    Abstract: Drug-associated cues can elicit relapse to drug seeking after abstinence. Studies with extinction–reinstatement models implicate dopamine (DA) in the nucleus accumbens shell (NAshell) and dorsolateral caudate-putamen (dlCPu) in cocaine seeking. However, less is known about their roles in cue-induced opiate seeking after prolonged abstinence. Using a morphine self-administration and abstinence–relapse model, we explored the roles of NAshell and dlCPu DA and the D1/D2-like receptor mechanisms underlying morphine rewarding and/or seeking. Acquisition of morphine self-administration was examined following 6-Hydroxydopamine hydrobromide (6-OHDA) lesions of the NAshell and dlCPu. For morphine seeking, rats underwent 3 weeks’ morphine self-administration followed by 3 weeks’ abstinence from morphine and the training environment. Prior to testing, 6-OHDA, D1 antagonist SCH23390, or D2 antagonist eticlopride was locally injected; then rats were exposed to morphine-associated contextual and discrete cues. Results show that acquisition of morphine self-administration was inhibited by NAshell (not dlCPu) lesions, while morphine seeking was attenuated by lesions of either region, by D1 (not D2) receptor blockade in NAshell, or by blockade of either D1 or D2 receptors in dlCPu. These data indicate a critical role of dopaminergic transmission in the NAshell (via D1-like receptors) and dlCPu (via D1- and D2-like receptors) in morphine seeking after prolonged abstinence.

    Abstract: Folate (FA) plays a key role in the biosynthesis of amino acids, purines, and pyrimidines in the human body, and intracellular folate metabolism has become an attractive target of tumor chemotherapy. In this work, an inclusion interaction was found between FA and cucurbit[7]uril (CB[7] ), and the formation of a CB[7]-FA 2:1 supramolecular inclusion complex was confirmed by fluorescence spectra, UV-Vis absorption spectroscopy, 1 H NMR, and molecular modeling calculations. In addition, FA is generally determined through the indirect fluorescent method because it shows weak fluorescence in aqueous solution. Therefore, a simple, direct fluorescence probe method for rapidly measuring FA was investigated, and the linear equation of FA was ΔF = 14.691C + 37.366 within the concentration ranges of 0.82 ~ 18.31 µg mL –1 . The proposed direct fluorescence method was applied to the determination of spiked plasma. We demonstrated that this method could provide an experimental basis for the targeted administration of the CB[7]-FA complex, and it could be extended as a promising fluorescence detection method for drugs in vivo.

    Abstract: Introduction Wear particles induced inflammatory osteolysis is the most important initiating factors in the mechanism of aseptic loosening. However, the molecular network changes in this process remain largely elusive. Methods Here, we performed whole transcriptome analysis using Ti particles induced RAW264.7 cell model to identify specific genes and pathways. Results Sequencing results totally identified 159 mRNAs, 96 lncRNAs, 31 circRNAs, and 12 miRNAs were significantly differently expressed. Of these, we selected two of each RNA for qRT-PCR validation and the results were highly consistent with the RNA-seq data. GSEA analysis shows that upregulated gene sets were related to the three classical inflammation pathway, cytokine–cytokine receptor interaction, TNF, and NF-kappa B signaling pathway. The enriched genes included not only IL-1β and TNF- α, which were independently verified before sequencing, but also other inflammatory osteolysis-related genes such as Mmp9, Fas, and Ccl2. Co-differentially expressed RNAs were employed to construct the ceRNA co-regulatory network. Conclusion: The results revealed that 4 lncRNAs and 2 circRNAs formed a regulatory network to simultaneously regulate miR-3065-3p targeting Myo18a. The present study helps to comprehensively understand the molecular mechanisms and regulatory interaction networks during early inflammatory response.

    Abstract: This study aims to investigate the therapeutic effect of vitamin D (VD) combined with interferon (IFN) type I (IFN-α) on mice with hepatitis B and to explore the possible mechanism. The mice were divided into control group, model group, IFN-α group, and IFN-α  +  VD group. After 4 weeks, the mice of four groups were weighed, and the thymus and spleen indexes were calculated. The activity of alanine transaminase (ALT) and aspartate transaminase (AST) in the serum was detected while pathological changes of the liver were observed. The levels of interferon-γ (IFN-γ), interleukin-2 (IL-2), and interleukin-4 (IL-4) in serum were measured by enzyme-linked immunosorbent assay (ELISA). The proliferation of spleen lymphocytes was detected by MTT assay. Flow cytometry was used to detect the level of CD4 + , CD8 + , and CD4 + /CD8 + in peripheral blood. The levels of ALT and AST in serum were significantly lower in the IFN-α  +  VD group than those in the IFN-α group, but the thymus and spleen indexes were significantly higher. Although both IFN-α and IFN-α  +  VD can improve the damaged structure of live tissue, IFN-α  +  VD achieved higher efficacy than IFN-α alone. The serum IFN-γ, TNF-α, and IL-2 levels were lower in the IFN-α  +  VD group compared with the IFN-α group, and no significant difference was found in IL-4. Compared with the IFN-α group, the percentage of CD4 + and the D4 + /CD8 + ratio were significantly increased, but the percentage of CD8 + was reduced. The proliferation rate of splenic lymphocytes was higher in the IFN-α  +  VD group compared with the IFN-α group. IFN-α  +  VD was found to achieve higher efficacy than IFN-α alone for the treatment of hepatitis B in mice, possibly through increasing the immune level of mice.

    Abstract: Animacy plays an essential role in survival and adaptive behaviour. Previous studies have found that dangerous or threatening animals can capture and hold attention. However, it is unclear whether and how neutral animate objects guide attentional allocation. It is also uncertain whether the modulation of animate objects on attentional allocation is based on the object itself (object-based attention) or its location (space-based attention). Therefore, the present study adopted the well-established two-rectangle paradigm and used animate and inanimate objects as stimuli to test the abovementioned problems. The results revealed that object-based effects were obtained for both animate and inanimate objects. However, the object-based effects were larger when the cue appeared on the animate objects than on the inanimate objects, due to faster response to invalid same-object trials and slower response to invalid different-object trials. Beyond that, we also further confirmed that animacy itself, not the low-level visual complexity, led to the differential object-based effects. These results suggest that neutral animals also mattered to our attentional allocation and animacy can modulate object-based attentional selection by capturing and holding visual attention on the animate objects. Ultimately, the present study not only enriches our understanding of how neutral animate objects guide attentional allocation and support the attentional prioritisation theory, but also further extends and amends the animate-monitoring hypothesis.
