    Abstract: The pressure fluctuations and flow pattern in a mixed-flow pump are numerically investigated under design and off-design conditions. The accuracy and reliability are validated by good agreement of numerical results and experimental data of pump energy performance. At design flow rate, the dominant frequencies of pressure fluctuations are impeller rotation frequency or integer multiples of it, which are mainly induced by the rotor–stator interaction. At overload condition, the dominant frequencies of pressure fluctuations are similar as those of design flow rate, but the maximum amplitudes increase to about 2.5 times of those of design flow rate. At partial load condition, the pressure fluctuations and flow pattern are complicated. The low dominant frequencies below the impeller rotation frequency are related to the process of vortex emergency, movement and disappearance.

    Abstract: To explore the effect of virtual mass force, the unsteady two-phase flow in a multiphase rotodynamic pump impeller was numerically simulated, where the inlet gas void fraction was 4.9%, 14.9%, and 25.2%, respectively. The drag force and the virtual mass force were accounted for and the cases with and without the latter one were both analyzed for comparison. The results show that the trajectories of the gas bubbles are influenced by the virtual mass force evidently in the inlet extended region. Due to the effect of virtual mass force, some gas will firstly move to the shroud before accumulating in the hub region of the impeller. The characteristic of the pump head was discussed and the results demonstrate that the virtual mass force can decrease the pump head and lead to its fluctuation. In addition, the comparison between the steady and unsteady simulation shows that the virtual mass effect can be found only by unsteady simulation.

    Abstract: The influence of inlet guide vanes on unsteady flow in a centrifugal pump is numerically investigated. The independences of mesh elements, time steps and turbulence models are studied, and the satisfactory agreement between experimental and numerical results of the centrifugal pump performance validates the reliability and accuracy of the numerical model. The frequency characteristics of pressure fluctuations in impeller and volute are nearly the same for the pump without and with inlet guide vanes in the angle range from −36° to +36°. In the pump impeller, the dominant frequencies are mainly the rotational frequency f i (24.17 Hz) or 2 f i , and in volute they are the blade passing frequency f BPF (145 Hz). For the large inlet guide vanes angles of −60°and +60°, the maximum amplitudes of pressure fluctuations in pump impeller and volute are stronger than that in pump without inlet guide vanes. Therefore, the influence of inlet guide vanes on unsteady flow in the centrifugal pump is slight when the inlet guide vanes angles are regulated in a suitable region.

    Abstract: A combined direct and inverse iterative design method was developed for the hydraulic design of centrifugal pump impellers. This method is based on the fluid continuity and motion equations and solves for the meridional velocity taking into account the effects of the blade shape on the flow. The blade shape is drawn by point-by-point integration with blade thickening and smoothing using conformal mapping. Two examples designed using the direct and inverse iterative design method are compared to results using the traditional design method with significantly different meridional velocity distributions and three-dimensional blade shapes. Numerical simulations and tests show that the highest pump efficiency is 2.2% higher with this design method than with the traditional design method. The numerical results agree well with the experiments with a smoother flow pattern than with the traditional design, especially in the volute.

    Abstract: Long noncoding RNAs (lncRNAs) have an effect on the occurrence and progression of a considerable number of diseases, especially cancer. Existing research has suggested that MAGI2 antisense RNA 3 (MAGI2-AS3) takes on a critical significance in the development of hepatocellular carcinoma and lung cancer. However, the functions of MAGI2-AS3 in laryngeal squamous cell carcinoma (LSCC) remain unclear. In this study, MAGI2-AS3 expression level in LSCC tissue and cell lines was detected, and the effect of MAGI2-AS3 overexpressed on LSCC phenotypes and the possible influence mechanisms were examined. MAGI2-AS3 was downregulated in the tissues of LSCC patients versus non-tumor tissues, and it was correlated with advanced TNM (tumor, node, metastasis) stage and lymph node metastases, as indicated by the results of this study. MAGI2-AS3 inhibited the proliferation, migration, and invasion of LSCC cells in vitro and in vivo. Furthermore, the hypermethylation level of the MAGI2-AS3 promoter region was indicated by bisulfite genomic sequencing and methylation-specific polymerase chain reaction, such that MAGI2-AS3 expression was downregulated. Besides, MAGI2-AS3 promoter hypermethylation was regulated by DNA methyltransferase 1 (DNMT1), and MAGI2-AS3 expression was reversed by 5-Aza-2′-deoxycytidine (5-Aza). Moreover, the result of the RNA pull-down experiment suggested that 38 proteins were enriched in the MAGI2-AS3 group versus the control group in TU177 cells. To be specific, SPT6 (ie, a conserved protein) was enriched by fold change 〉 10. SPT6 knockdown reduced the antitumor effect of MAGI2-AS3 in TU177 and AMC-HN-8 cells. Meanwhile, SPT6 overexpression inhibited the proliferation, metastasis, and invasion of TU177 and AMC-HN-8 cells. As revealed by the above findings, DNMT1-regulated MAGI2-AS3 promoter hypermethylation led to downregulated MAGI2-AS3 expression, such that the presence and progression of LSCC were inhibited in an SPT6 binding-dependent manner.
