    Notes: Summary A quick, simple and inexpensive technique utilizing an isotonic matrix of semi-solid agarose for immobilizing channel catfish (Ictalurus punctatus) peripheral blood leucocytes onto microscope slides before cytochemical staining is described. The technique was demonstrated to be superior to the more conventional means of immobilizing cells for procedures requiring either fixed (Sudan Black B and nonspecific esterase stains) or viable (Nitro-Blue Tetrazolium stain) cells. Although the agarose immobilization technique was developed with channel catfish leucocytes, its utility with other fragile cell populations seems likely.

    Notes: Summary Surgical exploration of three patients who had a radiological diagnosis of sphenoid pneumosinus dilatans disclosed an intracanalicular meningioma of the sheath of the optic nerve (MSON), bilaterally in two patients and unilaterally in the other. Two cases of sphenoid pneumosinus dilatans, previously reported, are reviewed, and a new case is presented. The relationship of sphenoid pneumosinus to MSON is discussed. Sphernoid pneumosinus dilatans may be an early sign of MSON in the optic canals or at the optic foramens.

    Notes: Summary The phospholipase C ofclostridium welchii (α toxin) has an absolute requirement for trace quantities of Ca2+. It attacks pure phosphatidylcholine particles (smectic mesophases) having a close-packed bilayer structure only when their surface zeta potential is made positive by the addition of certain divalent ions (e.g., Ca2+) to the aqueous phase or by the presence of low concentrations of long chain cations to the lipid. Alternatively, if the rotational freedom of individual phospholipid molecules is increased by the insertion of shortn-alkanols (e.g., hexanol) into the bilayer or when a monolayer of the substrate at an air/water interface is expanded, enzymic hydrolysis can occur without any requirement for a net positive charge on the surface.

    Notes: Summary It has been found that then-alkyl bromides are capable of inducing the fusion of unilamellar liposomes. These compounds can bring about fusion of liposomes composed of either pure phosphatidylcholine or phosphatidylcholine + phosphatidic acid. Fusion of unilamellar liposomes gives rise to multilamellar structures, the morphology of which has been examined by negative staining and freeze-fracture techniques. It has been shown by microelectrophoresis that then-alkyl bromides have no effect on the surface charge of liposomes, and fusion has been further characterized by use of light scattering and differential scanning calorimetry, the latter indicating that true mixing of the fatty acyl chains occurs upon fusion. Finally, fusion occurs atn-alkyl bromide levels below that required to saturate the aqueous phase of the system.

    Notes: Abstract A rapidly growing, long-term suspension culture derived from Triticum aestivum L. (wheat) was synchronized using hydroxyurea and colchicine, and a chromosome suspension with chromosomes was made. After staining with the DNA-specific fluorochromes Hoechst 33258 and Chromomycin univariate and bivariate flow-cytometry histograms showed 15 clearly resolved peaks corresponding to individual chromosome types or groups of chromosomes with similar DNA contents. The flow karyotype was closely similar to a histogram of DNA content measurements of Feulgen-stained chromosomes made by microdensitometry. We were able to show the stability of the flow karyotype of the cell line over a year, while a parallel subculture had a slightly different, stable, karyotype following different growth conditions. The data indicate that flow cytometric analysis of plant karyotypes enables accurate, statistically precise chromosome classification and karyotyping of cereals. There was little overlap between individual flow-histogram peaks, so the method is useful for flow sorting and the construction of chromosome specific-recombinant DNA libraries. Using bivariate analysis, the AT:GC ratio of all the chromosomes was remarkably similar, in striking contrast to mammalian flow karyotypes. We speculate about a fundamental difference in organization and homogenization of DNA sequences between chromosomes within mammalian and plant genomes.

    Notes: Abstract A PCR-based codominant marker has been developed which is tightly linked to Mi, a dominant genetic locus in tomato that confers resistance to several species of root-knot nematode. DNA from tomato lines differing in nematode resistance was screened for random amplified polymorphic DNA markers linked to Mi using decamer primers. Several markers were identified. One amplified product, REX-1, obtained using a pair of decamer primers, was present as a dominant marker in all nematode-resistant tomato lines tested. REX-1 was cloned and the DNA sequences of its ends were determined and used to develop 20-mer primers. PCR amplification with the 20-mer primers produced a single amplified band in both susceptible and resistant tomato lines. The amplified bands from susceptible and resistant lines were distinguishable after cleavage with the restriction enzyme Taq I. The linkage of REX-1 to Mi was verified in an F2 population. This marker is more tightly linked to Mi than is Aps-1, the currently-used isozyme marker, and allows screening of germplasm where the linkage between Mi and Aps-1 has been lost. Homozygous and heterozygous individuals can be distinguished and the procedure can be used for rapid, routine screening. The strategy used to obtain REX-1 is applicable to obtaining tightly-linked markers to other genetic loci. Such markers would allow rapid, concurrent screening for the segregation of several loci of interest.
