    Abstract: It has been a significant and challenging issue to determine the elastic deformation of parallel manipulators for their precision analysis and control. A new method is proposed and studied for solving the elastic deformation of some parallel manipulators with linear active legs using computer-aided design variation geometry. First, an original simulation mechanism of a parallel manipulator is constructed; each of the vectors in the force transformation matrix of the parallel manipulators is constructed by this simulation mechanism. The active/constrained wrench and their pose are determined based on the Newton–Euler formulation. Second, the elastic deformed dimensions of the active legs are determined based on the elastic deformation equation and the active/constrained wrench. Third, a new simulation mechanism of this parallel manipulator is constructed by replacing the original dimensions of active legs with the deformed dimensions of active legs and the elastic deformations of parallel manipulators are solved using the pose difference between the original and new simulation mechanisms. Finally, two parallel manipulators are illustrated and their elastic deformations are solved and verified by both analytic approach and finite element method.

    Abstract: Highway traffic incidents induce a significant loss of life, economy, and productivity through injuries and fatalities, extended travel time and delay, and excessive energy consumption and air pollution. Traffic emergency management during incident conditions is the core element of active traffic management, and it is of practical significance to accurately understand the duration time distribution for typical traffic incident types and the factors that influence incident duration. This study proposes a dual-learning Bayesian network (BN) model to estimate traffic incident duration and to examine the influence of heterogeneous factors on the length of duration based on expert knowledge of traffic incident management and highway incident data collected in Zhejiang Province, China. Fifteen variables related to three aspects of traffic incidents, including incident information, incident consequences, and rescue resources, were included in the analysis. The trained BN model achieves favorable performance in several areas, including classification accuracy, the receiver operating characteristic (ROC) curve, and the area under curve (AUC) value. A classification matrix, and significant variables and their heterogeneous influences are identified accordingly. The research findings from this study provide beneficial reference to the understanding of decision-making in traffic incident response and process, active traffic incident management, and intelligent transportation systems.

    Abstract: Prostate cancer is a common malignant tumor with a high incidence. MicroRNAs (miRNAs) have been shown to be important post-transcriptional regulators during tumorigenesis. This study aimed to explore the effect of miR-144 on PCa proliferation and apoptosis. Material and Methods: The expression of miR-144 and EZH2 were examined in clinical PCa tissues. PCa cell line LNCAP and DU-145 was employed and transfected with miR-144 mimics or inhibitors. The correlation between miR-144 and EZH2 was verified by luciferase reporter assay. Cell viability, apoptosis and migratory capacity were detected by CCK-8, flow cytometry assay and wound healing assay. The protein level of EZH2, E-Cadherin, N-Cadherin and vimentin were analyzed by western blotting. Results: miR-144 was found to be negatively correlated to the expression of EZH2 in PCa tissues. Further studies identified EZH2 as a direct target of miR-144. Moreover, overexpression of miR-144 downregulated expression of EZH2, reduced cell viability and promoted cell apoptosis, while knockdown of miR-144 led to an inverse result. miR-144 also suppressed epithelial-mesenchymal transition level of PCa cells. Conclusion: Our study indicated that miR-144 negatively regulate the expression of EZH2 in clinical specimens and in vitro. miR-144 can inhibit cell proliferation and induce cell apoptosis in PCa cells. Therefore, miR-144 has the potential to be used as a biomarker for predicting the progression of PCa.

    Abstract: Adenomyosis is a uterine condition in which endometrial glands and stroma are commonly pathologically observed in the myometrium. In this study, we sought to determine the effect of resveratrol on the progression of adenomyosis. Adenomyosis was induced in mice given tamoxifen neonatally. All mice were subjected to body weight measurement and hotplate testing every four weeks beginning four weeks after birth. All mice with adenomyosis were randomly separated into 3 groups at 16 weeks: untreated, low-dose resveratrol (25 mg/kg), and high-dose resveratrol (50 mg/kg). After 3 weeks of treatment, final hotplate test and body weight measurement were performed, and the uterine horn blood samples were collected. Adenomyosis in mice caused body weight loss and uterine weight gain, reduced hotplate latency, and progression of endometrial fibrosis. The underlying biological process could be coupled with the overexpression of many cells’ proliferation and immune-regulation–related genes. Resveratrol treatment could slow the progression of adenomyosis by enhancing hotplate latency, lowering endometrial fibrosis, and restoring cell proliferation- and immune-regulation-associated gene expression levels in endometrium and plasma. However, resveratrol treatment also reduced the body weight and uterine weight. In conclusion, Resveratrol might be a potential compound for treating patients with adenomyosis.

    Abstract: Mastoscopic axillary lymph node dissection (MALND) is a currently used and safe surgical treatment option for breast cancer. However, the extensive application of MALND is still debatable because of the use of conventional axillary lymph node dissection (CALND). Therefore, in the current study, we aimed to compare the efficacy and safety of MALND and CALND for obtaining evidence-based conclusions about the short-term and long-term outcomes of MALND for patients with breast cancer. PubMed, Web of Science, Cochrane Library, and CNKI were comprehensively searched for articles published between January 1998 and January 2019. Then Newcastle-Ottawa scale was used for quality assessment. The Review Manager software version 5.0 was utilized for generating forest maps and funnel plots. Twelve studies including 2157 patients were selected for the meta-analysis. There were no significant differences in the number of lymph node dissections, tumor recurrence rate, axillary drainage, postoperative hospitalization time, and tumor size between the MALND and CALND groups ( P 〉 .05). In the MALND group, the surgery time was longer, while the incidence of intraoperative bleeding was lesser and the duration of drainage was shorter than those in the CALND group ( P 〈 .01). The complications in the MALND group were also fewer than those in the CALND group ( P 〈 .05). The results of the current study showed that MALND is reliable and feasible for breast cancer owing to the lesser incidence of intraoperative bleeding, shorter drainage duration, and lower incidence of complications compared to CALND.

    Abstract: This paper presents a numerical and experimental result of the end-wall vortex generator jets for controlling corner separation and enhancing the aerodynamic performance in a high subsonic (Ma = 0.7) compressor cascade. The experiments were carried out on a compressor cascade at design point ( i = 0°) and off-design points ( i = −2°, 2°, and 4°). At design point, the total pressure loss coefficient could be reduced up to 12.1%.With the increase in the incidence, the control effect is enhanced first and then reduced. The maximum total pressure loss reduction is up to 14.6% when the incidence is 2°. The numerical study is further conducted to analyze the flow pattern and the vortex structure. The jet vortex is formed downstream of the jet hole using the vortex generator jets, the cross flow on the end wall is also suppressed.

    Abstract: Vortex generator jets usually use a circular hole. The hole exit shape is also an important parameter, so the change of the shape will have a significant influence on the control effect of the compressor. In this article, the optimization design of the hole exit shape is studied in order to find the optimal geometry. The 2D hole exit shape is represented with a binary coded and initialized with the B-spline. The surrogate-based optimizer consists of a parametric design code, a computational fluid dynamics solver, a Kriging model, an infill sample criterion, and a genetic algorithm. First, the design of the symmetric hole is studied in a low ratio of the jet to inlet mass flow. The circumferential length of the designed hole is fixed. Second, both the symmetric hole and the asymmetric hole are studied. And the area of the hole exit shape is fixed. The results show that the optimal hole shape can enhance the strength of the jet vortex and reduce the total pressure loss of the compressor cascade. A better control effect can also be obtained in a high jet mass flow.

    Abstract: We assessed the surrogacy of objective response rate (ORR), disease control rate (DCR) and progression-free survival (PFS) for overall survival (OS) in anti-PD-1/PD-L1 trials of metastatic melanoma through a meta-analysis of randomized controlled trials (RCTs). Methods: PubMed and EMBASE were searched for phase II/III RCTs till June 2019 investigating anti-PD-1/PD-L1 agents. Treatment effect (hazard ratio or odds ratio) on potential surrogates (ORR/DCR/PFS) and OS were collected. At trial level, we assessed the correlation between treatment effect on potential surrogates and OS, weighted by sample size, fixed and random effect models, and calculated the surrogate threshold effect (STE). Sensitivity analyses and leave-one-out cross-validation approach were performed to evaluate the robustness of our findings. Results: We included 8 RCTs (4110 patients; 11 comparisons). We did not identify strong correlations between ORR [coefficient of determination ( R 2 ): 0.09–0.25], DCR (0.41–0.57) and OS. However, we noted a strong correlation between PFS and OS, with R 2 of 0.82 in sample size, 0.75 in fixed effect and 0.72 in random effect model weighting, the robustness of which was further verified by leave-one-out cross-validation approach. Sensitivity analyses with restriction to trials with less than 50% crossover, phase III trials, large trials and first-line trials strengthened the correlation (0.78–0.94). The STE for PFS was 0.78. Conclusions: PFS may be the appropriate surrogate for OS in anti-PD-1/PD-L1 trials of metastatic melanoma. A future anti-PD-1/PD-L1 trial would need less than 0.78 for PFS of the upper limit of confidence interval to predict an OS benefit.

    Abstract: Various prediction indices based on the single time point observation have been proposed in chronic obstructive pulmonary disease (COPD), but little was known about disease trajectory as a predictor of future exacerbations. Our study explored the association between disease trajectory and future exacerbations, and validated the predictive value of the modified and simplified short-term clinically important deterioration (CID). Methods: This study was a multicenter, prospective observational study. Patients with COPD were recruited into our study and followed up for 18 months. The modified CID (CID-C) was defined as a decrease of 100 mL in forced expiratory volume in 1 second (FEV1), or suffering exacerbations, or increase of 2 units in COPD Assessment Test (CAT) during the first 6 months follow-up. Simplified CID was defined when excluding CAT from the CID-C model. Results: A total of 127 patients were enrolled in our final analysis. Compared with patients without exacerbations during the period of the 6th to the 18th month, patients with exacerbations were more likely to have frequent short-term exacerbations in the first 6 months (2.14 versus 0.21, p  〈  0.001). The short-term exacerbations were the best predictor for future exacerbations [odds ratio (OR): 13.25; 95% confidence interval: 5.62–34.67; p  〈  0.001], followed by the history of exacerbation before study entry, short-term changes in FEV1 and CAT. CID-C and Simplified CID were both significantly associated with exacerbations (OR: 7.14 and 9.74, both p  〈  0.001). The receiver operating characteristic curves showed that the Simplified CID had slightly better predictive capacity for future exacerbation than CID-C (0.754 versus 0.695, p = 0.02). Conclusion: Disease trajectory, including both the CID-C and the Simplified CID had significant predictive value for future exacerbations.     Abstract: The gap leakage between the impeller ring leads to the change of the pump cavity flow characteristics, resulting in the uneven pressure distribution of shroud, which causes the axial force of the cavity to change. In this paper, the flow in the front and the rear cavity of the multistage centrifugal pump was taken as the research object. Through the numerical method, the radial flow velocity, the leakage flow size and its direction, the core zone rotation factor of the front and the rear cavity of the multistage pump impeller at all stages and the axial force of pump cavity were studied. The results show that the leakage in the front cavity of multistage pump impeller at all stages flows inward along radial direction (i.e. it flows from the inlet of pump cavity to the front ring clearance). The rotation factor in core zone is higher than 0.5, and with the increase of rotation factor, the axial force of the front pump cavity increased. The leakage in the rear pump cavity flows outward along radial direction (i.e. it flows from the rear ring to the inlet of pump cavity). The rotation factor in core zone is less than 0.5, and with the increase of rotation factor, the axial force decreased gradually. Besides, the radial velocity and rotation factor in the front and the rear multistage pump of impellers were obviously along the axial direction at three regions, the regions are pump case boundary layer, core zone and impeller boundary layer. The flow in the core zone is dominated by circumferential circular motion, and the radial velocity in the core zone is 0. It is shown that the direction of the leakage in the pump cavity and the rotation effect of the flow micelle in the mainstream core zone are the main factors affecting the axial force of the pump cavity, and the research results can provide theoretical guidance for the calculation and suppression of axial force of multistage pumps.
