Notes: Abstract Combinations of feeding deterrents are additive in their effects on the feeding behaviour of Locusta migratoria. The effect has been shown within and across different chemical classes. A mixture of fourteen different phenolic compounds from Sorghum bicolor tested in naturally occurring concentrations is deterrent although the individual levels of deterrence are not measurable. The significance of the findings in relation to plant resistance is discussed.

Notes: Summary Strains of Volvox carteri forma nagariensis derived from Japanese and Indian isolates (“J” and “I” strains, respectively) exhibited length differences (RFLPs) for approximately 90% of the restriction fragments detected by hybridization with a variety of unique-sequence, small-gene-family and repetitive-element probes, including heterologous probes of chloroplast and mitochondrial origin. Extensive post-zygotic mortality was observed among the zygotes produced by crossing J and I strains, suggesting some form of genetic incompatability between them. Most of the viable progeny exhibited recombinant patterns of nuclear inheritance and maternal inheritance of mitochondrial and chloroplast markers. However, many progeny exhibited exclusively uniparental (usually maternal, but in one case paternal) inheritance of both nuclear and organellar markers. Some of these non-recombinant individuals may be derived from “parthenospores” (dormant asexual cells resembling zygospores). Others may be a result of “pseudogamy,” in which one of the parental pronuclei is excluded from the zygote, followed by selective exclusion of both the mitochondrial and the chloroplast genomes derived from that same parent. When segregation patterns for 44 nuclear markers were analyzed in 90 recombinant progeny, statistically significant, locus-specific deviations from expected Mendelian transmission ratios were observed for a sizeable fraction of all markers in both reciprocal crosses: some markers were preferentially transmitted by the J strain, while others were preferentially transmitted by the I strain. It is speculated that these transmission distortions may be related to the regions of inter-isolate genetic incompatability, and may complicate the use of JxI crosses to establish a RFLP-based linkage map for the species.

Notes: Summary Rabbits were fed a cholesterol-enriched diet to render them atheromatous. After 3 months on this diet they were switched to a low-lipid stock diet. Some animals were killed at this point, while the rest were divided into (a) a group allowed to eat ad-libitum and (b) a restricted group allowed to eat half by weight of what the ad-libitum group consumed. Most animals were killed at 9 months (i.e. after 6 months' regression). The group allowed the restricted diet showed a 27% weight loss, but their serum cholesterol fell slightly more slowly than that of the ad-libitum animals. Likewise, atherosclerosis was slightly worse in the restricted than in the ad-libitum group. The results do not support the view that severe dietary restriction causes atherosclerosis to regress.

Notes: Summary The histopathology of 40 cases of achalasia of the cardia, 6 cases of oesophageal spasm-incoordination and 4 cases of scleroderma was examined. Three cases of carcinoma and 6 cases of reflux oesophagitis were used as a control group. A nearly complete loss of myenteric ganglion cells was found in the upper thickened segment in achalasia. Some surviving ganglion cells were found in the lower segments in half the cases of achalasia; in two cases counts were normal in this segment. The occurrence of neuronal chromatolysis in 9 biopsies of achalasia supports the view that an active disease process was involved. The preganglionic parasympathetic fibres in two cases of achalasia were normal in appearance and number; this somewhat limited evidence tends to count against a primary disorder of the preganglionic neurone in this condition. The 6 cases of oesophageal spasm-incoordination showed similar neuronal loss to that in the lower segment in achalasia. Possibly “oesophageal spasm” represents an early stage or incomplete expression of achalasia. One case of scleroderma showed loss of ganglion cells, but the myenteric plexus was here involved by the disease process. None of the 9 cases in the control group showed any loss of ganglion cells or chromatolysis. Acute and chronic inflammation was not convincingly associated with loss of ganglion cells in either achalasia or oesophageal spasm.

Notes: Summary Using the monoclonal antibody HAM-56 with the avidin-biotin method on recent or archival paraffinembedded tissue from multiple sclerosis brains, we have been able to distinguish between acute, chronic active and inactive lesions. HAM-56 stains all macrophages, monocytes and at least some microglia; it is the only pan-macrophage marker to our knowledge that can be satisfactorily used on conventional paraffin sections. A much narrower range of mainly perivascular macrophages in acute plaques of multiple sclerosis is stained with MAC-387, anti-muramidase and anti-alpha1-antitrypsin. The acute plaques show HAM-56-stained macrophages throughout the lesion, and these macrophages exhibit profiles of phospholipid-rich myelinic bodies, which are also usually stainable with Luxol fast blue. Active ongoing lesions show a rim of macrophages at the edge of the lesion. These macrophages show profiles of large vacuoles, thought to represent the sudanophilic esterified cholesterol formed during demyelination. Inactive cases show none of these features; the few perivascular macrophages present often contain the end product of lipid peroxidation, ceroid-lipofuscin.

Notes: Summary An analysis is presented of the immunohistological and ultrastructural features in a series of 118 surgically removed pituitary adenomas all of which were studied immunohistologically using antisera to growth hormone (GH), prolactin (PRL) ACTH, βFSH, βLH and βTSH, and 75 of which were studied ultrastructurally. Results were analysed according to the mode of presentation of patients. Forty-one (35%) of the tumours were from patients with acromegaly or gigantism, ten (9%) from patients with Cushing's syndrome or Nelson's syndrome, 19 (16%) from patients with clinical features associated with hyperprolactinaemia and 48 (40%) from patients with space occupying lesions which appeared clinically to be overtly endocrinologically functionless. By light microscopy, using the immunoperoxidase (PAP) technique, immunoreactive GH was demonstrated in all the tumours from patients with acromegaly or gigantism, immunoreactive ACTH in all tumours from patients with Cushing's syndrome or Nelson's syndrome and immunoreactive PRL in 95% of tumours associated with effects of hyperprolactinaemia. Forty-five percent of the tumours from acromegalic patients contained some PRL-positive cells as well as GH-positive cells. Among the tumours which appeared clinically to be endocrinologically functionless were three tumours (from males) uniformly stained for immunoreactive PRL. Of the remainder, 60% were negative for immunoreactive hormones and 40% contained small numbers of cells which were positive for a variety of immunoreactive hormones. ACTH-cell and PRL-cell tumours had ultrastructural features as described in previous studies. Fifty percent of GH-cell tumours examined at the EM level contained fibrous bodies, while in the remainder these structures were not identified. Tumours with fibrous bodies were more likely to contain PRL as well as GH with immunoperoxidase. All tumours that were endocrinologically functionless and which were examined at the EM level contained secretory granules. Oncocytic change was common in these tumours. No ultrastructural differences were observed between those which contained immunoreactive hormones by light microscopy and those which did not.

Notes: Summary The effectiveness of 200 μg oestradiol benzoate (ODB) given at various times following ovulation, to overcome a 3–4-day difference in ovulation times between donor and recipient was examined. In approximately half of the recipients in which the interval from the administration of human chorionic gonadotrophin (hCG) to ODB was 2 days, some of the eggs implanted. With a 4-day interval, however, neither implantations nor degenerate blastocysts were found at autopsy on days 11 or 12 in recipients of either tubal or uterine eggs.

Notes: Summary The indications and results of transsphenoidal surgery for pituitary adenomas are analysed. The advantages and disadvantages of Bromocriptine and radiotherapy are considered. Transsphenoidal surgery does not damage pre-existing pituitary function. It produces excellent results in micro and mesoadenomas, curing about 70–80% of patients irrespective whether the tumour produces ACTH,prolactin or growth hormone. Moreover there is a very small relapse rate. Macroadenomas of non-functioning type are also well treated by transphenoidal surgery, with a small recurrence rate as judged by CT scan follow-up. But macroadenomas secreting prolactin or growth hormone are less effectively treated by transsphenoidal surgery, only about 40% being cured. Invasive macro prolactinomas are not helped by surgery and should be treated with Bromocriptine and radiotherapy. Radiotherapy should be used sparingly and has inevitable complications. Post-operative delayed visual deterioration is caused either by recurrent tumour or radiotherapy. There is no good evidence to support the secondary empty sella syndrome as a cause of such delayed visual deterioration.

Notes: Summary Osmium tetroxide and OTAN histochemical reactions have been reappraised following recent publications in this journal by Ellbder and Lojda. Provided that the standard OTAN reaction is used on frozen sections of conventional thickness, unsaturated hydrophilic polar lipids are stained in an orange or red shade while unsaturated hydrophobic non-polar lipids are stained brown-black or black. We have confirmed the anomaly — pointed out by Elleder and Lojda — that the “polar lipid” reaction of the atherosclerotic plaque is essentially extinguised by acetone. Solubility and blockading methods provide circumstantial evidence that Elleder and Lojda's assumed OsO4-protein reactions are due to the lipid moiety of a lipoprotein complex.
