    Notes: Abstract A variety of detrital foods derived from marsh plants were fed to the copepods Eurycemora affinis and Scottolana canadensis. The copepods did not survive well or produce eggs when feeding on detritus with smaller amounts of microbiota, but did well when a rich and abundant microbiota was present. Ciliated protozoans appear to be particularly important in the transfer of detrital energy to copepods.

    Notes: Abstract Adult female Calanus helgolandicus were transferred, immediately after collection in the English Channel in June 1984, to two unialgal diets one of which, the flagellate Cryptomonas maculata, was rich in starch, while the other, the diatom Thalassiosira weissflogii, contained no starch. The activity of the digestive enzymes amylase, trypsin and laminarinase was measured in these two populations under foodsaturating conditions over an acclimation period of eight days. Ingestion rates were measured on a daily basis and the results confirmed, together with a constant level of body protein, that the experimental conditions were above the incipient limiting concentration. In the long-term (4 to 8 d), the activities of all three enzymes were significantly elevated in the C. maculata-fed copepods, whereas ingestion rates were lower than those on T. weissflogii. These results observed under food-saturating conditions indicate a compensatory mechanism between digestive enzymes and the substrate ingested. They are consistent with previous work on Artemia sp. suggesting that the rate of assimilation is a function of the digestive enzyme activity and ingestion rate. Enzyme activity exhibited differing shortterm responses (〈48 h) on transfer to the two algal diets, which are interpreted in relation to the in situ activity of the field population.

    Notes: Abstract Accumulation of the bi-cyclic aromatic hydrocarbon 14C-1-naphthalene in adult female Calanus helgolandicus Claus and adult female Eurytemora affinis Poppe in sea water concentrations of hydrocarbon ranging from 0.2 to 992 μg/l was studied during exposure periods of up to 15 days as part of an investigation of the possible effects on marine zooplankton of persistent exposure to low levels of petroleum hydrocarbons. With both species the body levels of radioactivity increased rapidly during the first few days of the exposure period, but after exposure for 7 to 8 days to sea water containing 50 μg hydrocarbon/l an equilibrium condition was approached; in some experiments where E. affinis was exposed to 1.0 and 10 μg hydrocarbon/l for 15 days there was no further increase in body levels of radioactivity after 7 to 8 days. Using a low concentration of hydrocarbon (1 μg/l), the quantity of radioactivity accumulated after 10 days was found to be nearly fifty times greater in the smaller species, E. affinis, than in C. helgolandicus, when expressed in terms of body weight. After they had been exposed to the hydrocarbon for several days the copepods contained a considerable proportion of radioactivity that was no longer identifiable as naphthalene and was presumably present as metabolites. Radioactivity accumulated in the copepods after several days was rapidly lost after they were transferred to uncontaminated sea water: e.g. C. helgolandicus lost nearly 90% of its body level of radioactivity in 24 h. Thereafter the rate of loss was greatly reduced, and 5% of the original body level of radioactivity still remained in the copepods at the end of 11 days. Experiments on the breakdown of naphthalene added at low concentrations to sea water samples containing natural microbial populations indicated degradation rates of 0.1 to 0.2 μg/l/24 h in oceanic water, and 2.6 μg/l/24 h in inshore water samples. The results are discussed in terms of the possible transfer of hydrocarbon to a higher trophic level in areas subjected to constant low-level inputs of petroleum hydrocarbons.

    Notes: Summary Analyzes were made on muscle samples taken from the lateral part of the m. quadriceps femoris of man (lactate, pyruvate, and pH) on venous blood (lactate, pyruvate) and on capillary blood (pH). Samples were taken at rest, immediately after termination of dynamic exercise and during 20 min recovery from exhaustive dynamic exercise. Muscle pH decreased from 7.08 at rest to 6.60 at exhaustion. Decrease in muscle pH was linearly related to muscle content of lactate + pyruvate. The relationship was slightly different from what has been obtained after isometric exercise and this difference was ascribed to acid-base exchange with the blood during dynamic exercise. Lactate content was highly elevated in muscle after exercise and the concentration was 2–3 times higher than in blood. Pyruvate content was, however, only slightly higher than that at rest. During recovery lactate content of muscle decreased exponentially with respect to time, whereas pyruvate content increased. The half-time of lactate decrease was 9.5 min. From the lactate dehydrogenase equilibrium relative values on NADH/NAD ratio could be calculated. It was found that NADH/NAD was highly increased after exercise and that it had not returned to the basal value after 20 min recovery.

    Notes: Summary The time course of phosphorylcreatine (PC) resynthesis in the human m. quadriceps femoris was studied during recovery from exhaustive dynamic exercise and from isometric contraction sustained to fatigue. The immediate postexercise muscle PC content after either form of exercise was 15–16% of the resting muscle content. The time course of PC resynthesis during recovery was biphasic exhibiting a fast and a slow recovery component. The half-time for the fast component was 21–22 s but this accounted for a smaller fraction of the total PC restored during recovery from the isometric contraction than after the dynamic exercise. The half-time for the slow component was in each case more than 170 s. After 2 and 4 min recovery the total amounts of PC resynthesized after the isometric exercise were significantly lower than from the dynamic exercise. Occlusion of the circulation to the quadriceps completely abolished the resynthesis of PC. Restoration of resynthesis occurred only after release of occlusion.

    Notes: Abstract Acquisition of a shock avoidance task was impaired in mice after cessation of chronic consumption of ethanol, tertiary-butanol (t-butanol), or pentobarbital. The drugs were administered in liquid diets for 7 days and avoidance behavior was impaired in a test 1 day after withdrawal of the drugs. The avoidance deficit was also observed 8 days after withdrawal from chronic pentobarbital. There was no apparent relationship between the avoidance deficit and physical dependence, as measured by a decrease in body temperature or convulsions on handling, since at 6 h after withdrawal only moderate withdrawal signs were seen in the mice consuming ethanol or t-butanol, and no withdrawal signs were seen in any of the mice at the time of avoidance testing. These results suggest that impairment of avoidance behavior after chronic exposure is a general effect of central nervous system depressants and, in the case of ethanol, is not due to the production of acetaldehyde.

    Notes: Abstract Cholinergic stimulation of chloride secretion involves the activation of a basolateral membrane potassium conductance, which maintains the electrical gradient favoring apical Cl efflux and allows K to recycle at the basolateral membrane. We have used transepithelial short-circuit current (I SC), fluorescence imaging, and patch clamp studies to identify and characterize the K channel that mediates this response in T84 cells. Carbachol had little effect on I SC when added alone but produced large, transient currents if added to monolayers prestimulated with cAMP. cAMP also enhanced the subsequent I SC response to calcium ionophores. Carbachol (100 μm) transiently elevated intracellular free calcium ([Ca2+] i ) by ∼3-fold in confluent cells cultured on glass coverslips with a time course resembling the I sc response of confluent monolayers that had been grown on porous supports. In parallel patch clamp experiments, carbachol activated an inwardly rectifying potassium channel on the basolateral aspect of polarized monolayers which had been dissected from porous culture supports. The same channel was transiently activated on the surface of subconfluent monolayers during stimulation by carbachol. Activation was more prolonged when cells were exposed to calcium ionophores. The conductance of the inward rectifier in cell-attached patches was 55 pS near the resting membrane potential (−54 mV) with pipette solution containing 150 mm KCl (37°C). This rectification persisted when patches were bathed in symmetrical 150 mm KCl solutions. The selectivity sequence was 1 K 〉 0.88 Rb 〉 0.18 Na ≫ Cs based on permeability ratios under bi-ionic conditions. The channel exhibited fast block by external sodium ions, was weakly inhibited by external TEA, was relatively insensitive to charybdotoxin, kaliotoxin, 4-aminopyridine and quinidine, and was unaffected by external 10 mm barium. It is referred to as the KBIC channel based on its most distinctive properties (Ba-insensitive, inwardly rectifying, Ca-activated). Like single KBIC channels, the carbachol-stimulated I SC was relatively insensitive to several blockers on the basolateral side and was unaffected by barium. These comparisons between the properties of the macroscopic current and single channels suggest that the KBIC channel mediates basolateral membrane K conductance in T84 cell monolayers during stimulation by cholinergic secretagogues.

    Notes: Abstract Isometric contraction of the quadriceps muscle sustained to fatigue with a force of 66% of the maximum voluntary contraction force resulted in a mean glycogen utilization of 80.4 (S.D. 58.4) mmol glucosyl units/kg dry muscle (d.m.) and an accumulation of glycolytic intermediates and glucose corresponding to 82.9 (S.D. 17.5) mmol glucosyl units/kg d.m. Accumulation of hexose phosphates (principally glucose 6-phosphate) accounted for 35.4% (S.D. 4.1) of the total increase and lactate for 59.3% (S.D. 2.8). During a 4 min recovery period glucose 6-[hosphate content showed a linear decrease with a half time of 2.0 min and lactate decreased exponentially with a half time of 2.5 min. The rate of lactate disappearance from the muscle was approximately 4 times as fast as that observed previously after maximal bicycle exercise. This was probably due to a lower lactate concentration in blood after isometric contraction resulting in a larger muscle-blood gradient for lactate. Muscle content of free glucose was increased after contraction and increased further during recovery. It is concluded that the glucose increase is confined to the intracellular pool and is an effect of hexokinase inhibition by accumulated glucose 6-phosphate. Occlusion, of the local circulation after the contraction inhibited the recovery processes for lactate and glucose 6-phosphate.

    Notes: Abstract  Amphibian larvae vary tremendously in size at metamorphosis and length of larval period. We raised pond-dwelling four-toed salamander (Hemidactylium scutatum) larvae to test two models that predict a larva’s age and size at metamorphosis. The Wilbur-Collins model proposes that the developmental rate of a larva responds to changes in growth rate in an adaptive manner throughout the larval period, and that metamorphosis can be initiated after a minimum size has been reached. The Leips-Travis or fixed-rate model states that developmental rate is set early in the larval period, perhaps by early growth rate or food availability and their positive correlation with developmental rate, and that changes in growth rate during the larval period affect size at metamorphosis, but have no effect on the age of an individual at metamorphosis. A modified version of the Wilbur-Collins model suggests that a larva’s developmental rate becomes fixed about two-thirds of the way through the larval period, with changes in growth rate after that point only affecting size at metamorphosis. Larvae were raised on eight different feeding regimes which created two constant and six variable growth histories. Growth history did significantly affect size at metamorphosis. However, an a posteriori statistical test revealed a group of seven and an overlapping group of six treatments with indistinguishable lengths of larval period, indicating a general picture of a fixed developmental rate regardless of growth history. This result is unique among similar studies on invertebrates, fish, and frogs. There was no association between early growth or food level and development rates. Neither the Wilbur-Collins nor the Leips-Travis fixed-rate models were supported. The invariable developmental rate of Hemidactylium and recent osteological evidence from the literature suggest that larvae begin the process of metamorphosis as soon as they hatch, probably a trait selected for by strong predation pressure in the aquatic environment. A variety of different approaches (ecological, developmental, phylogenetic) are necessary to fully evaluate the adaptive nature of the timing of transitions between life cycle stages.
