    Notes: Abstract The aim of this study was to investigate the pharmacological profile of the kinin B1 and B2 receptors in isolated stomachs from wild-type control and B2 receptor knockout mice. Isometric contractions evoked by bradykinin (BK) (9 nM) and desArg9BK (28 nM) were shown to be different. The contraction induced by desArg9BK had a longer duration than that evoked by BK and increased during incubation in vitro in stomachs of wild-type controls, while in the transgenic B2 receptor knockout mice, the contractions evoked by desArg9BK and BK were similar and followed the B1 receptor agonist pattern. BK but not the carboxypeptidase-resistant analog, [Phe8ψ(CH2-NH)Arg9]BK, was found to be active in the stomach of B2 receptor knockout mice. BK-induced contractions were prevented by mergetpa (a carboxypeptidase M inhibitor) (10 μM) and by a the B1 receptor antagonist, AcLys[DβNal7,Ile8]desArg9BK (R 715) (0.88 μM), while not being influenced by the B2 receptor antagonist HOE 140 (0.38 μM). BK and [Phe8ψ(CH2-NH)Arg9]BK were potent contractants of the wild-type mice stomach and their effects were not influenced by mergetpa or by the B1 receptor antagonist: they were reduced by HOE 140. After incubation in vitro for 3–4 hours, the tissues were treated with HOE 140 (4 μM) and FR-173657 (17 μM) to eliminate B2 receptor function. In these tissues, BK evoked a B1-like contraction which was inhibited by mergetpa (10 μM) and antagonized by R 715 (8 μM). The results indicate that BK acts primarily on B2 receptors. However, after intramural conversion to desArg9BK, activation of B1 receptors of the mice stomach occurs. In the tissues of B2 receptor knockout mice, BK behaves as a pure B1 receptor agonist while in stomachs of control animals, the B2 receptor contribution is overwhelming. After complete blockade of the B2 receptor, BK is able to evoke B1-mediated responses similar to those observed in tissues of B2 receptor knockout mice. It is concluded that the disruption of the B2 receptor gene eliminates the B2 receptor without influencing the B1 receptor system.

    Notes: Summary Dissociation and elutriation procedures were applied to the OTT-6050 mouse teratoma line carried in subcutaneous implants in 129/J mice in order to enrich the differentiating neuroepithelial cells. Subsequent renal subcapsular implantation of one of the resultant cell fractions (IB-9) in syngeneic mice led to the constant production of macroscopically pigmented tumors which, in addition to undifferentiated stem cells, contained primitive neuroepithelial populations composed of medullary epithelium, neuroblasts, and numerous ependymoblastic rosettes. Melanin pigment, confirmed by the presence of melanosomes and premelanosomes, was found in medullary epithelium and other primitive neural cells. The tumors preserved their characteristics through 65 sequential transplants over a period of 5 1/3 years. The pigment was maintained in vitro for up to 3 months in an organ culture system. Subcutaneous or intracerebral transplantation of the renal tumors of the IB-9 fraction accentuated the capacity of these primitive cells towards further neuroepithelial differentiation into mature synapse-forming neurons, and was associated with a decrease in primitive neuroepithelium and an absence or a marked decrease of melanin. Return of the tumor to the kidney resulted in the reappearance of melanin after one to three passages, again associated with the presence of primitive neuroepithelium. The recognition of melanin pigment in the OTT-6050 mouse teratoma transplants could be a useful marker for the successful selection of primitive neuroepithelial cell populations in this experimental tumor system. These populations may help to study the relationship between melanin production and certain types of primitive neuroectodermal tumors in man.

    Notes: Abstract Results of epidemiological cross-sectional studies of chronic non-specific lung diseases (CNSLD) in the Netherlands suggest that the exogenous factors smoking and atmospheric pollution have an effect on chronic production of phlegm. These factors seemed to have no effect on bronchial obstruction, except in one subgroup of men, in whom there was a relationship between the exogenous factors and the prevalence of dyspnoea, although no relationship was found between these factors and impairment of FEV1. The implications of the findings are discussed.

    Notes: Zusammenfassung Überblickt man die gesamten hier mitgeteilten Versuchsergebnisse, so kann man folgendes feststellen: Die Alkoholnarkose läßt sich durch Cardiazol, Coramin, Pervitin und Cycliton antagonistisch beeinflussen. Bei Cardiazol ergibt sich ein Optimum mit guter Weckwirkung in der Dosierung von 2,5 mg/100 g. Die höhere Dosierung von 5 mg hat geringere Weckwirkung. Auch bei Coramin ist ein Optimum bei 15 mg vorhanden, während die höhere Dosierung von 30 mg synergistisch mit dem Alkohol wirkt, im Sinne einer Verlängerung der Narkose. In gleicher Weise verkürzte auch Pervitin die Alkoholnarkose, eine lähmende Wirkung bei höherer Dosierung war nicht erkennbar. Cycliton wirkte in der niedrigeren Dosierung antagonistisch, während höhere Dosen wie bei Coramin die narkotische Wirkung des Alkohols verstärkten. Auf die Chloralosenarkose wirkte keines der Analeptica antagonistisch ein, doch war die Beurteilung durch die starke Reflexerregbarkeit, die die Chloralose hervorrief, praktisch unmöglich. Cardiazol und Pervitin verlängerten beide etwas die Narkose. Coramin und Cycliton wirkten ausgesprochen toxisch, sie töteten schon in niedrigen Gaben eine große Zahl der Tiere in der Chloralosenarkose.

    Notes: Summary Epidermal growth factor (EGF) is a peptide which stimulates cell mitotic activity and differentiation, has a cytoprotective effect on the gastroduodenal mucosa, and inhibits gastric acid secretion. The immunohistochemical localization of EGF in the Brunner's glands and the submandibular glands is well documented. The localization of EGF in other tissues is still unclarified. In the present study, the immunohistochemical localization of EGF in tissues from rat, man and a 20 week human fetus were investigated. In man and rat, immunoreaction was found in the submandibular glands, the serous glands of the nasal cavity, Brunner's glands of the duodenum, the Paneth cells of the small intestine, and the tubular cells of the kidney. In the fetus EGF was found in the kidney and in the intestinal Paneth cells. Antisera raised against rat submandibular EGF did not recognize EGF in human tissues, whereas antisera against human urinary EGF worked in rat as well as man. EGF was found only in cells with an exocrine function.

    Notes: Summary Surface membrane microprojections and coated vesicles of a well differentiated (Kernohan-Sayre grade I) cerebral fibrillary astrocytoma were compared with those of anaplastic astrocytic (grade IV, glioblastoma) cerebral tumors. Both phenomena increase with increased malignancy. This provides further evidence for membrane alterations with malignant change in human astrocytic cells. It is the first comparison of fibrillary astrocytoma surface phenomena with those of more malignant astrocytic tumors of the cerebrum.

    Notes: Abstract Background: Multimodal therapy is used increasingly in advanced gastrointestinal tumors. Potential benefits of using an intraoperative adjuvant therapy during laparoscopy for cancer have been documented in animal studies. The aim of this study was to develop a device that could deliver such an intraoperative drug therapy. Methods: We developed a micropump suitable for minimally invasive surgery procedures that allowed microdroplets of therapeutic substance to be distributed into the pneumoperitoneum (CO2), creating a ``therapeutic pneumoperitoneum.'' A closed-loop control system regulates drug delivery according to the gas flow. In vitro, the micropump is able to aerosolize various aqueous and ethanol solutions, including cytostatic and bacteriostatic drugs and adhesion-modulating agents. The size of the microdroplets has been optimized to prevent visual artifacts. Results: The micropump was tested in an animal model (pig). The system was inserted into a 5-mm trocar. After insufflation of a 12-mm CO2 pneumoperitoneum, laparoscopic sigmoid colon resections could be performed with no special difficulties. No fog developed, and no system-related complication was observed. At autopsy, the active principle was distributed to all exposed peritoneal surfaces. Conclusions: As opposed to conventional peritoneal washing, therapeutic pneumoperitoneum reaches the entire peritoneal surface, allowing an optimal drug distribution. Drug diffusion into the tissues is enhanced by the intraperitoneal pressure. Precise determination of the instantaneous and total drug quantity is possible. Therefore, this drug delivery system has several advantages over conventional irrigation. Its potential domains of application are locoregional cancer therapy, prevention of port-site recurrences, immunomodulation, analgesia, peritonitis, and prevention of postoperative adhesions.

Mit der Untersuchung wurde das Verhalten des vom Subcommissuralorgan gebildeten Reissnerschen Fadens 1–24 Std nach Injektion von3H-Tyrosin,3H-Alanin und35S-Cystin in den Seitenventrikel von Katzen geprüft. Die Verabreichung der Aminosäuren erfolgte über eingeheilte Kanülen. 2. Die Art der Tyrosin-Markierung des RF läßt sich weder als Bausteinmarkierung, d.h. als im SCO stattfindender Einbau des Tyrosins in das Sekret, noch als postmortaler Artefakt, d.h. als Anlagerung von Tyrosin an den RF durch Formalin-fixierung erklären. Vielmehr muß angenommen werden, daß es in vivo zu einer chemischen Bindung zwischen Tyrosin und dem Glykoproteid des RF kommen kann. Die notwendigen Ankergruppen könnten zum Teil von der im RF enthaltenen N-Acetyl-Neuraminsäure geliefert werden.

The fibre of Reissner (RF) a secretion of the subcommissural organ (SCO) was proved 1 to 24 hours after injections of3H-alanine,3H-tyrosine and35S-cystine into the lateral ventricle of cats. The amino acids were given by a cannula implanted 14 days before. 2. The kind of the tyrosine-labelling of Reissner's fibre cannot be explained as labelling of elements, i.e. as an insertion of the tyrosine into the secretion taking place in the SCO, nor as a postmortal artefact, i.e. as attachment of tyrosine to the RF by formaline-fixation. But it must be accepted, that there exists the possibility of a chemical binding between tyrosine and the glycoproteid of RF in vivo. The necessary anchor-groups might partly be delivered by the N-acetylneuraminic acid of the RF.
