    Abstract: Lung cancer (LC), particularly nonsmall cell lung cancer (NSCLC), is one of the most prevalent types of neoplasia worldwide, regardless of gender, with the highest mortality rates in oncology. Over the years, treatment for NSCLC has evolved from conventional surgery, chemotherapy, and radiotherapy to more tailored and minimally invasive approaches. The use of personalised therapies has increased the expected efficacy of treatment while simultaneously reducing the frequency of severe adverse effects (AEs). In this review, we discuss established modern approaches, including immunotherapy and targeted therapy, as well as experimental molecular methods like clustered regularly interspaced short palindromic repeat (CRISPR) and nanoparticles. These emerging methods offer promising outcomes and shorten the recovery time for various patients. Recent advances in the diagnostic field, including imaging and genetic profiling, have enabled the implementation of these methods. The versatility of these modern therapies allows for multiple treatment options, such as single-agent use, combination with existing conventional treatments, or incorporation into new regimens. As a result, patients can survive even in the advanced stages of NSCLC, leading to increased survival indicators such as overall survival (OS) and progression-free survival (PFS).

    Abstract: Riverine nutrient loads are among the major causes of eutrophication of the Baltic Sea. This study applied the Soil & Water Assessment Tool (SWAT) in three catchments flowing to the Baltic Sea, namely Vantaanjoki (Finland), Fyrisån (Sweden), and Słupia (Poland), to simulate the effectiveness of nutrient control measures included in the EU’s Water Framework Directive River Basin Management Plans (RBMPs). Moreover, we identified similar, coastal, middle-sized catchments to which conclusions from this study could be applicable. The first modelling scenario based on extrapolation of the existing trends affected the modelled nutrient loads by less than 5%. In the second scenario, measures included in RBMPs showed variable effectiveness, ranging from negligible for Słupia to 28% total P load reduction in Vantaanjoki. Adding spatially targeted measures to RBMPs (third scenario) would considerably improve their effectiveness in all three catchments for both total N and P, suggesting a need to adopt targeting more widely in the Baltic Sea countries.

    Abstract: The present paper offers a brief assessment of climate change and associated impact in Poland, based on selected results of the Polish–Norwegian CHASE-PL project. Impacts are examined in selected sectors, such as water resources, natural hazard risk reduction, environment, agriculture and health. Results of change detection in long time series of observed climate and climate impact variables in Poland are presented. Also, projections of climate variability and change are provided for time horizons of 2021–2050 and 2071–2100 for two emission scenarios, RCP4.5 and RCP8.5 in comparison with control period, 1971–2000. Based on climate projections, examination of future impacts on sectors is also carried out. Selected uncertainty issues relevant to observations, understanding and projections are tackled as well.

    Abstract: Evidence shows that soil moisture (SM) anomalies (deficits or excesses) are the key factor affecting crop yield in rain-fed agriculture. Over last decades, Poland has faced several major droughts and at least one major soil moisture excess event leading to severe crop losses. This study aims to simulate the multi-annual variability of SM anomalies in Poland, using a process-based SWAT model and to assess the effect of climate change on future extreme SM conditions, potentially affecting crop yields in Poland. A crop-specific indicator based on simulated daily soil moisture content for the critical development stages of investigated crops (winter cereals, spring cereals, potato and maize) was designed, evaluated for past conditions against empirical crop-weather indices (CWIs), and applied for studying future climate conditions. The study used an ensemble of nine bias-corrected EURO-CORDEX projections for two future horizons: 2021–2050 and 2071–2100 under two Representative Concentration Pathways: RCP4.5 and 8.5. Historical simulation results showed that SWAT was capable of capturing major SM deficit and excess episodes for different crops in Poland. For spring cereals, potato and maize, despite a large model spread, projections generally showed increase of severity of soil moisture deficits, as well as of total area affected by them. Ensemble median fraction of land with extreme soil moisture deficits, occupied by each of these crops, is projected to at least double in size. The signals of change in soil moisture excesses for potato and maize were more dependent on selection of RCP and future horizon.

    Abstract: In non-uremic subjects, IFNL4 rs368234815 predicts HCV clearance. We investigated whether rs368234815 is associated with spontaneous HCV clearance in haemodialysis patients and whether it is a stronger predictor of HCV resolution than the IFNL polymorphisms already associated with HCV clearance in dialysis subjects. We also evaluated an association of rs368234815 with patients` survival and alterations in transcription factor binding sites (TFBS) caused by IFNL polymorphisms. Methods Among 161 haemodialysis patients with positive anti-HCV antibodies, 68 (42.2%) spontaneously resolved HCV infection, whereas 93 remained HCV RNA positive. Patients were tested for near IFNL3 rs12980275, IFNL3 rs4803217, IFNL4 rs12979860, IFNL4 rs368234815, and near IFNL4 rs8099917. IFNL4 rs368234815 polymorphism (TT/TT, ΔG/TT, ΔG/ΔG) was genotyped by restriction fragment length polymorphism analysis; other IFNL polymorphisms - by high resolution melting curve analysis. We used the Kaplan-Meier method with the log-rank test for survival analysis. In silico analysis included the use of ENCODE TFBS ChIP-seq data, HOCOMOCO, JASPAR CORE, and CIS-BP databases, and FIMO software. Results The probability (OR, 95%CI, P) of spontaneous HCV clearance for rs368234815 TT/TT patients was higher than for the ΔG allele carriers (2.63, 1.38–5.04, 0.003). This probability for other major homozygotes varied between 2.80, 1.45–5.43, 0.002 for rs12980275 and 2.44, 1.27–4.69, 0.007 for rs12979860. In the additive model, rs368234815 TT/TT was the strongest predictor of HCV clearance (6.38, 1.69–24.2, 0.003). Survival analysis suggested an association of the ΔG allele with mortality due to neoplasms (log-rank P  = 0.005). The rs368234815 ∆G allele caused TFBS removal for PLAGL1. Conclusions In haemodialysis patients, the association of rs368234815 with the spontaneous HCV clearance is better than that documented for other IFNL3/IFNL4 polymorphisms only in the additive mode of inheritance. However, identifying the homozygosity in the variant ∆G allele of rs368234815 means a more potent prediction of persistent HCV infection in haemodialysis subjects that we observe in the case of the variant homozygosity of other tested IFNL3 / IFNL4 polymorphisms. Removal of PLAGL1 TFBS in subjects harbouring the rs368234815 ∆G allele may contribute to cancer susceptibility. The association of rs368234815 with cancer-related mortality needs further studies in HCV-exposed subjects.

    Abstract: Paraoxonase 1 (PON1) is known for preventing atherosclerosis through lipid-modifying features, antioxidant activity, anti-inflammatory, anti-apoptosis, anti-thrombosis, and anti-adhesion properties. Uremic patients requiring haemodialysis (HD) are especially prone to atherosclerosis and its complications. We analysed the PON1 gene ( PON1 ) polymorphisms and serum PON1 (paraoxonase) activity concerning dyslipidaemia and related cardiovascular diseases and mortality to show how they associate under uremic conditions modified by maintenance HD treatment. The rs662 AA + AG (OR 1.76, 95%CI 1.10–2.80, P = 0.018), rs854560 TT (OR 1.48, 95%CI 1.04–2.11, P = 0.031), and rs854560 AT + TT (OR 1.28, 95%CI 1.01–1.63, P = 0.040) contributed to the prevalence of atherogenic dyslipidaemia diagnosed by the triglyceride (TG)/HDL-cholesterol ratio ≥ 3.8. The normalized serum PON1 activity positively correlated with atherogenic dyslipidaemia (ẞ 0.67 ± 0.25, P = 0.008). The PON1 rs854560 allele T was involved in the higher prevalence of ischemic cerebral stroke (OR 1.38, 1.02–1.85, P = 0.034). The PON1 rs705379 TT genotype contributed to cardiovascular (HR 1.27, 95% CI 1.03–1.57, P = 0.025) and cardiac (HR 1.34, 95% CI 1.05–1.71, P = 0.018) mortality. All P-values were obtained in multiple regression analyses, including clinical variables. Multifaceted associations of PON1 with dyslipidaemia, ischemic cerebral stroke, and cardiovascular mortality in HD patients provide arguments for the consideration of PON1 and its protein product as therapeutic targets in the prevention of atherosclerosis and its complications in uremic patients.

    Abstract: Uncertain results of the status assessment performed as required by the Water Framework Directive can be responsible for misclassification of a water body’s status and may lead either to risk due to undertaking unnecessary remediation actions or risk of penalties for refraining from any action and not reaching environmental goals. Based on Bayesian decision theory, optimal decision tables are shown for two examples of water quality indicators, for a river catchment in central Poland. To overcome the problem of scarcity of publicly available monitoring data, the existing SWAT model for the studied catchment was used to generate nutrient concentration time series for the baseline conditions and under different scenarios. The status classes assessed based on annual mean concentrations of daily values for total phosphorus and total nitrogen were adopted as the ‘true’ status classes of the water bodies based on each indicator. SWAT simulation results enabled calculation of probability distributions of concentrations for the stochastic states of the water body, both for the period before and after the performance of corrective actions. Results Bayesian decision tables consisted of alternative management decisions including modernization of the existing wastewater treatment plants in the case of phosphorous and also of fallowing agricultural areas in the case of nitrogen. An example of a penalty calculation procedure is presented in the event that the subject of the case before the EU Court of Justice would be failure to achieve the environmental objectives by all water bodies belonging to the selected catchment. Conclusions Detailed discussion of this analysis indicates the potential benefits in terms of minimization of costs/losses that the proposed methodology may bring to the protection of surface waters. The presented method of risk analysis for making decisions on remedial actions when uncertainty exists about the water status assessment, can be considered as a prototype of a general methodology prepared for implementation in water protection. Unfortunately paying fines instead of taking remediation measures might be optimal for uncertain status of water bodies.
