    Abstract: Heat shock protein 70 ( HSP70) genes play essential roles in guarding plants against abiotic stresses, including heat, drought, and salt. In this study, the SlHSP70 gene family in tomatoes has been characterized using bioinformatic tools. 25 putative SlHSP70 genes in the tomato genome were found and classified into five subfamilies, with multi-subcellular localizations. Twelve pairs of gene duplications were identified, and segmental events were determined as the main factor for the gene family expansion. Based on public RNA-seq data, gene expression analysis identified the majority of genes expressed in the examined organelles. Further RNA-seq analysis and then quantitative RT-PCR validation showed that many SlHSP70 members are responsible for cellular feedback to heat, drought, and salt treatments, in which, at least five genes might be potential key players in the stress response. Our results provided a thorough overview of the SlHSP70 gene family in the tomato, which may be useful for the evolutionary and functional analysis of SlHSP70 under abiotic stress conditions.

    Abstract: Objectives: Carallia brachiata Merr. is a Vietnamese medicinal plant, used to treat mouth ulcers, tongue sores, sore throats, and malaria. This paper focuses on the chemical constituents of C. brachiata and their inhibition of NO production. Methods: The dried and minced sample was extracted with methanol. The crude extract was isolated by various chromatographic methods. The structures of the isolates were determined by HR-ESI-MS, NMR, and CD spectra. The NO inhibition activity was evaluated in LPS-activated RAW264.7 cells. Results: Two new catechin derivatives (carabrachiatanins A (1) and B (2)) and seven known proanthocyanidins (3-9) were isolated from C. brachiata by bioguided fractionation. All isolates significantly showed NO inhibition effects with IC 50 values ranging from 13.72 ∼ 34.28 µM. Conclusions: Two new and seven known compounds were isolated from C. brachiata. This is the first report of compounds 3-9 from C. brachiata. Compounds 7 and 8 inhibited NO production with their IC 50 values of 16.31 ± 0.61 µM and 16.53 ± 0.83 µM, respectively, compared to that of dexamethasone, which showed IC 50 value of 14.94 ± 1.11 µM.

    Abstract: We recently reported a standardized xeno- and serum-free culture platform to isolate and expand umbilical cord-derived mesenchymal stem/stromal cells (UC-MSCs). Comparing populations from the same passage, cells that were cryopreserved and culture-rescued exhibited characteristics similar to those of their fresh counterparts, continuously cultured cells without interim cryopreservation. The culture rescue after thawing allowed for the cells to be fully recovered. However, since it would be more cost-effective and timesaving if cryopreserved cells can be used as an off-the-shelf product, we set out to compare the bioactivity of freshly thawed UC-MSCs versus culture-rescued UC-MSCs of the same batch that were recultured for an additional passage under our xeno- and serum-free protocol. UC-MSCs showed high viability in both the freshly thawed and the re-cultured group. Both populations displayed a similar proliferation capacity which is indicated by a comparable population doubling time and colony-forming ability. Both freshly thawed and culture-rescued UC-MSCs expressed the characteristic immunophenotype and were capable of differentiating into osteocytes, chondrocytes, and adipocytes. On the other hand, culture-rescued cells appeared to be more potent in immunosuppression than freshly thawed cells. In conclusion, freshly thawed and culture-rescued cell products share comparable bioactivity in cell growth and proliferation, immunophenotype, and differentiation potential. However, the culture-rescued cells that were allowed to grow for an additional passage appear to display a more favorable immunomodulatory potential when compared to their freshly thawed parent cells.
