    Notes: Summary Lithium 3,5 diiodosalicylate (LIS), a chemical utilized for the noncytolytic extraction of cell surface antigens, was used in this study to extract glycoproteins from the cell membranes of L2C-EN leukemic blast cells. The crude soluble antigen (LIS-L2C) preparation was found to confer immunoprotection in syngeneic guinea pigs against a lethal challenge of L2C-EN. Titration of the crude LIS-L2C soluble antigen extract revealed that 1 mg antigen gave 100% protection against a 2×105 viable tumor cell challenge 2 weeks after immunization and that immunizing doses of 0.1 mg, 0.25 mg, and 0.5 mg soluble antigen afforded 17%, 66%, and 83% protection, respectively. The specificity of this immune response was demonstrated by the failure of guinea pigs immunized with 1 mg LIS extract prepared from another guinea pig tumor (line 10 hepatoma) to be refractory to a similar L2C tumor cell challenge. A cell-mediated immune response to the LIS-L2C soluble antigen was observed in animals, based on a positive delayed hypersensitivity response to the soluble antigen 5 weeks after immunization. Similarly, in vitro testing revealed a specific blastogenic recognition of the soluble antigen by immune leukocytes.

    Notes: Abstract Herbivores and their forage interact in many ways, in some instances to the benefit or detriment of herbivore and vegetation. Studies of wildebeest (Connochaetes taurinus) in Africa and snow geese (Chen caerulescens) in the Arctic have suggested that these grazers enhance graminoid production in certain sites by repeatedly using them. Other studies have concluded that herbivores are sensitive to local variation in forage quality and quantity, and preferentially use those sites that are intrinsically more productive. In this study, caribou (Rangifer tarandus) were observed foraging at different densities on two adjacent Alaskan ranges, within which particular feeding sites contained predictably high, medium, or low densities of caribou. Vegetation from one high- and one low-use site on each of the high- and low-density ranges was sampled and monitored for productivity, measured as re-growth following clipping, with the objectives of determining which forage characteristics influence usage by grazers and whether the productivity and nature of graminoid growth after clipping were related to grazing history. Forage biomass density (g/m3), shoot density (number/m2), stand densities of nutrients and minerals (g/m3), and forage concentrations of nutrients and minerals (g/100 g tissue) correlated positively with use of sites by caribou. Productivity was independent of previous use by grazers, but consistent within ranges. These results indicate that caribou are sensitive to local variation in forage quality and quantity, preferentially use those sites with higher returns of nutrients and minerals, and have the potential to enhance graminoid growth on sites that are inherently more productive.

    Notes: Summary By means of immunocytochemistry retinal S-antigen is selectively demonstrated in retinal photoreceptor cells of the rat and in pinealocytes of the hedgehog, rat, gerbil and cat. Brain areas surrounding the pineal organ are immunonegative. The immunoreactive material is evenly distributed in the perikarya of the cells. Occasionally, inner segments of retinal photoreceptors and processes of pinealocytes are also stained. The outer segments of retinal photoreceptors display a strong immunoreaction. In both pinealocytes and retinal photoreceptors the intensity of the immunoreaction varied considerably among individual cells. The immunocytochemical demonstration of retinal S-antigen in mammalian pinealocytes indicates that these cells still bear characteristics of photoreceptors. This finding is in accord with the concept that mammalian pinealocytes are derived from pineal photoreceptor cells of poikilothermic vertebrates.

    Notes: Summary The aim of this study was to examine whether rod-opsin and S-antigen immunoreactions were present in the pineal organ of adult man and how these immunoreactions were correlated with neuronal markers, e.g., synaptophysin, and neurofilaments L, H and M. Three perfusion-fixed epithalamic regions including the pineal organ and five pineal glands obtained at routine autopsy were used. The specimens were taken from female or male patients, 25 to 85 years of age. All immunoreactions were performed using highly specific, well-characterized antibodies. Rod-opsin and S-antigen-immunoreactive pinealocytes occurred in all pineal organs investigated; however, the immunoreaction was restricted to small subpopulations of pinealocytes (rod-opsin immunoreaction: approximately 3%–5%; S-antigen immunoreaction: approximately 5%–10% of the total population). In contrast, immunoreactions for synaptophysin and neurofilaments M and H were present in numerous pinealocytes. Immunoreactivity for neurofilament L was not found. These data suggest that the cellular composition of the human pineal organ is heterogeneous. Moreover, the presence of rod-opsin and S-antigen immunoreactions in the human pineal organ indicates that it may be affected by autoimmune retinal diseases that are provoked by antibodies against these proteins, as is the case in rodents and non-human primates.

    Notes: Summary A therapeutic regimen is described for sedative, analgesic, and anti-emetic effect in patients receiving intraarterial carmustine (BCNU) for malignant gliomas. This regimen consists of nalbuphine, 30 mg, i.v., and droperidol, 2.5 mg, i.v., given immediately prior to intra-carotid BCNU infusion. Droperidol, 2.5 mg, i.v., is then administered on four hour intervals for sixteen hours post-procedure. This combination provided excellent effect in nine patients treated for twelve intra-carotid infusions. None of the nine patients experienced vomiting, one experienced mild nausea several hours post-infusion, and non complained of severe pain or discomfort. Thirteen additional patients received diazepam, 10 mg, P.O., prior to the intra-carotid BCNU infusion, with fentanyl, 100 mcg, i.v., and prochlorperazine, 10 mg, i.m. at the onset of infusion. All thirteen patients suffered from severe nausea, vomiting, and orbital pain. The nalbuphine/droperidol combination is thought to provide a superior alternative to the traditional narcotic/pheonothiazine/benzodiazepine combination for carotid BCNU infusion. This combination has theoretical advantages for the patient with intracranial mass lesions by providing analgesia and sedation with minimal potential for respiratory depression and carbon dioxide retention.
