    Notes: Abstract The applicability of a chlorophyll fluorescence assay for kanamycin (Km) resistance screening in transgenic tobacco (Nicotiana tabacum) and Arabidopsis thaliana plants was investigated. In wild-type leaves incubated in the presence of 200 mg/l Km, a decrease in maximum variable fluorescence ((Fv)m) and a significant increase in constant fluorescence (Fo) were observed. Using (Fv)m/Fo as a screening parameter, we were able to distinguish Km-treated samples from untreated samples within 4 days. This parameter was applied to Km resistance screening using tobacco plants transformed with the nptII gene via Agrobacterium. Among 74 shoots selected on medium containing 200 mg/l Km, 37 plants were scored as Km sensitive by the chlorophyll fluorescence assay. These 74 scorings proved to be accurate, as reconfirmed by (1) polymerase chain reaction amplification of the transgene, (2) enzymatic assay of neomycin phosphotransferase and (3) leaf disc assay. Using the chlorophyll fluorescence assay, we could also screen 3-week old Arabidopsis plants carrying the nptII gene. These results clearly demonstrate the reliability and efficiency of this nondestructive assay for Km resistance screening of transgenic plants.

    Notes: Abstract. We have investigated the origin and nature of chromosome spatial order in human cells by analyzing and comparing chromosome distribution patterns of normal cells with cells showing specific chromosome numerical anomalies known to arise early in development. Results show that all chromosomes in normal diploid cells, triploid cells and in cells exhibiting nondisjunction trisomy 21 are incorporated into a single, radial array (rosette) throughout mitosis. Analysis of cells using fluorescence in situ hybridization, digital imaging and computer-assisted image analysis suggests that chromosomes within rosettes are segregated into tandemly linked “haploid sets” containing 23 chromosomes each. In cells exhibiting nondisjunction trisomy 21, the distribution of chromosome 21 homologs in rosettes was such that two of the three homologs were closely juxtaposed, a pattern consistent with our current understanding of the mechanism of chromosomal nondisjunction. Rosettes of cells derived from triploid individuals contained chromosomes segregated into three, tandemly linked haploid sets in which chromosome spatial order was preserved, but with chromosome positional order in one haploid set inverted with respect to the other two sets. The spatial separation of homologs in triploid cells was chromosome specific, providing evidence that chromosomes occupy preferred positions within the haploid sets. Since both triploidy and nondisjunction trisomy 21 are chromosome numerical anomalies that arise extremely early in development (e.g., during meiosis or during the first few mitoses), our results support the idea that normal and abnormal chromosome distribution patterns in mitotic human cells are established early in development, and are propagated faithfully by mitosis throughout development and into adult life. Furthermore, our observations suggest that segregation of chromosome homologs into two haploid sets in normal diploid cells is a remnant of fertilization and, in normal diploid cells, reflects segregation of maternal and paternal chromosomes.

    Notes: Summary A rice plant resistant to 5-methyltryptophan (5MT) was selected from mutagenized M3 seeds (Oryza sativa L. var. Sasanishiki) originating from panicles treated with ethylene imine (0.2%) 2 h after flowering. When germinated on 5MT-containing medium, the seeds (M4) from selfed plants segregated with a 3 resistant:1 sensitive ratio, indicating that the plant was heterozygous for a resistance gene and that the resistance was dominant. The resistance was also expressed in callus derived from seeds. Analysis of the free amino acids in seeds, seedlings, and calli showed that homozygous resistant plants (TR1) contained higher levels of total free amino acids than sensitive plants. In particular the levels of tryptophan, phenylalanine, and histidine were, respectively, 8.5, 5.4, and 4.9 times higher than those in the sensitive plants.

    Notes: Abstract This paper proposes a new crosstie overlay slow-wave structure for a traveling-wave electro-optical modulator and presents a design method including design considerations for the optimum structure. This structure can be designed to satisfy the phase velocity and impedance matching conditions simultaneously over a broad spectral bandwidth. This idea can be applied to a number of electrode structures for broadband traveling-wave optical modulators.

    Notes: Abstract This paper proposes and analyzes a GaAs traveling-wave optical modulator which uses a modulated coplanar strip electrode with periodic cross-tie overlay. This slow-wave structure can be designed to satisfy phase velocity and impedance matching conditions simultaneously. The dominant conductor loss in the slow-wave structure is reduced using the modulated coplanar strip electrode. The calculated 3-dB modulation bandwidth (100 GHz) is much wider than the bandwidth limit (30 GHz) of conventional electrode structures that are limited by phase velocity mismatch.

    Notes: Abstract. An all-optical computing device is proposed to perform incoherent digital operations, including the NAND, NOR, and XOR gates. Instead of using interferometer-based methods, making use of self-generating noise gratings and the erasure of them by another beam in two photorefractive self-pumped phase conjugators (SPPC), the device is able to process mutually incoherent as well as mutually coherent inputs. Changing the orientations of two polarizers and a photorefractive crystal, we are able to perform incoherent parallel NAND, NOR, and XOR operations with this proposed setup.

    Notes: Methods are presented for examining the consistency of experimental data for microbial growth where light energy is converted to chemical energy through photosynthesis. True growth yield and maintenance parameters are estimated for several sets of available experimental data. Methods of parameter estimation are presented which allow all of the measured variables to be used simultaneously for parameter estimation. The results show that a wide range of values have been found for the true growth yield and maintenance parameters. Values of the true growth yield range from 0.04 to values above those predicted by the Z-scheme model for photosynthesis.

    Notes: Available electron methods are presented and used to estimate theoretical energetic growth yields for photoautotrophic, mixotrophic, and photoheterotrophic growth of algae and photosynthetic bacteria. The theoretical yields are compared to experimental values reported previously. For photoautotrophic and mixotrophic growth of algae experimental values that approach and even exceed the theoretical values have been reported in the literature. For photosynthetic bacteria experimental yields are much smaller than thetheoretical maximum values.
