    Abstract: A feature of allergic airway disease is the observed increase of nitric oxide (NO) in exhaled breath. Gram-negative bacterial infections have also been linked with asthma exacerbations. However, the role of NO in asthma exacerbations with gram-negative bacterial infections is still unclear. In this study, we examined the role of NO in lipopolysaccharide (LPS)-induced inflammation in an ovalbumin (OVA)-challenged mouse asthma model. To determine whether NO affected the LPS-induced response, a NO donor ( S-nitroso- N-acetylpenicillamine, SNAP) or a selective inhibitor of NO synthase (1400W) was injected intraperitoneally into the mice before the LPS stimulation. Decreased levels of proinflammatory cytokines were demonstrated in the bronchoalveolar lavage fluid from mice treated with SNAP, whereas increased levels of cytokines were found in the 1400W-treated mice. To further explore the molecular mechanism of NO-mediated inhibition of proinflammatory responses in macrophages, RAW 264.7 cells were treated with 1400W or SNAP before LPS stimulation. LPS-induced inflammation in the cells was attenuated by the presence of NO. The LPS-induced IκB kinase (IKK) activation and the expression of IKK were reduced by NO through attenuation of the interaction between Hsp90 and IKK in the cells. The IKK decrease in the lung immunohistopathology was verified in SNAP-treated asthma mice, whereas IKK increased in the 1400W-treated group. We report for the first time that NO attenuates the interaction between Hsp90 and IKK, decreasing the stability of IKK and causing the down-regulation of the proinflammatory response. Furthermore, the results suggest that NO may repress LPS-stimulated innate immunity to promote pulmonary bacterial infection in asthma patients.

    Abstract: Extracranial metastasis from glioblastoma multiforme (GBM) is rare, especially multi-site metastases without intracranial recurrence. However, the metastatic mechanism of GBM remains unknown and there is currently no consensus regarding the best therapeutic regimen. We report the case of a 46-year-old man with primary GBM who developed scalp metastases and subsequent multiple pulmonary metastases. He was treated with the Stupp regimen after surgery for the intracranial tumor. However, a series of soft masses in the scalp were subsequently identified, and new nodules were found in his left eyebrow arch during chemoradiotherapy. Despite salvage chemotherapy and targeted therapy, the patient eventually died of respiratory failure with multiple pulmonary metastases. This case highlights the need for rigorous follow-up, including brain magnetic resonance imaging, in patients with GBM. The occurrence of extra-central nervous system symptoms indicates the possibility of metastasis, and the relevant examinations should be conducted promptly. Positive therapies may help to relieve symptoms and prolong survival in patients with metastatic GBM.

    Abstract: The development of diabetic macrovascular complications is a multifactorial process, and melatonin may possess cardiovascular protective properties. This study was designed to evaluate whether melatonin attenuates arteriosclerosis and endothelial permeability by suppressing the myosin light-chain kinase (MLCK)/myosin light-chain phosphorylation (p-MLC) system via the mitogen-activated protein kinase (MAPK) signaling pathway or by suppressing the myosin phosphatase-targeting subunit phosphorylation (p-MYPT)/p-MLC system in diabetes mellitus (DM). Rats were randomly divided into 4 groups, including control, high-fat diet, DM, and DM + melatonin groups. Melatonin was administered (10 mg/kg/d) by gavage for 12 weeks. The DM significantly increased the serum fasting blood glucose and lipid levels, as well as insulin resistance and endothelial dysfunction, which were attenuated by melatonin therapy to various extents. Importantly, the aortic endothelial permeability was significantly increased in DM rats but was dramatically reversed following treatment with melatonin. Our findings further indicated that hyperglycemia and hyperlipidemia enhanced the expressions of MLCK, p-MYPT, and p-MLC, which were partly associated with decreased membrane type 1 expression, increased extracellular signal-regulated kinase (ERK) phosphorylation, and increased p38 expression. However, these changes in protein expression were also significantly reversed by melatonin. Thus, our results are the first to demonstrate that the endothelial hyperpermeability induced by DM is associated with increased expressions of MLCK, p-MYPT, and p-MLC, which can be attenuated by melatonin at least partly through the ERK/p38 signaling pathway.

    Abstract: To evaluate the feasibility and accuracy of three-dimensional (3D)-printed individualised guiding templates in total hip arthroplasty (THA) for the treatment of developmental dysplasia of the hip (DDH). Methods: 12 hips in 12 patients with Crowe type IV DDH were treated with THA. A 3D digital model of the pelvis and lower limbs was reconstructed using the computed tomography data of the patients. Preoperative surgical simulations were performed to determine the most suitable surgical planning, including femoral osteotomy and prosthesis placement. Based on the ideal surgical planning, individualised guiding templates were designed by software, manufactured using a 3D printer, and used in acetabulum reconstruction and femoral osteotomy during surgery. Results: 12 patients were followed up for an average of 72.42 months (range 38–135 months). During surgery, the guiding template for each case was matched to the bony markers of the acetabulum and proximal femur. Preoperative and follow-up Harris Hip Scores were 34.2 ± 3.7 and 85.2 ± 4.2; leg-length discrepancy, 51.5 ± 6.5 mm and 10.2 ± 1.5 mm; and visual analogue scale scores, 6.2 ± 0.8 and 1.3 ± 0.3, respectively, with statistical difference. Shortened deformity and claudication of the affected limb were obviously improved after surgery. However, 1 patient had artificial hip dislocation 2 weeks after surgery, and another patient had sciatic nerve traction injury, both of whom recovered after physical treatment. Conclusions: Preoperative surgical simulation and 3D-printed individualised guiding templates can fulfil surgeon-specific requirements for the treatment of Crowe type IV DDH. Accurate THA can be achieved using 3D-printed individualised templates, which provide a new personalised surgical plan for the precise positioning and orientation of acetabular reconstruction and femoral osteotomy.

    Abstract: Leucine zipper tumor suppressor 2 (LZTS2) is implicated in several cancers; however, its biological mechanisms in non-small cell lung cancer (NSCLC) are not yet understood. We found that low levels of LZTS2 in NSCLC were correlated with tumor and nodal status. LZTS2 could inhibit cell proliferation and cell cycle transition at the G1/S phase and was implicated in the regulation of proteins associated with the canonical Wnt pathway, including GSK3β and β-catenin through inactivating the Akt pathway. These results provide novel mechanistic insight into the biological roles of LZTS2 in lung cancer cells.

    Abstract: The Zoige Basin on the eastern Tibetan Plateau has the largest area of highland peatlands in China. However, the development history of these peatlands is still poorly understood. Understanding how these carbon-rich ecosystems responded to change in the Asian summer monsoons during the Holocene will provide insight into the peatland carbon accumulation processes under different climate boundary conditions. Here, we document the timing of initiation and expansion histories of these peatlands using 59 new basal peat ages across the Zoige Basin, with 29 ages for initiation analysis and 30 additional ages for lateral expansion analysis. Also, we synthesized basal ages from 26 sites and carbon accumulation records at four sites from previous studies in this region. The results show that the peatland initiation is widespread at 11.5–10 and 7–6 kyr (1 kyr = 1000 cal. yr BP) and the minimum initiation periods occurred after 5 kyr. Our multiple basal ages along eight transects show that slopes are a dominant control on peatland lateral expansion rates, with very slow and less variable rates at slopes 〉 0.4°. Furthermore, we found a significant relationship between peatland basal ages and peat depths from 85 sites, suggesting relatively uniform peat properties. Carbon accumulation rates from detailed downcore analysis at four sites and on the basis of peat depth–basal age relationship show similar patterns with a peak carbon accumulation at 10–8 kyr. On the basis of estimated mean values of bulk density and carbon content from the region, the Holocene average C accumulation for the Zoige Basin is 31.1 g C/m 2 /yr. The widespread peatland initiation and rapid accumulation in the early Holocene were likely in response to higher temperature and stronger summer monsoon intensity, while the slowdown of peatland development during the late Holocene might have been caused by climate cooling and drying.

    Abstract: The coking coal resource in China is quite rich but is relative poor in the shallow coal beds. The total coals for coking are abundant, but the main coking coal resource is limted too. The coking coals distribute in 29 provinces (or autonomous regions), but they mainly concentrate only in a few provinces in which Shanxi province is the most important. Coking coal in China has rich in types, but among them the Gas coal and one-third of coking coal account for large proportion of proven resources reserve of coals for coking, meanwhile the coking coal and fat coal have low percentage, which are the main coal types in coking. In the current years the coking coal resource in shallow coal beds are becoming poor and poor because of the large production. Meanwhile, in the deep coal beds with more than 800 m contain lots of coking coals resources. According to the coal metamorphism theory the coal rank will increase gradually with the buried depth. When the mining depth is reach up to 1200 m in Northern China a large amount of fat coal, coking coal would be found and exploited in the coming years. This new prediction for coking coals in deep coal beds will resolve the problem of short supply of coking coal in China.

    Abstract: Aflatoxins are extremely harmful carcinogens to humans and animals. In recent years, attention has been directed toward the application of molecular imprinting polymers for the separation and detection of aflatoxin. In this review, polymerization methods for the preparation of molecular imprinting polymers for aflatoxin detection, such as lump-bulk polymerization, spherical molecular imprinting polymer synthesis, surface-imprinted polymerization, and electropolymerization, are described. The applications of molecular imprinting polymers in solid-phase extraction, biosensors, and the surface-enhanced Raman detection of aflatoxin are also reviewed in this paper.

    Abstract: The flammable and the mechanical effects of silica (SiO 2 ) on the halogen-free, flame-retarded ethylene–octene copolymer (POE)/polypropylene (PP) composites were investigated. Through the thermal stability analysis and the combustion test, it was found that the SiO 2 had a synergistic effect on the intumescent flame retardant (IFR), especially when the SiO 2 content was 3 wt%. According to the results of mechanical properties, the tensile strength of composites increased with the rising SiO 2 content and most of the impact strength is retained. The fracture morphology showed the state of SiO 2 /IFR particles in the POE/PP blends, which reflected that with the increasing SiO 2 content and the decreasing IFR content, the aggregates became less compact and the size of aggregates became smaller.

    Abstract: Although the clinical application of osimertinib, a third-generation epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI), has been a new step forward in the first-line treatment of non-small cell lung cancer (NSCLC), an increasing number of patients with progression on osimertinib represents a great challenge clinically. The patterns of resistance mechanisms and subsequent treatment strategies after first-line osimertinib resistance are not well established. Methods: Between January 1, 2016 and October 31, 2020, a consecutive of 56 EGFR-mutant lung cancer patients treated with osimertinib as first-line therapy at Daping Hospital (Chongqing, China) were retrospective screened. The samples of pre-osimertinib and osimertinib-resistance were all detected by next-generation sequencing (NGS) panels. Statistical analyses were carried out using SPSS 23.0 software. Survival analyses were performed using the Kaplan–Meier method and compared using a log-rank test between groups. Results: Among 47 patients with osimertinib effectiveness analysis, the median progression free survival (mPFS) was 15.4 months (95% confidence interval [CI]: 12.2-24.9 months), and median overall survival (mOS) was 35.5 months (95% CI: 23.9 months -NA). A total of 21 patients underwent repeated NGS tests upon osimertinib resistance. MET amplification was the most common resistance mechanism (6/21, 28.6%), followed by C797S mutation (5/21, 23.8%). A total of 15 patients received subsequent treatments, with mPFS of 7.3 months (95% CI 5.0 months -NA). Among them, 7 patients with EGFR C797 S or/and MET amplification received subsequent second-line targeted therapy, achieving mPFS of 7.3 months (95% CI 4.5 months -NA). Of note, 3 patients received immunotherapy as second- or third-line treatment after osimertinib resistance, achieving median clinical benefit of 37.3 months. Conclusions: MET amplification and C797S mutation are main resistance mechanisms, which could be targeted by crizotinib and gefitinib, respectively. More than 50% patients could receive subsequent anticancer targetable therapies after first-line osimertinib resistance. Immunotherapy may also be an acceptable choice after osimertinib resistance.
