the oxygen consumption rate. Methods In total, 942 zygotes had their oxygen consumption rates measured. In total, 282 zygotes were assessed by using time‐lapse imaging. In total, 121 zygotes were examined by using both their oxygen consumption rate and time‐lapse imaging. Results The embryos with moderate respiration rates of between 0.41 and 0.61 (×10 14 /mol s −1 ) on day 3 had a 22.1% chance of becoming good‐quality blastocysts; those outside that range had a 14.3% chance. With the time‐lapse system, when the first division was within 24 hours, 22.3% of the embryos grew to good blastocysts. After 24 hours, the rate dropped to 8.6%. The intervals between two consecutive cleavages were calculated and the duration of the second cell cycle was defined. When the time was between nine hours and 13 hours, there was a higher rate of good blastocysts. Regarding both criteria, when the embryos had progressed in the optimal range, a high percentage of them had become good blastocysts; it was 8.0% outside of that range. Conclusion Individual embryos with the potential to develop into good‐quality blastocysts could be selected at day 3 of culture using these systems.

    Abstract: The oxidation behavior of high‐purity silicon carbide (SiC) prepared by chemical vapor deposition was investigated by thermogravimetry, transmission electron microscopy, and Raman spectroscopy in the temperature range 1534–1902 K in pure O 2 . The carbon layer formed at the SiC/SiO 2 interface upon oxidation above 1784 K. Raman peaks corresponding to D ‐ and G ‐bands could be identified from the carbon layer. Bubbles were observed in the SiO 2 scale after the oxidation at 1873 K. This could be attributed to the accumulation of CO gas at the SiC/SiO 2 interface, resulting in the formation of the carbon layer and bubbles. These suggest that the oxidation rate of SiC is limited by the outward diffusion of CO in the SiO 2 scale in this temperature range.

    Abstract: Over the last decade, the importance of Advanced Care Planning has been recognized in Japan. However, it is still difficult for healthcare providers and patients to verbalize how they want to spend the last days of their lives and what unfinished business they have left to accomplish. This study aimed to clarify how nurses intervened to assist patients with cancer at the end of life to express their thoughts and concerns. Twenty‐four nurses were interviewed regarding how they intervened for patients' thoughts and concerns, and data were analyzed qualitatively using a modified grounded theory approach. A 25‐item scale was selected, and the questionnaires, which included the scale, were distributed to nurses in general wards, palliative care units, and home visiting nurse services. A total of 1864 responses were analyzed. Factor analysis and structural equation model analysis were used, and three factors were identified: “assessment and intervention,” “basic care,” and “knowledge and communication,” which led to the formulation of an intervention structure model for uncovering thoughts and concerns. These results indicate the importance of basic care and communication during interventions for patients.
